Species: P. Cubensis
Varieties: KSSS, B+

Tried (and failed) a couple of times previously with the infamous "broke boi" TEK but this will be my first time working with agar.

Edit 1/1:

Made ~200ml of LMEA and poured 8 90mm petri dishes. I probably could have done the entire sleeve of 10 but I poured them a little thick. There's definitely a learning curve here, but when I do my first round of transfers I should have an easier time.

Four plates each of either variety, both from MSS. I used an inoculation loop. Excited to see all the wonderful bacteria in my dishes!

14/1 Update

One plate of Amazon germinated well, took a couple transfers today. I haven't been doing agar work in a while so my cuts were not super clean but I think the myc looks good. Here's a post-shot of the germ plate. Vets, how does it look?



21/1 Update

Finally got around to taking T2's. Growth is still disorganized as all hell. Hoping there's something nice that I can take to grain by T3. Need to whip up another batch of no pours in the next couple of days. Had a vendor for cheap plates on mercadolibre but they've all disappeared and been replaced with ludicrously priced petris so I may bike up to the science store this weekend.



Bonus pic, here's my little habanero. About two months old, just moved her to a new pot



28/1 Update

Got some real petris poured. Oh my god the clarity. Probably just gonna use the no-pours only for germination at this point, if at all. Anyhoo, I put the rest of the plates in the sleeve, where do I need to store them? Is room temp ok? Hoping for some pretty myc, I'm super paranoid that there has been some bacteria hanging around in my no-pours that I haven't been able to see.

4/2 Update

Alright so after doing more reading I think that the clear halos on all my plates is just happy myc in a nutrient rich environment. Took some T4's today and I've got a candidate from T3 to go to grain in a week or so.

Let's do this!

I will be growing some P. Cubensis: Melmac (Homestead PE) and PES Hawaiian.

Jan 1:
Used a Melmac spore swab on 5 agar plates. This is my first time working with spore swabs (in the past I have used spore prints and syringes). Used the method of tearing off a bit of cotton from the swab with sterilized tweezers and spreading that among 4 plates. For the 5th plate, I just broke off the end of the swab and left it there.

1/2/2020
Just streaked 2 plates with PF Redspore.  Had a spore syringe, soaked a sterile swab with the syringe and streaked the plate.  Repeat.  Agar blend was 3g DME, 3g agar and 150 ml distilled water, poured into 5 plates.  PC for 35 minutes in an instant pot.

It didn't really feel like I did anything, but will find out in a few days. :tongue:

Late to the party but I finally swabbed some B+ spores to two plates along with some t2 TOC and RW. Not much to update as of yet but here we go!

LAGM 2021


1 JAN 2021

Plates were poured a few weeks ago, standard LME agar. Swabs arrived in the last few days from generous Shroomery members. In my SAB I swabbed three plates each of APE and Melmac, burying the swab into the agar on the last plate. Now we wait.



6 Jan 2021

I’ve got germination on my APE and Melmac, but only where I jammed the swab into the agar. My streaks haven’t taken off yet.

Some nice myc off the Melmac swab.


Just the tiniest bit of white on the dark background of spores on the APE swab.


9 JAN 2021 update

Low temps in the house have really got things moving slowly. Everything still looks clean, so that’s good. I’ll wait a few more days before I take any transfers.



13 JAN 2021

So this is what I’m working with twelve days in. Still have nothing on three plates, but I’m going to move along today. I’ll grab that rhizo growth breaking free on the Melmac plate, and something white from the APE plate.


Hope I got some clean transfers. Everything seems so slow.


Also moved the swabs to new plates. There’s still so many spores on them.


20 JAN 2021

Had to pour a sleeve of plates today, as well as do some other stuff in the SAB, so I figured I’d make some progress here too. Took a couple of transfers from the Melmac plate that’s looking solid. Also grabbed a couple from a sketchy looking APE plate.


My growth has been slow, and not all looking the way I’d like, so I also went back to my germ plates to take some more T1’s. The germ plates were a bit better looking now that they have grown out a bit.


I decided to try a little experiment while I’m waiting. I think I picked the slowest varieties at the time of year my house actually cools off. Maybe this will get me some fruits quicker, or it will be a total shit show.


28 JAN 2021

A few days late updating, and no pics, boo me. All my T2’s look fucked, so they went in the trash. My second round of T1’s I took last time actually look pretty good, so an APE wedge went to a pint of grain for g2g. Also blended a Melmac plate into LI and dumped a bit into a few quarts. Speaking of grain, I’m teaching my girl to grow and this was her first time prepping and PCing grain. I only had shitty WBS on hand, but she did a bang up job of it. Might have underfilled the jars a bit, but it will work.

6 FEB 2021

Melmac on shitty WBS a few days past full colonization. Spawning these three short quarts to two shoeboxes today. The pint jar is a wedge of APE that will get a shake today.


12 FEB 2021

The two Melmac shoeboxes are moving along nicely. I’m going out of town for a few days, and expect some little mush when I return.


APE recovering from a shake a couple days ago. This stuff is slow.


18 FEB 2021

APE almost ready.


Melmac did not disappoint! Not a bad pinset for an MS culture.


20 FEB 2021

APE transfers as insurance. My pint of WBS looked sketchy, but I put it to 2 quarts of wheat to see what happens.


Melmac shoeboxes coming along.
