Lmaooo I'm jc bc I had test plates that I'm 90% were good and then most everything shit the bed anyway. Im jw if bad LC will look obviously messed up like yours when testing on agar. I feel like Lew told me that it's not a guarantee, but my memory is dog shit.



I wrote this a while back, and I post here to anyone wondering about LC not just you:

Liquid Culture. Don't do it until you master clean agar plates.

LC can be a great way to grow a lot of shrooms fast, but it's also a great way to grow a lot of mold and bacteria fast.

I don't see the point of doing LC early on, or when still having any contam issues.

LC is unneeded unless you're running a commercial grow and need to grow kilos. If not, why not just toss agar slices into grain jars, and shake at 30 percent? You're much more likely early on to get clean grows.

It's true that you can't always tell if it's clean in the liquid, but if someone must do LC, I'd recommend start using nothing but 4 grams dextrose in 100 ml distilled water, PCed for 25 min, cool to room temp before nocing.

It's as clear as water, which gives a better chance of seeing contam.

Good LC basically looks like sperm in water. (still should be tested on agar)



Oh I knew I was gambling. I just did it anyway bc I like ice cream and have an IQ of 48.

I went from LC to agar for the sole fact that i'm trying to keep that strain going for a long time.(and make spore prints when i have a good grow, But i knew from the start that the LC looked funky with white growth surrounding a large spore clump.

It would appear that the cabin didn’t do anything for the scalloped culture.  So… could it be that if it is bacteria, is the bacteria being pushed through with the myc?  Does that happen?  That’s a real ? for anyone that might know.  If it does happen, could a stiffer agar mix help?  I used 1.8% on this plate. 

If it isn’t bacteria, could it be viral?  I suppose there’s really no way to find that out visually unless you have an electron scope readily available lol.  Maybe we could test the presence of the virus through a contact test with a healthy culture.  Just spitballing but possibly have a transfer of a healthy culture grown out like a couple inches or less.  Then take a transfer of the scalloped culture and flip it so that the myc is facing downward and touch it to the healthy culture a little bit in from the leading edge; being careful to only let the mycelium of the two touch so that the scalloped myc doesn’t touch agar underneath the heathy myc.  If at some point the heathy leading edge starts to scallop, that could indicate a transfer of the virus occurred. 

Or is it something else that’s not a contagion and genetic.  An “error” in the code.  Maybe the myc has an extra chromy.  It’s the nicest, warmest myc you’ll ever meet and deserves all the chances other myc deserves with a heart of gold but it probably isn’t solving the equation of everything anytime soon. 

It does appear to be a contagion of some kind tho because not all my samples of this culture are scalloped.  This is just from one dried grain that I revived.  The rest were fine.  If it were genetic, then some kind of isolates mutation would have to happen

I did take an early transfer of that original scalloped plate and put it to a 4oz grain jar.  After seeing it grew out scalloped, I didn’t expand it and spawned it to a sandwich baggie so we’ll see how it does.  It’s almost at the point it should be knotting within a few days or so.

It’s kind of a bummer.. I wa hoping the cabin would fix the issue.

Maybe I am missing something, but in the picture the growth is stil inside the cabin.

Take a look at this, I got, because I really wanted it, this FP+(F8) wedge in a ziplock bag.


After water agar and T6(I've never done T6 before) i got this.


I got my slides today, but of course they are to big, so I need to get a glass cutter before I can move on, but I will take this FP+ through cabin sequestering and using a glass slide as my roof. But scalloping seems to be bacterial very often, agar roof is sketchy in my opinion.

:rofl: That's funny, but for a noob like me it gotta make sense. The problem with a agar roof is the fact that bacteria could easily hop along the roof and agar is pretty soft and would bulged down in to the cabin...

How I understand cabin sequestering is that bacteria won’t grow through.  It grows on the surface but doesn’t burrow.  The same reason cold pours and trap doors and sandwiches work.  Unless I’m completely misunderstanding the intended use of them.  I don’t think I am though. That’s why I asked if the myc could be pushing the bacteria through with it. 

Technically the transfer inside the cabin is upside down but in my mind, when you make the cuts for the transfer, you’re pushing fragments of whatever was on the surface down to the bottom anyway.  Unless the transfer in the cabin is supposed to be shorter than the thickness of the cabin plate so it doesn’t touch the roof so it grows along the plate bottom and then up the insides of the cabin.  That doesn’t make sense to me tho.. why even have a roof at all then?

If it’s because the meeting of the cover slip and the rest of the plate is supposed to stop bacteria, why wouldn’t the roof itself stop it unless it’s being pushed through.

Every time I see this technique I feel like it’s different. I thought cabin sequestering was a variation of trenching. I thought you needed a trench around it for it to work.

Or did I just see someone do that? I still have no idea what it’s for.
Seems like a lot of effort to replace regular careful culture cleaning 🤷‍♂️

Well, the way I understanding it, the myc is supposed to grow between the cabin and the roof, not through the roof. Furter more, glass is not as attractive as agar.

It's similar to sandwich, but the roof is not supposed to touch the culture one is cleaning.

I simply rarely ever clean anything 😂 a clones gotta be super special fore to even bother but when info I keep it simple and scalloped cultures have never landed in my criteria, oddly enough the only one that's done it for me this year was also FP + though but I had good plates so I just tossed it , I'd assume it's meshed with bacterial rods but no long own a scope so idk

I'd assume bacteria most molds are pretty good at making themselves be seen eventually, IME spawning scalloped cultures usually just leads to a mess of nothing but I can't recall ever getting any mold from it
