Oh...one other question.

Condensation formed inside the plate then dripped onto the agar....any problems with this?

According to the MMGG, you should stack your petri dishes (about 4 or 5 high) while the agar is cooling to keep condensation to a minimum. I would think since it's all sterile, drippage wouldn't hurt.

Sorry, I don't know how long you can save agar dishes. But since it is fertile conditions for mold and bacteria, I wouldn't chance it after too long.

Colonization should take 4 to 5 days. Did you add the spores to the center or multiple spots in the dish? Use the mycelium closest to the center (away from the sides) to avoid contamination when you take the samples to innoculation your substrate (rice cakes).

Check out your mycelium under a microscope. It's kewl!

I used the innoc. loop to swipe some spores on the plates. Just innoculated last night.

Standard agar techniques require that the dishes be stored with the agar on the top and the water drops on the bottom so as to not let the water affect anything that might be growing on the agar.

I've noticed that if one injects a small drop of spore juice onto a dish it will take about 2 days to show signs of growth and about 10-13 days for complete colonization. Spore print scrapings take slightly longer.

Thanks man. That helps! I notice some growth today. I am incubating at 70-75 degrees.

I guess all I really needed to do was to wait till the agar set then turn the plates upside down? Maybe I should be incubating that way also?

Another tip here is to put the plates back in the bag to incubate. This eliminates some of the nasties getting at your stuff over time.

What works really well for me is to stack them in stacks of 10 after pouring and when they are cooling. There will always be some condensation on the top ones. I let them incubate with the lids loose in the dish and when all condensation has disappeared i used a small roll of saran wrap (or the dutch equivalent,made of "breathing" polyethylene)to tape the edges of the dish shut. Just cut a small roll of 2 cm wide or so with a sharp knife from the big roll. Taping will keep all the contaminants out while still allowing gas exchange.
