What to do with a bunch of half colonized agar plates?
    #2925597 - 07/25/04 11:35 AM (16 years, 3 months ago)

They seem to have stalled at half colonization, is this normal?

I dont know why I made them but I did and I want to use them for future grain innocs. Should I wait until they are done colonizing? Then what? I got 10 plates of Z+ going..

I've heard of sucking up some water (what kind? just tap?) into some syringes sticking them in a jar and PCing them (where can I get syringes that will stand up to PC and how long do I PC?). Then while using the needle to scrape at the mycelium squirt the PCed water into the dish and resuck the water. I could make tons of mycelium syringes this way. Would this be what you guys would do? And would I need full colonization for this? All the mycelium looks ropey and beautiful..

IMO you should harvest some of the cleanest fastest growing sectors and inoc some fresh plates of agar , then once you get a sweet strain a goin either inoc some grains or do the old syringe tek . What type of agar are you using? it is quite possible that the ph may be a little off for the particular species you are trying to germ , thus the apperance of stalling . The myc you have so far will colonize the next batch of agar with no apperant stalling if my theory is correct , due to fact it is happy with food source .
Good Luck

They have been in constant 83-85* temps.

I dont want to go to another plate tripndicular, I'm just looking for the best way to store it until I'm ready for it. I figure myc syringes will be best. I'm not sure how cautious I need to be of the uncolonized agar.

Wow, that sounds pretty warm. I've read temps are best between 78-80, but it will depend on your variety. This range has worked well for me so far.

Check the thread dates, most of the suggested threads are old.
