Speaking of azurescens, is this germ plate trying to pin?

Out of curiosity @Adas, could you make it pin if it had sawdust or something else added to the agar? Do you or others here know why it doesnt? Do other woodlovers pin on agar?

Had 2 jars of cyans which had trich in it.. so I removed the green as much as I could. Jar was already 25% colonized by trich.. nevertheless I decided to do a little experiment.. so I plucked the trich outside of some solid chucks of mycelium (3cm diameter). And added some ripened old chips which were laying there in the garden and had already been mixed with some soil and have been laying there for some months... Just grabbed a few hands of that stuff and added the mycelium chucks (didn't break it up).. was laying in the cold outside open to the elements.. (sheltered from the rain though) 7 days later.. didn't see any trich pop up.. so I moved the bags indoors.. again week later.. still no trich.. decided to add some more spawn now which I had laying around. But yeah.. maybe a way to save partially colonized grain without having to start all over? I wonder if a natural biofilm just fights of trich..

Added more spawn intentionally, just to see what will happen..

I mean I guess you could, but you'd also have to keep it for a very long time to starve it, and then introduce the right temps. MAYBE then they could

I personally have never observed knots in my in vitro experiments ever, but my friend who expands the myc using just woodchips had this happen at least once. It's still a mystery to me what exactly causes knotting. The knots were on the woodchips against the container, not on the surface.

I have a cyanescens plate going that really looks like it's trying to pin (apparent knots, and a couple brown points that look like early pinheads). This plate is also hilariously contaminated and the mycelium appears to be trying to hold off the mold at one end and racing to the edge of the container (as in your friend's case) to fruit at the other. I'll see if I can get a picture, I'll just need to open it somewhere else. FWIW, as mentioned earlier I do use wood soak water in my agar- but I would expect that to mainly be primary decomposition-esque sugars and not lignin. Hell if I know though.

As far as knotting is concerned... I think you actually said it- starving has got to be a factor IMO. It seems they are very sensitive to the availability of nutrition, and when that runs out, and especially when the furthermost strands run into a barrier (guaranteeing no further nutrients along that path), that's when they tend to pop up. Plenty of examples of outdoor wood lovers showing up along edges of garden beds, sidewalks, etc. and I've seen this observation/theory floated a number of times before.

No way can I say this with any authority but if I had to guess, this is part of why cultivated wood lover experiments seem to a) almost invariably take about as long as wild ones to fruit- 9 months or so?- and b) produce so much overlay and often won't fruit at least for a long time in fully colonized chip tubs. Compared to the nutrients available in dune grass roots and partially decomposed hardwood, spawn coming from grain with a concentrated abundance of uncolonized wood is probably pretty fat and happy for a while. The need to reproduce wouldn't become urgent until they've really depleted that supply, which is gonna take a while. (Agar might be different because it's also less nutritious than most cultivated substrates.)

All speculation though.

Here's the plate in question. Like I said, the situation is p ugly. That divot before the strands pick up again was from me trying to swipe enough for a new plate.

And... enhance!

Enhance again...

It's weird, right? If those aren't knots, my best guess would be that's where the mycelium is overtaking the mold and the discoloration is from metabolites produced in response to that collision. But it doesn't quite look like that to me. And the bigger ones look suspiciously different.

Finally, and I'd be curious to hear from more experienced cultivators on this (so pretty much everyone), that sort of "fan-tail" pattern of growth- a long combined strand that frays and fans out at the end- seems to show up late in the cycle around when knots should or do start to form. Sometimes the exploratory growth on chips does that too, but something about the shape feels different to me.

Time will tell!

I'm not seeing knots, it appears to be a continuation of the contaminate patches outside of the myc.

But hey, let it grow.  FAFO.

Oh yeah that looks heavily contaminated to me.. when I move too swiftly in the sab or drop something.. bump the tote, I can get a few contaminated spots on my plate. Apart from that your transfers should be clean.. even if you end up exposing receiving jar/plate for a number of seconds and multiple times. but this is quite a bit.. you could start with germination on water agar for T0 next time and then move clean parts to normal agar, but yeah would expand this more agar and see if those stay clean.. if the plate stay clean but contamination rides along try water agar to clean them up, I would wait with moving to grain untill you feel confident. Good luck!

Ya that's not knots. My Azzies turn blue and cream even being completely sterile. It's just a normal aging reaction of the myc, especially on nutritious substrates.

Thanks all for the input!

Fortunately this is just one plate of a few going from that print. It's definitely been a process trying to isolate them out, but I let that one keep going because it seemed to be pushing back against the mold and it's always interesting to see how that plays out on agar. The transfer on that one was before it got nearly that bad so it should be recoverable eventually.

Woodlovers can be very easy to grow and to maintain in the right area. I received a simple envelope with a handful of colonised Ps cyanescens woodchips, expanded the chips, made some beds, and 20 years later I can still harvest from said patches, mainly due to the fact the gardeners that maintain the area keep adding new woodchips to new places in the area (I did nothing to expand, just spread some of the stembuts and some mycelium in new piles of wood.) I don't know weather the spores from the original beds keep spreading, if it's the mycelium that keeps expanding, or it's purely the great habitat, but I love the fact that I did a little work 20 years ago and still can harvest every autumn around said spots and have the feeling I attributed to that. Plus it's public domain, so anyone who can recognize them, can take a bunch.
