06/01/21 Update

On 5/29/21 I made three transfers from one of the swabbed plates. I would have made more but didn’t have any more plates to use. The first two pics is what they look like today, two days later.

The last picture is of the top part of the stem that was all black with spores that I had transferred. Kinda cool how it looks don’t you think with the different types of myc on the same plate?

These look clean to me now so do I really need to transfer again? Am I being niave to think that? 

I'd sure like to try a Josex Poke on that last plate if you guys think it's good to go... somewhere from the left side... but that plate is a biopsy with no transfers made from it yet...

I don't quite know what I'm looking for in 'clean agar.' To me, clean is basically nothing else growing, no spots, no goo, all white. Lot's to read out there... I better get on it!

Of those 3 lined up, the last one looks the best but I'd still do a few transfers and see what they do... you want a more uniform growth pattern. Or at least that's what everyone says... lol. I like to take as many xfers as I can to see how things work. But im also team Full Send... if its clean, get it on grain! Lol.

So yeah, take a few xfers if you can, if not, get it chowing down on some grain!

  • How long do you wait, how far do you let the myc grow out, before you transfer again? I mean is there an ideal time to transfer?

  • lol... that 3rd pic certainly is. Is it not good to let them grow out that much before you do a transfer?

I like to let it get a bit more than quarter sized to show some identifiable traits. But I have let it go much longer many times. Ideally you want to take transfers before the myc gets too close to the edge of the plate because of the increased risk of edge contamination.

You could take transfers on the first two, but I would give it a day or two more to see if there are any clear areas of faster healthy looking growth.
The third is getting pretty close to the edge. That is not actually fatal, but you do run a higher risk of grabbing an unseen contam with the transfer by the edges.

Lost A/C for 3 to 4 days. Inside temps got to around 90f/32c so the tamps never recovered well. I figured this was the end of LAGM2021 for me. So I decide to look for stones.

My first stones.
Again, not many pulled from the sub.
But the sub was pretty dry after the A/C issues, so I decided to soak the crap out of the other tub and let it go a bit longer to see if I get more stones from that one. After another week, it even started pinning again. So LAGM2021 continues for me for a bit longer.

#1- It depends on your intentions... if you're looking for fast propagation, you'll want to select faster myc, earlier... in that scenario, faster = better...

#2- yes... lol. You can take transfers as soon as you can see mycelium...

#3- ill let em grow and over grow... I like to use plate pins... no sense in doing all the work and not seeing it thru til the *proverbial* end, right?

Week 22 Check in...

Texas Orange Caps - T2 Plate to WBS


Little bit of a lull in my LAGM21 action.  I lost 3 of my TOC T2 plates to some type of invader.  I think it was because I let my plate area get dusty and then I was handling the plates too much. It's hard to resist picking them up and turning them over and lookking at the myc. The 4th plate looked okay.  I should have taken transfers to T3 but... I split it and dropped it into two 1/2 pint WBS jars.  Growth seems a little slow, so it may be infected, but I'm going to let it grow out.

Trying to decide if I want to re-start TOC just in case these jars don't make it or if I want to try APEU or PESA. If I don't restart the TOC, I'll have to do some reading to make sure I have handle on the right shoebox conditions for APEU or PESA.  The good thing about starting for spores, I'll have plenty of time before I have to put it to grain.

I realize there are all sorts of growing environments, mine being a dedicated 10x12 room... and since this project is bringing these thoughts up again I'm going to ask. I apologize ahead of time if this is not appropriate for here... :bow2:

How do you all implement good practice and sterile techniques?

I want to be fast, clean and efficient :super:

I know it's all important, but there's got to be a happy medium somewhere? Between all the various tek's there's:
  • room, air, and SAB prep where you wipe and spritze EVERYTHING down with ISO, bleach
  • Lysol or Oust for the air
  • spritze everything that goes into SAB with ISO
  • shower, brush your teeth, put on clean clothes, cover hair, mask up, put on gloves
  • spritze everything again and spritze your gloved hands again if you take them out of the SAB
  • and then there's the tek about working in a dry SAB...


What if you simply put on a clean long sleeved t-shirt and didn't worry about the bottom half of your body? You're supposed to be sitting still and not touching ANY of your body while working in the SAB, right [wink wink]?
  • Or maybe soapy water spritze your lower half to keep any contams from flying around?
  • Or maybe wear one of those longer lab coats that cover knees on up?
  • And then if you do wear a lab coat, do you have to wash it between each use?
  • If you hang the coat in the room on the back of the door does it gather dust that can cause contams?
  • If you don't wear a lab coat but a long sleeved t-shirt, since you're only working in the SAB could you reuse the same t-shirt?
  • Would spraying down arms of the t-shirt with ISO and soapy water work if wearing it again?


Do you take your shoes off before going in the room?

As for the room prep
  • you already keep the room clean as in periodically dust and mop the floor
  • you've closed all windows and turned off anything that causes the air in the room to move (AC, fans, HEPA filter, etc.)
  • and you'll be working in the back portion of the SAB.


Would it be enough to clean only the immediate area, SAB and SAB contents?
  • make sure your immediate work area (like the top of the table you'll be working on) is wiped down with 70% ISO
  • wipe down inside of SAB and spritze with soapy water
  • wipe down everything that goes into the SAB with ISO as you put it into the SAB


And what if you have to leave the room to go do something like turn the PC off that's in another room?
  • Do you have to start all over again with clean clothes, new pair of gloves, etc.
  • or can you just spritze your clothes and spritze/wipe your gloved hands?


My mind goes in circles over this... :facepalm3:

I may get shouted down, but I like the 80/20 rule. The first 20 percent of effort gets you 80% of your results. The closer you want to get to 100% perfect, the more effort you have to apply.

So you can do all of those things to try to get as close as possible to perfection. But ime it doesn't take nearly that level of care in a normal environment. I have been using a SAB that I set up on my Kitchen table. Sure, I wipe down the table and and use a fairly generous amount of ISO from a spray bottle on gloves and anything I put in the SAB. I torch my scalpel and wash my hands (and hairy arms). But I don't even turn off the AC. Are you kidding me? It will be 110 here this week.

I try to use care and if I find that I am having contam issues, I try a bit harder to tighten up my technique. But I have had a lot of success without driving myself too crazy with it. Environments will vary so what works for me may not work for you. I suggest finding a middle path and trying it. Then if you get good results, you can try being a bit more lax. If you are breeding a crap farm, take a few more precautions.

Don't get me wrong. I would love to get a ffu and set up a dedicated lab. But this is just a hobby and passion project for me and I already get better results that I can handle. Trying new stuff is half the fun.

I love your enthusiasm! I've been if front of a hood for over a year so sadly, my SAB advice is gonna be less than helpful...

I honestly had no problems using my SAB dirty, after work, same-ol-clothes.... I quit spraying my SAB down early on... the biggest thing is slow, precise movements... hell, half the time I didn't even wear gloves with my unwashed hands...

It's hard to know when you're unfamiliar with all this rather scientific stuff, no-one to bounce things off of and need to be fruggle and careful not to error or waste. Working in the flow is much more forgiving... I tried to go back to my SAB once after I built my hood... couldn't figure out how I had grown anything!! I have mad respect for all the SAB warriors out there!

And, just so you know, I have a list of all the members that will get a free flowhood when I win the lottery, and your definitely on it!!
