Just more aggressive genes. Isolate that..

And for me, I cleaned up Ps. pelliculosa. 11 transfers and only 1 came out worthy enough to continue working with!

Not at home right now and wish I had a picture of the two T1 plates. Will have to share later

Edit:

The T1 plates

This is t1, should I do another or is this decent to go to grain?

Take transfers from 3 places to one plate and send that one.

LC is looking clean. I would let them go for another week before transferring. Also JMF is probably B+ or GT or any other regular variety

Hello! Noob here on my first time trying agar. Got some growth from golden teacher ms syringe in 1 out of 5 plates. Would love a more experienced opinion of it before doing my first transfer. How does it look? Is it even cube or am I growing a mold ?
Thanks in advance!

Hi! I transferred some wedges from this dish and they are all looking a bit like this one (this was the best looking photo). The original dish is still growing and looking the same, no apparent contams.

How is this transfer looking? Are those things to the left an attempt at rhizomorphic growth?

Any help is appreciated, thank you all!

Hello again thread!

I poured 20+ plates of Holy Grail and I'm waiting several days to see if my tek is solid and clean before I do some spore scrapes.

Now, I'm concerned that the 2.5" plates wont be able to show much sectoring before it hits the sides. Is that a valid concern?

I say this because I'm a stones throw from Michigan State, and they have A FUCKING TON of glasswear, and what looks like an infinite supply of sterile styrene plates for sale in a sleeve ($17 bucks per 100, not bad) and I'm getting itchy on going over and buying several hundred.

So, is the benefits of 3.5in plates soo much that I should just abandon HG plates? As a side note, HG plates are pretty good in terms of keeping contams out. I found a 2 year old one with poured agar in it and it had -no- contams present.

When using a spore swab, do you not just rub it on the agar, rotating and zig zagging as you go?

I rolled one pretty good over 5 plates and don't see anything at all in 2-5 days, did I do it wrong?

2-5 days for spore germination?
Be more patient

Like D ^ Said... that's not long at all.

However I've done that same thing before and nada but when the whole tip was cut off and buried in the agar, was successful.

Next time cut the tip off on to the last plate, since it's already been used it's not likely to be able to be used again (cleanly). Or just strands off the main to each plate.

:whathesaid:

Definitely preferred to bury the swab or tulip the swab and transfer a petal rather than using the entire swab.

By tulip, you mean spread some petals and rip off the cotton for a bury transfer?

Yes, cut the swab tip into 8 petals which you unfold and remove.
Imagine the swab like a lotus with each petal being a section of the swab.

Do you do that with a flamed blade? Doesn't that burn it?

It burns the edges, but the swab absorbs liquid from the plate and hydrates the spores
