After all the time I've spent in the hobby I still have trouble with my germ plates and figuring out what to transfer from. I made a bunch of new plates tonight pretty sure these were no good but transfered the spots you mentioned.

That’s why I grab three spots and put those three on one plate

My T1 plates always have 3 pieces. I just know I’m not gonna get clean transfer obviously from a T1 and I can then observe them and pick one to carry forward.

I just try to find fuzzy white threads and go from there.

The bigger growth one I would have grabbed two cause it’s the one I have the most confidence you can work out contam. Looks like is see a grow ring.

The upper left on plate one Icant decide what it is cause of how thick it is, but if your transfer tech is good, you’ll identify it fast.

The uniform growth on the second pic is what caught my eye. Something is riding along but again, good transfer tech and you should be able to separate it once you see it grow out.

Just kinda how I go about it and what I look for.

Hey shroomers,

I have 5 jars of different cube strain in my fridge for 6 month and tested them on agar. here is the result:







I think they all contaminated, or is this looking normal to you? The smell is actually also not really nice.

Hard to tell from the pics but if not nice white myc then you're probably right,  and will need to clean up with subsequent transfers

Is.that a film on the surface or reflection

From what im seeing (on.phone screen)
Looks like there's at least some healthy myc in there that could easily be transferred to a cabin. Or hell I'd try a hot pour over them (2 layers if adequate space)

Some cyan peace river porn.  I put four wedges from a grab and drag plate in a square hole together.  So that I could have a better chance of a fruiting strain, I can observe how they behave together, and hopefully catch any bacteria in the hole.

I like the results so much I'll likely keep doing my t1s like this.  And maybe add some trenches next time for fun :smile:

Im curious about the mechanisms behind this. I can see how doing this can select for more robust and vigorous mycelium since it has to span a gap, but how does this translate to a good fruiting strain? But in any case, a neat technique, I will have to try it out :lol:

Grab and drags spread the spores out, making the chances of transferring a mono higher.

Additionally not all mating types are well suited for fruiting.

Essentially, I am giving them more opportunity to find mating partners well suited to fruiting.

It doesn't guarantee that it will be a good fruiter.  But it increases the odds.

The top culture, was likely two dikaryons or a mono and a dikaryon, that mated and made the top strain.

Edit:  And the bottom strain didn't want to mate with either of its neighbors. So I will likely run that one too.

Thats really interesting, I guess I will have to read up on the details of mycelium reproduction in the near future then :lol: thanks for clearing it up.

So if im getting this right, grabs and drag have a higher chance to produce monos and then by placing the potential monos next to eachother in the hole the ones with good compatability will mate together and have a higher chance of fruiting?

Clear as mud and I've been reading that book off and on for a year :lol:

Honestly a lot of it for me is muscle memory (or eye memory more accurately).

Mycelium behaves very differently when there is competition on the plate. And giving them competition from the same print lets me observe if they're still looking for suitable mates and are vigorous compared to other strains from the germ plate.

It's also a way to introduce competition without contamination.

The square just traps bacteria.  And is something new I'm playing with.

But multiple cultures on one plate is something I picked up from Julian and have been doing awhile.



Edit:  It also cuts down on dishes.  I don't like washing glass plates.

Excuse the very open and simple question, but when having multiple cultures on one plate, what are you keeping an eye on?

Genetic compatibility and growth behavior/rate.

This is two strains that crossed late.





And this plate has one promising looking culture on it and two the don't look great.  Because the cultures are right next to each other, it makes observing bacteria and other sus growth easier.



This one knotted and canopied.



And this is the same plate under black light, this lighting bolt structure is a cord.  Which is a type of non-vegetative growth.  Since there is competition and they run out of room faster, non-vegetative growth shows up earlier and more consistently.



All of these are Copelandia by the way.  Since cubes make rhizo growth, I'm sure the technique would work out differently.  (Hence eye memory).

But I do imagine it would encourage plins.

I would love to see more folks playing with it.  I have learned a lot observing them with neighbors that I don't believe I would have learned keeping them separate.
