How to best inoculate agar with mycelium from fully colonized rye jars?

I have some fully colonized jars and want to transfer my culture to agar petri dishes I made yesterday. What will be the best way to transfer mycelium from fully colonized grain jar to agar petri dish?

I plan to do some experiments with this: Part of dishes will get a colonized rye grain with mycelium; Others I intend to inoculate with straight fluffy mycelium on inoculation loop;

What other ways are there to do G2A transfer? Google didn't relieved my interest on this topic, so I decided to start a new thread about this important matter in mycology - I have no cloneable/spore-printable mushrooms growing yet to save my culture for years to come.

you can attempt to stab threw your port/ polyfill with a clean flame sterilized needle and get a tissue sample like that.
Or open the jar, grab A grain out with a scalpel. Usually there is enough growing on the lid and you won t have to stab the grain.

do it as you said. and possibly a direct mycelium using scalpel. some dishes may be contaminated, but you could, if they grow strong enough, do another cleaning transfer. especially if you inoculate jar by spores.

I wrap tweezers in aluminum foil -> Sterlize in the PC (usually just along side the agar) for 25 min at 15PSI -> Open tweezers in SAB and use to transfer one grain.

Tweezers are 1-and-done too. If you need more than one transfer put a handful of tweezers in a glass bottle and PC them all together. Open them all in the SAB and use one per transfer. Do not flame sterilize between uses, one-and-done only.
