Big or small agar wedges to jars

So im getting close to transferring my agar plates to grain and im just wondering what would be better a whole bunch of small pieces of agar or 3 or 4 bigger pieces im gonna have enough plates to put one plate per jar so i could make lots of agar wedges for the jars and speed up colonization also should i shake the jars immediately after putting agar wedges in or just leave them on top of grain until they start colonizing then shake the jars

I was doing that for a while, now I just scrape the surface of the agar with an inoculation loop.

approximately 1cm x 1cm put 3 pieces in whatever your colonizing and shake :brainscream:

First off, I use no-pour half-pint jars that have been PC'd.  I then scrape the surface off the agar with the inoculation loop (after sterilizing with an alcohol lamp) into the cracked lid of the grain jar.  I do this partly so I don't have to drag the piece of agar through the air.  I just slide it into the jar like dumping spaghetti into a trash can.

i have 1 plate per jar so i would be using alot more than 3 pieces but ill do 1cm by 1cm pieces

So you pour the mycelium liquid into the jars and leave the agar in the no pour ?

well that's when I'm making the agar.  but what i was talking about comes after a colony has been established after about five days.

you should just shoot over to a no-pour tek to understand that part better...

All up to personal preference.  You can do the tic tac toe method, especially if you're using no-pour plates.  Mentioned here:

You can do the same thing with pour agar plates, but it's more tedious. 

Personally, I just cut a very large chunk of agar or two or three (depending on how many clean plates I have ready at the moment) and dump them in. 

Small pieces = more inoculation points
Larger pieces = easier to get one section of the jar to colonize quickly and a little bit less tedious and less chance to introduce contamination into your dish/jar due to more time spent cutting on the dish.  However, if you're sterile technique is on point, you don't really have to worry too much about the higher risk of contams being introduced.

But, assuming your plate is clean, grains are clean, and technique is sound you'll be fine whichever way you choose:goodluck:

I wouldnt put an entire plate to a jar. The point of using wedges is to leave behind latent/invisible contamination. I use a .5x.5x.5 triangle and either throw the culture in cold storage or in the trash.

If you have contamination, you shouldn't be using that plate on your grains.  Nothing wrong with using a whole plate to a jar if you have a clean culture.  People have been doing the tiger drop for years now with no problem.

No no youre definitely right

OP, dont take what im saying as its okay to use a plate with contamination on it. If it is visibly contaminated ANYWHERE on the plate, its a toss. Or try to clean it.

What i was trying to say was, if there is anything wrong on the plate, it will be exacerbated by grain. I have always heard to use as little of the plate as possible, but maybe that's outdated?

If your plate is clean use as much or as little as you want, I usually cut a cross then a circle about 1-2cm away from the edge. Sometimes I make a quick cut to remove the donor agar, this gives me 4 big wedges and I use anywhere from 1-4 per jar depending on how many jars I need to make, usually 2 because I like to have a backup.

If your plate aint clean then dont use it.

A larger wedge will colonize quicker and faster colonization usually means less chance for contamination

The way I like to do It,
1 agar puck to a grain jar.
Chop the puck into 20 to 30 pieces in the jar.
Put lid on jar and shake lightly until agar pieces are distribute evenly..

Use as little of the plate for a transfer normally, for the reasons you mentioned in your previous comment.  Leaving behind possible contaminants that aren't readily apparent or maybe aren't so obvious to a more novice grower. Also it's important to take a small transfer wedge if you're trying to minimize genetics, such as going for an isolate, although that isn't exactly applicable to this situation. 

However, when you go to grain you should be sure that whatever piece you are using (whether it a wedge or a whole plate) is 100% clean.  If not, or if you're unsure about it, go ahead and take at least one more transfer to a clean plate just for safe measure.  You can take a smaller wedge to grain, but it will take ages to colonize, which can cause problems on its own.  Like I said, I like to take 1/3 to 1/2 of a plate or so.  Sometimes smaller, sometimes larger.  While I'm not a huge fan of putting a whole plate to a jar, it is certainly possible and many people here do it with great success.

okay to clear things up my plates are 100% clean and they are in standard 100mm petri dishes on the third transfer from the original ms plate
I think ill make my plates into about 6 wedges each put them in the jars and shake to break mycelium up. I like the idea of chopping it up in the jar however it seems that may be bad because then i would have to put the handle of the scalpel into the jar and the handle is not sterilized just sanitized with alcohol

I'm a believer that the more culture you add the faster your jars will colonize. No brainier..
The faster they colonize the healthier the jar will be, that's If the culture is clean.., obviously..

I have a method that works for me, using plastic table spoons to transfer the culture from the dish to the grain jar and use the plastic spoon to chop up the puck. I do the process in fifteen second with the lid back on the jar.. I do 10 jars just about ever other day in the morning while I have my first cup of coffee. I use 10 spoons to do 10 jars.. No messing around torching scalpels and cooling them down.. The spoons are seven inches long. I tilt the jar so my hand isn't over the jar. When I drop the agar puck in the jar It only two inches from the top of the jar.

It's a unorthodox method that works for me.
