How do you guys feel about "treated" vs "untreated" petri dishes?

Almost all my plates have this wispy border outside the rhizo growing, is this bacteria? How I get rid of it? just doing more transfers?

It's the leading edge of your mycelium beneath the surface of the agar. You often see this with germ plates before they fill in. At least it always appears to be just below the surface when I see it, it could be the case that it's sitting on the surface but hasn't quite filled in yet.

Bacteria looks like a milky residue on your plate.

Cannot comment on the treated vs untreated question. Those celltreats are pricey in comparison to what i've been getting.

My normal 500ct are out of stock so i'm going to have to grab some of those or the Thomas brand ones on the Zon.

When I first ordered dishes I bought treated ones and have seen no difference between them and the untreated I use now.

I really like the grip ring celltreat dishes. I wrap mine outside of my SAB so it provides a little more peace of mind there even though I've never really noticed problems doing it that way. The lid doesn't slide around if you are holding it by the side with one hand but I'm not sure if they're worth the extra money.

As far as I understand, it doesn't make a difference for our purposes.  Treated petri dishes are made hydrophilic for adherent cell cultures that grow under a growth medium so that they can adhere to the dish, otherwise they grow in weird clumps.  We're growing cultures suspended on agar, so it doesn't matter if the plastic is hydrophobic.

Hi guys, it's been 1 week since I put GT ms syringe to agar and one plate looked ready for a transfer while the others seem like they need 3 more days until big enough. Interestingly it's the top plate in the stack so maybe because it got more light it grew faster.

Unfortunately the lid of the petri had too much condensation to take a Pic so I did 3 transfers and then took a Pic with the lid off. Would you guys comment on if it's healthy mycelium and my cutting?

First Pic is the transfer, second is one still growing in a zip bag. Third and fourth Pic is the same dish, just trying to get shots of the spores growing at the edge.

I transfer with a needle biopsy as soon as I see germination and would feel comfortable transferring from your plates at their current stage. The growth on your 3rd plate near the edge seems a little suspicious, since it's so close to the edge mainly, but its difficult to really evaluate it from that picture.

Oh that's good to know. So I shouldn't let it grow out bigger then. Guess I'll transfer the others tomorrow after making more plates today.

It was hard getting a pic because of the glare, but it is a bit more clumped up and fuzzy. Maybe I'll avoid that plate

The grip ring are nice and sturdy but when I pour a little too hot the condensation takes forever to clear up in front of the hood.

I never had any problems with the slippable Celltreat either they were as strong as they needed to be honestly and I liked how light they were.

I've also noticed that condensation is worse with the grip ring plates but I took this as a testament to the efficacy of the grip.

I have 5 grip plates that I didn't bother wrapping and there is still a light layer of condensation after almost 2 weeks.

Gang - for the first time, 600+ plates into the game, I finally did 25 plates of germination/transfer with only ONE contam. Bout time!  :smile:

So I germed some melmac swabs from QM - nothing came up except on the one where I actually brok the swab-tip off in the ag.  I took transfers from that small bit of myc.  They're growing super unevenly and weakly, it seems. I'm gonna transfer again, but can someone tell me what they think?
 
Second batch is from LC.  I made 4 different bottles, using 1 plate of B+ that had "gone to pin" and 2 syringes. Used a sterile swab in the bottle, then swabbed on plate.  One has a couple bacteria blobs, but they aren't on the actual parts where I streaked, I don't think (number 2 in this row).  Any thoughts on these?  Couldn't have done this without y'all.  Reddit, other forums, even published books--nothing has helped me like this community. Cheers!

Both of these plates are direct from the stipe clones, T0, never transferred.

Can I go to grain or should I do a few transfers. I see some disorganized growth of :shrug:, so am unclear how to prudently proceed.
