I envision a scenario where many prints of the same species could be added to a jar or media bottle filled with distilled water, with added surfactant like tween-20 to prevent clumping, and a stir bar. Then pasteurizing that jar to produce reliably clean, commercial quantities of spore syringes.

The jar and surfactant might have to be sterilized first, but may be worth the effort.

Syringes aren't technically necessary for  home cultivation, especially if you're streaking/spreading the spores for future culture work. A small jar could be used to dip into with an inoculation loop whenever  necessary. The stir bar would be helpful when either filling syringes or for re-suspension of the spores prior to use.
Much like I do for spore rehydration, but with the pasteurizing twist.

Damnit Stipe this is absolutely 🔥! I'm so game! I have a brand new ultrasonic bath that has the capability of using temperature and/or ultrasonic. I have a few experiments on going with the thing and I'm now adding wild spores to the list! Let's see what happens!

Easy enough to preload the jars with distilled and sterilize before adding spores or prints. Use Jet dry rather than tween as a surfactant to keep it to what I can find under the sink? See how simple convection works rather than use a stir plate?
I’ll start this dumbed down basic and see what happens.

Jet dry definitely works, so does manual agitation. We need to confirm this works well enough first before we get there . :cheers:

I just remembered that I inoculated a plate with those old syringes and put it in my discard pile. It grew grey mold with white interspersed with it. I haven't bothered to confirm that the white was mushroom mycelium because the two were intertwined so much that it would've been a waste of time. I still have the plate.

This means I already have a 'before' plate.

I'll use my ninja 'keep warm' function and put the syringe in water for about 6 hours. Hopefully that's enough time to bring the liquid inside up to proper temperatures.

I'll do this the next time I'm home from work. I'll post pictures of the before plate, water temperature, and then inoculate a fresh plate this weekend sometime.

If it works for a jacked up 14 year old syringe, then it should work with any syringe.

Even getting the test plate to grow nothing would be a good point of data

Great thread! GreeneMyco linked it in my wild cubensis thread!

This could be a way to get crosses as well fairly easily for PF Tek, even with wild prints. Most importantly, it removes the need to go to agar, which is immediately creating genetic bottlenecks.

I think I'm going to have to scrape a few prints together into a mason jar of water, use a stirrer to get them really mixed up, then pasteurize it, then fill the syringes with it.

Stipes have you tried this already?

I mean, 6 hours at a very low temperature isn't synonymous with pasteurization but, give it a go and see what happens. I wouldn't hold out much hope for anything significant to happen, it may even make things worse by incubating microbes.

Would/ do you think the benzoyl alcohol in bacteriostatic water would kill spores?

I'm not sure what the sporicidal properties of that are nor aware of the hardiness of spores we use for cultivation. Should be easy enough to test though.

In the name of snience I will find out if they survive 45 mins in the instant pot while I make agar and if that is enough. Wish me luck.

I will mix Yeti with Husky, as it should be pretty obvious if one or the other dominates, or if they blend.

I do believe my keep warm heats it to somewhere between 140-160f. It's been a while since I've checked.

if I ever decide to grow some mushrooms again I'll be doing this... right now I don't see that happening anytime soon though, hell I haven't grown a single mushroom in almost a year... my movers fell off the face of the earth🤣

I have no experience with spores, but cubensis mycelium survives freezing for extended periods. I’ve revived cubensis mycelium that was in the freezer for a year. I would think spores would be even more cold hardy. I guess my question is what do you gain by freezing spores? I understand the utility of pasteurized spores when trying to save a contaminated culture, but freezing?




dont know, dont care!! just popped into my head, i told stipe i wasnt trying to derail the thread.

there are some papers about freezing cultures and or spores to deal with mites but i think heat might prove more beneficial for that as well
