Im willing to bet that if you grow mushrooms in here without going spores to agar - nobody will show up at your house to kill your family.

I HATE swabs, wasn't there a whole thread of peeps trying to
make swabs work? From wetting to fuckin breaking off the tip to
stick it in the agar. I got nuthin, nuthin but nuthin!  :rofl:

Well that goes back to the roots of LAGM.
I think there's always been a push to get noobs past agar fears.
And Comebackkid pushed that so that this affair is really to get
new peeps into agar.

Swabs are fun. :mad2:

I always do 2-3 plates per swab with last one being a dunk. I always get nothing on at least one plate but always get something on the rest. Sterile water soak and getting rough is the key. I'll be posting a video of my process for tomorrow. Of course this time it won't work 🙄

I will be growing Ps. Cubensis.  Ghost and Jack Frost varieties (much thanks to The Doobsker and everyone else who has helped me with spores, I am very grateful).

hahahah yeah. I'm not interested in any kind of prizes that come out of this and I also won't be able to inoculate everything on my list on NYD anyway so I will be a week or two late for some of the species I'm growing. I just saw bigfoot looking to get some non-cube varieties added into the mix so figured I'd jump in and share what I have going on.

That makes sense - I am always amazed at how many cake grows I see from newer growers, which is crazy to me in the year 2021. Once I got good at agar, I looked down on cake grows until I realized they are sometimes the ONLY way I could get growth from a syringe. Then it occurred to me that if you are doing MS and just looking for a good fruit to clone from, or if you are trying to stabilize a specific culture, cakes would be the fastest way to do it, so I think they still have a useful purpose.

Yeah funny story smartattack I actually got this swab from a complicated trade triangle with some folks and uh I think they're actually from you originally! lol! Good info though thanks I guess I need to PC a li'jar o'water soon

The streaking technique is dope if your goal is isolates, I've gotten one as fast as 4 transfers using it. Basically serial dilution on the surface

But do you really want isolates? Why?

If so, you need to test LOTS at a time to find a good one

For most people, you are better off going spore>agar>clean culture>grain then cloning some promising fruits/clusters, then testing those clone cultures  to find a good one and focus on that



I was steered away from wasting my time finding isolates by c10 years ago. You dont need an isolate to achieve what ur after. Infact you can waste a lot of time chasing a atrue isolate and it might not even be good. But if thats what ur after c10 has a good streaking tek for getting closer starting from MS.

RUSTYDICKS, 7th GENERATION!

January 1st

Got way lucky with the timing and could harvest today an MS tub of RustyDicks (F6) from which I took a couple swabs just in time for the new year and my LAGM '22.

I used 2 swabs from the fruit pictured (a 167 grammer from the first flush) to inoculate 8 of my 'ol trusty BRF/verm pucks:




This grow was struggling from the beginning and yield was likely compromised because I'm currently experimenting and messing around with somewhat aggressive grain preps (chemicals were used to wash and rinse my dirty ass grains, never again :facepalm:).
Here's some pics of the fruits from this F6 grow for reference:






Cut most of them in half, mostly solid!



January 17th

Late update considering I inoculated some pucks with a swab on the first.
Did the lazy thing to do and used pucks that were a bit old and definitely dry as a bone as I discovered days later.
Only got germination on 3 pucks out of 8, and growth on those 3 pucks was literally crawling, almost stalled.

The plan today was to transfer some tissue from the dry pucks to fresh pucks (which I did). But then I did 3 LC's too with the little tissue I had on those 3 pucks, like the one pictured:



I like to let things grow more from spores before doing LC so that it gets easier to see what's going on but the temptation of turning that little initial growth on those sad dry pucks into 3 potentially clean LC's was too strong, especially when all it takes is poking that shit with a needle. Game is on!

February 6th

Kept the three LC's in a rather cold place so they took longer than usual. Also waited some days more after fully colonized so that I could use my only day off (today) to inoculate some grain.

Here's some pics to go with the words. 3 LME LC's with a bit agar in them:


These gave me no reason to think they aren't clean so today I aspirated 60ml from each LC to be tested... on a good bunch of grain jars. :cool:


February 10th

The 3 LC's recovered just fine, albeit a tad slow due to low temps but they seem healthy otherwise.
36 jars were inoculated (3-4 ml per jar). Picked one jar from each culture for the pic:



Growth always starts off a bit wispy on grain with these but it thickens up considerably after a bit.
The myc of this cross always reminds me of the OG RustyWhyte.
Will post pictures as they progress.

February 19th

Was planning on updating the log the day I gave the jars a second shake but life is hella busy lately.

Here is a small sample of the jars from each of the three cultures. Some nearly done like the ones pictured, and many fully colonized already.
Extra cottony, fluffy growth this time, not exactly picture pretty:

Going to let these sit a few more days before spawning and then decide what I'm going to do with the 36 jars.
The original plan was to make 3 tubs from each culture (9 total), using different fruiting strategies, as these have shown a tendency to blob on these last generations. But that's also a lot of work lol

February 27th

I let the jars sit for a few days after full colonization was achieved and today I made 3 tubs, using 4 jars per tub and culture (1:2 ratio, grain/coir). No top layer or casing was applied (yet).

In addition, I spawned 4 jars from culture 8 (the best looking one), split into 2 buckets (2 jars per bucket, 2:3 ratio), and also did one bucket using 2 jars from culture 6.

As far as pics go, nobody needs to see some freshly spawned tubs. :lol:

March 29th

Just harvested the 2 buckets from culture 8 which produced some serious blobby action.
I decided against casing them in the end because I'd rather put some more effort into finding genetics that don't need a casing (obviously not the case here).

Got the fat guy on the upper part of the picture and took swabs from it.

I'm also seeing the beginning of what might be something interesting from culture 6. A couple of early birds fruits (just like ol' RW likes to do) and not a blob in sight yet:


There's some more stuff coming in a few days.

The following is a fat pin growing from the germination pf-tek puck used for this growlog, culture 8 to be exact.
It's actually a "normal" fruit, only that the cap is facing against the glass and can't be seen in the pic.
Took some tissue samples to agar:


The plan is growing out this invitro clone in the hopes it doesn't need to be cased and maybe take swabs from it too.
At the same time I'll knock up some plates with the swabs I took today, just in case the F8 grows from these spores turn out good.

I also inoculated an LC a few days ago with the F6 culture that was pictured at the start of the log. Planning to grow it out again and take some swabs because I don't know where I put the swabs I took  from that grow :facepalm:. Just in case I have to go back to F6 and start again from there.

April 2nd

Culture five doing gnarly things!

Lots to take in but would you take a look a the fat feisty lady on the upper right corner showing off her old rusty upskirts... indecent!




April 3rd

Some second flushers from culture 8 getting fat. Will take some swabs if/when they open up.



April 9th

The second flushers from culture 8 are done. Was waiting for them to open up but they didn't and were starting to get soft and degrade so I picked them. Some third flushers coming in, too.



There's a final update coming in a few days. Also had to bin the other day a tub that triched out, think it was culture 5.


April 10th

Ok so those early pins from culture 6 became the whole yield from that bucket, disappointing.

As long as agar is involved I'm good with it  :thumbup: I'm actually very fond of brf transfers to agar so I get it. Plus you're not doing cubes so you get extra leeway lol
