The floor geometry has risers/sprus that don't seem covered in the spec. What are these for? Would they not translate to air gaps in the final part?

I am excited (and optimistic!) about your efforts and contributions in this space.

Being asked to read LLM outputs, particularly undisclosed ones, feels bad.

Given your background, I presume you're pretty exposed to the discourse on this topic, but I'm happy to expand on this if it's helpful.

Oh yea, the computers are surprisingly bad with versioning, I'll go take a look at that pipeline and see what they are supposed to be if you're still interested.

This one fell by the way side as I have been using the serial dilution method to isolate. It is much faster and more effective than what I could do with these. I would highly recommend giving it a shot!

Wait what were these slides gonna do that competes with serial dilution?

Dilution is my preferred method since dedikaryotizing solutions remain beyond me for now, but the resulting cultures still need to be inspected for clamps.

I'm using these (experimentally and as a fallback/curiosity) to inspect the possible monokaryons obtained via serial dilution without the physical disturbance associated with other methods, since SD alone doesn't guarantee isolation and there still needs to be clamp connection inspection at some point.

Oh geez, well this is going to show how big of a noob I am.

I assumed that this was an alternative path to isolation, but I think that I understand now you are using the slides only for observation, possible mating. Is that correct?

And now I am realizing I made a much larger logical leap in my isolation methodology.

Back to the HHG for me!

Background: when a spore germinates, it creates monokaryotic (single nucleus) mycelium. This is an interim state that can't fruit on its own and only exists to find a mate. Two of these intersect and their genetics combine to create a dikaryotic (two nuclei) organism that's capable of producing fruit, whose spores will then encode some combination of the two parents' genetics. These nuclei are shared between cells via clamp connections.

(a single monokaryon can also borrow from an already mated pair, this is useful info for breeding because you only need a single monokaryon to start creating new things, and it allows you to use genetics from things you couldn't otherwise get an isolated spore from, like enigma).

In a multispore grow, huge numbers of spores germinate, meet, and form many different dikaryons that then all battle it out and occupy various parts of the substrate and fruit with various combinations of genetics.

--

"isolation" can mean a lot of things in cult.

Most people use it as in "not multispore", a ~single culture, usually a clone of some fruit etc, that behaves evenly and reliably.

But often these are only approximately isolated cultures. In a multispore grow it is possible to have multiple dikaryons coexisting in a space, and even in a single fruit. I don't really understand the details of how this works or is possible, just that it apparently is. So you can take a tissue clone from the interior of a single fruit and have something that is an isolate in the sense of being stable and single-physical-origin, but is not isolated in the sense of being one isolated dikaryotic organism.

A true "isolate" in the way it's usually used refers to a single dikaryotic culture.

Then there's the act of isolating monokaryons, which involves catching this unmated mycelium early in its life (or inducing separation later, whether chemically or by beating the shit out of it).

Because spores can be so tightly clumped together, even on a well separated streak plate, it can be difficult to identify whether a single visible germination point represents an actual single/unmated monokaryon. The confirmation process for this includes looking at it in a microscope and trying to find clamps as hard as you can, and presuming success if you cant.

I've been using this for clamp hunting, to help determine whether the germination points I've individually sampled from a streak plate are actually monokaryons or not.

So then what distunguishes a single dikaryotic culture from another single dikaryotic culture? Is it its genome?

From a genetic perspective, yes, similar to humans in that way.

From a cult performance perspective there's all the environmental stuff on top of that, it could have insane genetics but if it grew up ice cold with no food it might still be a lil stunted, compared to something average that was well taken care of, etc.

So if Im understanding this correctly, its like the analogy of perfect twins being seperated at birth and then growing up in vastly seperate environments.

They have the same genome at birth, but through their environment and diet, their epigentics change their pheno expression based on their lifestyle (and their genome would possibly mutate).

This would make them distinct (dikaryotic isolate cultures).

Am I getting this right?

That's a good stress test to expose the difference between humans / where the metaphor breaks down. The identical twins would just be one fused thing and, if separated then reintroduced, merge back into one, despite their differences.

this post is inspired. Besides the initial effort to make the vessels themselves, which is the main hurdle to streamline, it seems easy enough. It fulfills three requisites of being great imo, 1 it removes the need to make a slide because it already is one, 2 allows for narrower plane of viewing and reduces storage, 3 it can be put to both pour and no-pour applications and as a bonus it consumes already existing materials and preserves agar.
In its simplicity and reductiveness it is just brilliant. it may be some added effort, but is oozing with utility and crtitical thinking. As someone who prides themselves on improvising and being crafty, I must say it's efficient and very easily manufacturable. It functions on multiple levels, and kill 3 birds simultaenously. It seems straightforward enough to automate and is easily patentable with the right design and criteria. It is very clever.
maybe it already existed but I consider your application original; there are few teks that don't already inherit some existing knowledge and you clearly demonstrate here that you are A more than a hobbyist B sticking it out for the long haul. Nobody puts this much love, effort and attention into a passing leisure or something they are going to throw away.
Welcome to the community Ras, I can't wait to see how you develop this idea further, I'm certain it won't be your last one.:pimp3::hatsoff:
