I have 66qt tub with two holes cut in it. I spray really good on the inside with 90% ISO and let it sit for a few minutes before I do the Agar.

There are some good threads around here with pointers to help prevent contam. I'll see if I can find them and post the links.

^ That'll do.

70% alcohol is actually a better sanitizer than 90% but no worries, that's just splitting hairs.

No need to wait btw, with the walls wet and static from the tote, any contaminants are stuck to the sides and the air is still, almost instantly. -Ever been drenched in sweat 🥵 and close the window? You can feel how quick it becomes dead air - 🤣🤣🤣

As for isolating away from contaminants or cleaning up cultures... It's good practice to make transfers as soon as you see mycelium expanding & not wait for days. Takes more plates but easier to fix; at least until you get comfortable with your sterile procedure & get an eye for different baddies & proper growth. 👍🏻🫶🏻

Thanks, good info. I am still learning. I started off really good and had some really good tubs. I have a few going now with some pretty good shrooms. I have had some Trenching tubs going on lately. I am trying to figure out why. They start off good and then start Trenching. I moved the new tubs to a different room to try to insolate from the Trenching tubs. I am guess that the baterial is Trenching the new tubs.

This plate was taken from 2 sections of g2a where I could tell that the growth came from each grain.
I like that you can see a border, but there is a clean line where you see where they're getting along.

Life situation made me stop abruptly quite awhile ago.  I had a great ghost isolated that reproduced fantastic shrooms - alas, that is gone. :frown:.

I had 1 syringe stored in a fridge and hoped beyond hope it would be viable and I could start again. How long is normal to see some activity on an LME agar plate??

I am really worried I have nothing to actually work with.

Welcome back!

I have heard of spore syringes remaining viable for years under refrigeration.  If you don’t have luck though, slide on over to the Marketplace and get a fresh spore print from one of the giveaways. :mushroom2:

Id definitely give it the ol college try at least! I can't speak to syringe shelf life, but I've seen mention of people reviving some pretty old spore prints anyway. I hope it works out for ya though!

These are my first plates. I used LC to inoculate. I could use some advice as to where I should start trying to isolate and why. There is only one super obvious contamination in one of the plates. I appreciate any insight on my journey.

These spots all look pretty good to take a transfer.

If you see similar looking spots on the plates in person, and the pics didn't do it justice, then I would take some additional transfers from those spots as well.

No rush. 😀

I only have 3 fresh plates for the 3 best spots. Making more plates up today/tomorrow as I just got my order today.

I assume there are at least some promising transfers.

Thank you!

I was eyeing a couple of those areas as well. Certainly not all. Heading to farmers marker and getting something there that I'm going to clone as well. WTH
