I used a dedicated kitchen cabinet, emptys and flasks up top. But it's so dry here I think I have to put them in Ziplocs too just to preserve them from dehydration.
Micro Bio vastly different out here. You notice, but the "little things" really show the difference.

Inoculated some agar with LC about 10 days ago, how quickly should I transfer any good growth to new plates? They’ll growth is very small right now and looks clean.

I put mine in ziplocs after pouring. They dont get the glad wrap until inoculated.

Can’t seem to take my eyes of plates…:laugh:

Made a couple of transfers from some healthy natalensis rhiz…and a few plates are going vertical.

Is this because I took a slice too close to the edge of the plate as it was starting to go up sides of plate?




Is this little puppy β€˜pseudo-organised?’ It’s looked my β€˜pattern like’ over time…

I often get pretty aerial mycelium from the transferred wedge, with multiple species. Not sure what causes it but it's not anything to worry about.

Full plate looks good to my (still noobish) eyes. Natalensis has really weird morphology on agar.

Yeah not too worried about it..just curious as to why.

Has been a lot of trial and error with this strain…but plates are looking more promising now! Woohoo πŸ₯³

Hey folks, it's been a while, and I've not been idle - after these first not so good attempts, I've done 2 more batches of innoculations.

2nd batch was done on 12. July, using agar plates from the same pour as the first from 07. July.
Interstingly, no fungal growth at all on all 7 plates I inoculated. 3 had some bacterial contamination - one orange growth at the edge of the plate which showed up after 2-3 days, and 2 plates with tiny 2-3mm circular specs of white/light bacterial growth somewhere in the middle of the plates.

One possible cause of no fungal growth might be the spore solution, I re-used the solution which I had already drawn up and used on the first batch.
Didn't shake before use or anything, I recall reading that there is an effect that would cause contents to stick to the syringes walls after some time (something about static electricity or similar?) - so that might be the reason.


Now the third batch seems to be a success.
Changed the agar recipe and now I simply used a liquid malt extract, no potato flakes adding sediment and no additional sugar from syrup to add.
The agar didn't come out so well this time, lots of bubbles.
Probable cause: I had shaken up the agar bottle before pouring; I was using a 47Β° water bath  to get the agar bottle to pouring temp, but I guess the bottom of the bottle cooled faster, because the agar was already starting to solidify there.
Thought I could get it ready for pouring by shaking - and that did work - however I guess at this point the entire mass was already cooled so much that the air bubbles got contained in the solidifying agar.
Definetly felt like pouring a kind of sludge, instead of the clean, liquid consistency I had during the first pour.

However, barely any condensation on this pour, so in the end I still reaped one benefit of the low pouring temp. Just need to keep a better eye on the temps next time, or, if I screw up, just re-heat and try again (heard patience is rewarded in this hobby :tongue: ).

Anyways - this is the result so far:
Out of 6 poured dishes, 1 has a huge mold spot which has eaten up a tiny cluster of PC mycelium which I saw growing near a cluster of spores from the solution;
2 have what appears to be clean PC mycelium growth! :sun:






Would love an opinion from some veterans, but from what I've seen on posts here, and also from my previous failed cultures, this looks promising.



Bonus:
The tiny spec from my last post (from the magnifying glass shot) does seem to have grown into PC mycelium and is taking over the dish:

Hello!

I'm brand new to agar work, and am wondering what's going on on my plates. I poured a batch of ~50 LMEA agar plates and inoculated ~20 the next day with 2 different syringes, one MSS and one LC. 

I'm relatively confident in my sterile procedure up until the point of inoculation - none of my unused plates show contam, nor do a few "dummy plates" where I'd open up the plate in the SAB and go through the motions but not actually inoculate them.

I'm simply not sure what to look for when the agar growth isn't obviously rhizomorphic. Any help identifying what is growing on the plates and what I could have/should have done to isolate better would be appreciated.

The MSS batch is growing very thin and whispy, and in apparent sections. Isolations from leading edges result in the same thin and whispy growth.  Did I just isolate contam here? Or is this actually myc growth?



The LC plates all grew in big fuzzy clumps and didn't spread very far. The isolations did spread pretty fast, but were also pretty fuzzy across the whole plate. 



Were these plates ever viable? The only variable I can think to change is to lower the nutrition in the agar for the next round, as I've heard that can make myc growth stall. I'm not concerned about salvaging these plates per se, but learning from them.

Any one else have any other observations for me? I greatly appreciate anyone who can help!

Definitely listen for other opinions, but I don't see anything super concerning on those plates. Fresh germination plates and LC test plates always look more wispy and/or tomentose for me. Rhizomorphic growth is pretty and easier to distinguish as obviously what you're looking for, but it's not actually necessary to grow mushrooms, and it usually takes a few transfers, and maybe lower nutrients in the agar media, and might never actually appear for a given culture.

Bacterial contamination is usually pretty obvious - blobs of liquid that usually spread. Mold is similarly distinct, usually much more disorganized (threads growing both "out" and "across"), and will usually turn colors or develop black spots as it sporulates. There's a sticky thread on recognizing contam, and lots of examples of contamination in this thread and the "ask quick questions" thread. You can also just see for yourself what happens when you touch a plate with a gloved hand or open it outside of the SAB. :smile:

Nice first post and nice looking plates IMO. Do a few more transfers so you can get a feel for what clean, established mycelium looks like on agar, and then take the plunge and inoculate some sterilized grain!

I poured this agar WAYYY to hot, so I’m sorry for the horrible condensation in this photo. Feel free to flame me for this.

Anyway, this is a MSS plate that I took a few cuts out of 3 days ago. I set it aside just to watch and see what would happen with it. I was interested to see that the growth post transfer (at 11 o clock) is now exhibiting rhizomorphic growth.

Maybe this is common, but I am curious about this. Is this a stress response or something caused by the cut?
