Damn that’s more expensive than I thought it would be, what did you use to connect the plexiglass together?

Siliconed in place and to flow hood. 2 plexiglass gussets with 45/90/45 degree pieces to kinda keep it square.

It came later as I had to save for it. Real expensive because you buy the whole piece like 48" x 60"and then nothing is simple plexiglass. They make them clear but when viewed from the edge it looks blue like real glass. And added efficiency.. blah, blah. I wasn't able to source it cheaper really. Make them cut it. Thicker pieces have to scribed deep.

i liked that now you explained your point better for understanding, but i am still not convinced.

i will answer it the most respectful way and simpler way possible, trying to explain my point:

Quote:
The tip of the scalpel absolutely can be a contamination vector. The relevant issue isn't that the metal was sterile when I flamed it. The issue is what happens after that point. Once the sterile instrument and exposed material are operating inside the SAB, you're dealing with airborne particulate contamination: dust, skin particles, spores, droplets, and everything else that can settle onto an exposed surface. That's precisely why minimizing exposure and unnecessary manipulation matters. The same principle applies to your syringe.

not exactly. like i said, red hot sterilization, means the metal is at 600C. when you wait some seconds for it to cool down, doesn't mean the metal is at room temperature. the metal is still at 200C+. this means it is still "sterile", because *most* particles, droplet, or spores, will be deactivated very quickly in touch with 200C temperature of the metal. so even if any particle touch it, it will be dead in some seconds. maybe a endospore could survive, but a chance of having a endospore getting in touch with a needle of size of a couple of millimeters, inside of a SAB seems very unlikely to me.

Quote:
You're treating the LC as though it exists in a binary state of "sterile" versus "contaminated." Real aseptic work isn't that black and white

not exactly. i'm talking about more reliable methods and reducing contamination vector and bacterial load inside the LC, not about a binary "sterile/not sterile" thing.

Quote:
Your LC vessel has already been opened and exposed to the working environment during inoculation

not if i had used a SHIP. yes, it can still theoretically have some air passing through the SHIP, or the needle not be 100% sterile, or micropore tape not blocking 100% of contams, but is of course more reliable than any other method. you are comparing some millimetric hole done with the needle with opening an entire jar. the exposing area are like 100x bigger.

Quote:
If I can transfer material directly from the donor to the intended recipient with a sterile instrument, or no instrument at all, (gravity) then adding another vessel, a syringe, another needle, another penetration point, another handling operation, and another interface doesn't automatically make the procedure safer merely because the needle can be flame sterilized. A sterile needle is still another thing that has to be introduced, manipulated, cooled, positioned, and passed through another interface correctly. That's the distinction I posit you are missing: that the sterility of an individual component does not equal sterility of the entire process

this contradicts the idea of using agar to grain like you suggested. if i want to use agar, i need some kind of knife to cut the agar and put in the grain jar. what goes against your logic of "reducing steps". you are also simply ignoring the steps about the pouring method, opening jars to the air inside the SAB is also a step that can introduce contamination.

reducing steps don't automatically mean being safer.

@chiron thank you for you point. I wont take it into consideration because you're incorrect, but as far as I am concerned at this point our sxchange is null, and I wont respond to you further, respectfully. Please continue to engage on the boards; perhaps someone will reach you better than I did.

Bout to throw some jars in the IP and was wondering do I always have to put lids on the bottom so the jars don’t touch the metal of the pot on the bottom or can I set the jars on the base and be fine? It’s just that putting the lids in there when I’m sterilizing they get rusty really fast and it’s getting pretty annoying.
Also I’ve been slacking on transferring my agar plates. I have some plins and also just very colonized plates. Should I transfer before putting them to grain or are they fine as long as they’re still clean?

Because you are handling it more. Most particles settle to the bottom of the sab which is the point. You are trying to disturb them as little as possible. There are more surface you come in contact with. There are more moving parts for things to go wrong. My way is the accepted reliable way because it is less complicated and commonly received as accepted in theory and practice. The argument is null because it can't be proved objectively, only inferred through personal experience.

I am giving you the best advice I can, but there is no point trying to explain further because you did not come here to ask a question; you came searching for validity for an unsubstantiated hunch.

I recommend not letting the jars touch the base of the PC they might crack, my lids get rusty too, you can continue to use them just make sure to take them out as soon as possible and rinse and dry them to prevent rusting but it will happen. You could also get a raised trivet to put in and do away with the lids which is probably your best option. And as long as the plates are clean you should be good to go, Also if the pins are tiny if you wanted you could transfer them to another plate.

Thanks for the quick answer! Also I just literally grab the plin and chuck it into another plate right? I mean since they’re so small I can’t see grabbing a small piece of stipe easily

We can explain it for you, Chiron, but we can’t understand it for you.

Why is it every topic? Are you here to learn?

Like I said, it is hard for inexperienced people to understand without a foundational knowledge in their head, and they want to be argumentative for its own sake

not every topic. kloakk had teach me about not using gas exchange for LC and i agreed. some days ago fiddle_head had teach me about PC psi and time of sterilization and i agreed, and this happened other times too, just go on my profile and see it.

i'm a very simple person, if it makes sense i agree, if it doesn't i don't agree. is just that simple.

i'm here to learn, and if TC's are here to teach(what i think is what most TC want), then is a very simple matter, just explain it better and i will learn when it makes enough sense. i just don't agree with everything blindly, that's it.

the whole point is that for some reason it starts to be an ego debate for most of the discussions i had participated here, like what fiddle_head did some comments ago:

Quote:
i answered your question. by all means use a syringe and a ship. but dont pretend like you are capable of giving me advice or knowing more than me on something you've never actually carried out.

this is no "explanation" of anything, this is a "i'm smart, you dumb" argument focused not on arguments but on ego. so how can i understand something if there isn't enough, satisfactory explanation?

I’m sorry you see it that way. That’s not what he’s saying. He’s saying he knows more about a subject you have openly stated you’ve never practiced, so if you want advice he has it, but if you don’t want advice there’s no point in trying to tell him anything. That’s not ego, that’s truth that I think you find uncomfortable.

Idk man. I’ve taught dozens if not 100+ people all manner of thing on the forum over the handful of years I’ve been here. No one has had as much trouble with foundational concepts as you aside from people who legitimately had brain damage and learning disabilities. I honestly don’t mean that as an insult, but fuck dude.

If you’re not here to argue for no reason, I can’t figure out what you’re here for. The point of a system is what it does, right?

i want new insights from experienced people, what doesn't mean i will blindly accept them. is that simple.

experience are not omniscience. i will make a simpler example:

i had seen mushboy guide to liquid culture, mushboy is a TC. in his guide, he clearly stated that LC don't need gas exchange. then i posted it here to have a confirmation, and most people asked why would i want to do that, even if a TC recommends it.

if you go to bodhisatta oat prep guide(another TC), you will see him teaching prepping oats.

then you go to YoshiTrainer(another TC) guide to no prep grains, where he clearly states that he finds grain prep a waste of time.

do you understand what i'm talking about? there's many ways to reach the finish line, and many experienced cultivators had done it in different ways. then for something to make sense here, you have to state why your method is better than other methods, using arguments, not experience. many experienced and trusted cultivators had grown mushrooms reliably using different methods.

I find the idea that someone HAS to prove their option is superior for you to be erroneous. Why would we need to do that? We're all going to do what we want with the information here anyway.

I have done lots of prepped grains and no prepped grains. The conclusion I came to is that I get a more desirable end result with prepped grains. I'm never going to say Yoshi's way is inferior because there are use cases for his methods and grain choices that favor it. Lots of things here aren't black and white, they're options.

But when it comes to FOUNDATIONAL concepts like sterility, it's not really options.

Besides that, why do you think you're so special you're above "blindly accepting" advice when you don't know shit from fuck, dude? When you have a handful of people who know more about a subject than you all saying the same thing, maybe just listen? You've got a fat ego on you, and I find it a little silly that you're accusing fiddle of having one when you're flaunting your own.

Eat some humble pie big dawg. You stink.

nobody has to prove anything to anyone pal, and nobody needs to teach anything here either. people do that with their own volition, that's not the point.

the point is, if you are proposing to teach something, don't be mad if someone is willing to ask and defy what you are teaching.

there's several options using sterile techniques what the hell are you talking about. there's several guys who used syringes, bistoury, or nothing at all and got good results. there's people who uses 70% alcohol in the SAB and other people think this is useless. there's guys who spray soapy water inside of the SAB to decrease chance of contamination. there's others who uses nothing at all.

there ARE options when it comes to sterile techniques.

having a "big fat ego" means turning an argument that ain't personal in a personal attack. i never attacked the guy, neither you, just the argument. meanwhile, you called me a retard several times, and he said i'm inexperienced, so i just need to listen and not ask anything.
