As the dikaryotic hyphae lengthens it forms clamp connections between septa to transfer nuclei. Is this what you're getting at? Or do you mean fusion of the nuclei?

"The dikaryotic phase is technically not diploid, since the nuclei remain unfused until shortly before spore production.) "

"Each basidiospore germinates and generates monokaryotic haploid hyphae. The mycelium that results is called a primary mycelium. Mycelia of different mating strains can combine and produce a secondary mycelium that contains haploid nuclei of two different mating strains. This is the dominant dikaryotic stage of the basidiomycete life cycle. Thus, each cell in this mycelium has two haploid nuclei, which will not fuse until formation of the basidium. Eventually, the secondary mycelium generates a basidiocarp, a fruiting body that protrudes from the ground"

This should probably be its own thread.

Just started a thread, didn't see this until now

This topic has been discussed across too many threads. Post this topic there.

I was more talking about the sharing of genetic material as seen between two monokaryotes not the fusion of individual haploids which only occurs when spores are formed. I see people talk about "dikaryotes fusing" and was wondering what exactly this meant. I believe septa only allow material interchange between cells of the same dikaryotic hyphal strand.

But yeah, if we're branching into new thread territory that's fine.

EDIT: just saw your new thread link

Before a dikaryon creates 4 spores it goes through karyogamy (the fusion of the two nuclei) and then meiosis. Each set of those spores contain a random sequence of chromosomes. If a hyphae from spore A meets a hypae from spore B they become a dikaryon (assuming they’re compatible sex types)

In the case of crosses one hyphae from say Golden Teacher meets a hyphae from AA+ then the resulting dikaryon will produce 4 spores containing a random set of genes. By selecting and prolonging this process people are successfully able to stabilize a new cross, like Pasty did with RustyWhyte and Ted and the tribe are doing right now with Trump Dicks.

Exactly.

Here are some up close shots of cube mycelium. Seperating out a single thread and knowing you had done so would be pretty much impossible .

The first two pictures are a "clone" (tissue transfer from a fruit). Even the ones in these shots that look like individual strands could be 10-12 or 100 running together. They are not bound by limitations of relativity when it comes to cellular sizing lol. They are very plastic creatures and very aware.

The best methods I have seen involve a lot of calculated spore dilution (I'm sure there are better tech terms for this) and a lot of staring through a microscope and counting spores on a grid. It is totally unnecessary IME in order to get stellar results with cubes.

Great practice for working with cordyceps though. They are weird critters.

Very light fluffyness that is on top of the mycelium. It looks normal from first glance and can trick many unless you look closer. Between the rhizo strands you can see something else growing, which is white and fuzzy, and looking at it horizontally you can see the raised fluffy growth. Sometimes certain cultures and nutrient contents on agar can create similar vertical fluffy growth but it does take a trained eye to notice the difference, I can tell this isn't cubensis mycelium because of the way it looks comparatively to a clean culture of the same genetic

I think things will line up well for you. You have pretty mycelium and good things always tend to follow close on with pretty mycelium.
