Agar questions

I made an agar tek with BRF and I thought I had messed up a lot. It was really hard to measure the ingredients in grams since it was a very small amount. I also made a mess by spilling agar in my microwave and then tried to put the stuff back.
Then I used a very very small part of a fresh mushroom just below the cap and put in the petri dish after microwaving while it was still liquid.

However, it all seems to have turned out good. It seems agar work is quite forgiving.

There are couple of things I wonder.

1 - Does it seem to be ready to use?
2 - For how long can I keep it?
3 - How should I apply it to sterilized rye? Should I microwave the whole petri dish and then pour it in a container with rye? Will that not kill the mycelium?

Is it ready to use? sure, i would transfer a rice sized piece from the edge and make a more uniform plate.
You can keep it for a few month in the fridge if it is wrapped properly.Longer if it was a bit thicker.
Do not microwave your myc. If you want to transfer that to grain.In a still air box you need something sharp and stainless steel that you can flame sterilize, cut into the dish and transfer into sterilized grain.

Yes, that's what he means. Although, I can't see the clone piece in your agar dish. There doesn't seem to be a point of origin for your growth.

there are several actually. One at top right and one near my thumb for example. I used very small pieces, almost as small as dirt beneath fingernails.

I have three more questions.

1 - Should I have put it in the fridge directly after putting the mushroom in it or should I do it after colonization?

2 - Can I use a rice sized piece on sterilized rye as well? If so, Wow! I have a lot to colonize with already :laugh:!

3 - Should I be able to follow the same recipe but instead of a piece of mushroom use a spore syringe?

1. you put it in a fridge when you want to have backup culture aka storage. it grows and is kept on normal room temperatures in all other cases.

2. yes but will take ages to colonize. you will benefit from more than one inoculation point. dont put that myc on grains tho, plate does not look good.

3. yes but use PC and do it properly.

dont double post edit your last post if no one responds

1- you can put it in the fridge after it finishes colonizing
2-you can use a rice size piece of myc to colonize a grain jar but i would not recommend it. Maybe cut the dish so you can use 1/4 or 1/8
3- i dont know what recipe was used in the first place but you should be able to use the same recipe for both spores and tissue.

Why doesn't it look good? It's mostly white on top. On the bottom, it's a bit yellowish but mostly white. I PCed it properly but then I got some hair shaft in it.
Does it need to be completely white everywhere?

usually you want uniform rhizomorphic growth.Not splotchy with lots of different sectors.

I see! Thanks. That's why you suggested to make more agar from my agar. I will do that and then use that to inoculate grain. And then I will take pieces of rice sizes and inoculate grain from different ports.

to me it looks a bit bacterial, yellow on plate is not good sign and growth is all over the place.

when i clone i go at least one/two transfers from original tissue, even if clean. that way you can be sure its clean - growth is more uniform, organized and easier to spot contaminants. bonus is you have few dishes to play with.
when you get your agar down you can start playing with LI and LC... that way one plate can inoculate hundreds of jars in no time.

agar is easy but not THAT easy. :smile: you are doing good, don't stop you are almost there.

Thanks for the tips. I will try to create LC/LI too.

I'm sterilizing new agar with BRF right now to inoculate with spore syringe and realized I have another question.

Last time, after sterilization, I let the agar cool and then microwaved it later to make it liquid. When it was liquid I put the bit of mushroom in it. I'm thinking now, after microwaving, the agar is quite hot while it's still liquid. Will the heat not kill the spores in the syringe? Can I not inoculate the agar when it's cool and solid?

Oh, I read on Bod's agar BRF tek again. He microwaves it in order to be able to solve the ingredients in the water before sterilizing it. I didn't do that part now. I hope it turns out good anyway.

Okay, I opened the lid of the flask too soon and the agar blew up and I completely burned my arm. Also, now I have a gelatin mess in my kitchen. Do you have any tips on how to clean my kitchen? :smile:
