what's the longest you guys would allow your fully colonized dishes to sit out in room-temp before you want throw them in the refrigerator for storage?

I have a bunch of plates ready to test and i can only do so many at a time- are they okay to leave out?

most of my plates are okay- but some have been out for like two weeks or more now.

Potato flake agar that I decanted per suggestion from shroomery user Smartattack. Super happy with the result! I know its easy to get nice plates with LME but I wanted to use what I had for now.

I’ve used plates that sat out for a couple months. If I have a lot going I pour them thicker so they last a long time, but they do usually have old pins by then.

i see.

obviously we dont want to have our dishes dry out- but is it bad if the plates start pinning?

Yeah it’s fine.

I’ve knocked up jars using old plates covered in dead pins many times. Just takes a little longer to get moving.

I'm really struggling with satellite colonices lately. Not lately really, to be honest all my life since I got into agar haha but now I'm noticing them a lot, more than before.. I'm now almost a year and a half continually working with agar and never ever could get 100% rid of these .. I'm wanna throw the SAB through the balcony :mad2::mad2:

I'm trying my best and still having around 20-30% of my plates getting these, that's a lot!!:mad2::mad2::mad2:
I know it's my technique. I know it 100% but I don't know what to do now, I don't know what I'm failing at. I spray down my SAB before working, use a rack to separate the dishes from the floor, sterilize red hot the flame, steady but precise movements, avoid passing my hands on the dishes.. and still struggling with these fucking colonies!!!!!!!
I gotta go back to the fundamentals.. what are your best tips to avoid these? Any videos that I could check (Besides munchauzen and bod)?
Thank you guys

God damn I just realized I'm writing this just right after ending a session.. Ill try videotaping a little bit for next time

My satellite contams went to zero when I quit spraying anything in the sab. It doesn't clean shit anyway.
I know nobody wants to hear this, but that culture looks like mold to me.

Oh I forgot to mention it. Just saran wrap.
And now that you made me think, it might be that.. I'm noticing it more often since I bought the last cheap roll. I'm gonna buy another one tomorrow and see if it starts to improve

Well that's odd to hear. I'll try that way too then. I'll do anything to stop getting this shit :mad2:

thats what i thought lol. here is that pin plate. i had taken 2 transfers out of it a while ago, and yesterday i cloned the 2 biggest/first pins to pop out. Realized I could put this plate to grains first instead of waiting for the clones/transfers

gabbk,

use a dampened towel on the floor of your SAB as well- it well help catch all the falling particles in the air and keep them from potentially blowing back up when you work. Keep spraying the walls with soapy water for the same reason- but dont stress it if you've set everything up and its time to work but some spots are dry.

are you waiting an ample amount of time to let everything in the air settle on the surfaces before you work?

make sure you work in a room with as still air as possible- if you are using some sort of glove or gasket for your arms, back when i used a SAB i  found it better to just cover the holes with paper while i waited for shit to settle and then just left them as open holes. set up away from a window or keep it shut if its nearby, keep door closed.

these are the precautions i took when i was working with a SAB and had a mostly successful experience.

Thanks for your time Moshe. Yes I do wait a couple minutes for everything to settle and when I'm done I cover the holes with the same lid of the SAB. I do everything you said except for the towel. I used to do it but I got lazy and starting forgetting about it. At that time I had better contam rates as well now that I recall.. so I'm definitely going to implement it back again:thumbup::smile:

i bet the satellites are coming onto your plates because you're not waiting long enough. try setting up and waiting an hour before you work on the next batch.

I read people waiting much less, but i don't think even 15 minutes is enough for everything to settle properly, and its better to be sure than do it on guesswork.

Yo! Thinking I have some bacteria riding along both these plates

PE t3 has major off white discoloration in middle, also is wispy opposed to outer edge

APE t1 has that film surrounding myc - is that normal or bacterial?

If bacteria’s the case, would it make sense to keep taking transfers until
Culture is clean or is that just a lost cause?

Yes you are right Moshe I've always waited for 15 míns. I'll prepare it everything ahead with one hour:)

Lectric, I don't think that's lost at all. I'd hot pour these plates but first transfer the outter edges. I bet it'll recuperate quickly
