I don't see why you would want to wait to case the substrate because the longer you leave them out, the drier they get, and they are more prone to bacteria. I wouldn't wait longer than a week after they are fully colonized.

Are they fully colonized now?
If so, you can case them right away. You don't need to wait, but it's always best that you do, incase the center of the cake isn't colonized with mycelium. WOuldn't it suck if you cut your cake up to find that everything in the middle is just vermiculite?
Wait a week if you like, but i'd case them now.
