What about streaking? In micro lab we streak to produce multiple colony points... I get that isolating a problem in one spot makes sense, but you may have a chance at a good colony amongst many on one plate.

When I went syringe to agar,I dropped a drop of spore solution on a sterile swab,and used that.

I have some agar plates that are almost 100% complete. They have over grown most of the plate except for the outside edge close to the walls of my pastyplates. You can see the thin, almost clear, leading edge of the mycelium that does go to the wall but it's not as thick as the mycelium slightly behind it. I'll try to get a picture on here shortly but does this sound like bacteria? They've grown clean up to this point so don't see how it could be bacterial but I'm still new at this and could use a more experienced eye.

I dig. I use an innoc loop on the lab bench but I’m sure the needle on an MS syringe will serve the same purpose as long as the agar slice up doesn’t happen.

Here are those pics I mentioned two comments back. Notice how the mycelium doesn't reach the edge of the agar plate. These did have a lot of moisture coming out of the PC, not sure if that would cause this issue. Are these plates good for A2G?

It's actually better to go to agar first period... even with brf really; just has to be approached differently,  and requires a pressure cooker (PC)

Hard to tell anything about those plates from these photos... though the 3rd makes me think bacteria with the growth not betting uniform... condensation will spread bacteria, and judging that you said it's the edge, probably the case... I'd transfer from halfway between the canter and the edge, probably just get rid of the 3rd one... wanna take transfers from suspect trays as quick as you can identify it's mycelium to avoid meshing with contaminants

So I made three T2 pastys last night. Looks like I had too much condensation. How troubled does this look?

Just try not to cause it to run a lot, if the plate is clean it's not a big deal anyway

I'm hoping they are. I'm gonna order a 500pk of petris with quick delivery. Im tired of mold so I'm going to take like 3 T3s ea and then 5 T4 ea from those before I use em all to noc master pints. Then I'm gonna start another variety.

It says estimated shipping is the 22nd. If I place my order Sunday night I can get em on Amazon this week. That puts me one set of transfers ahead. If I maintain that pace I might order those as well hah

Transferring out to new plates today so I can get this to grain soon

I need some monkey love APE action in my life!


I've got 2 APE tubs that are overdue to pin. Not sure what they are waiting on.. Ugh! :rolleyes:
I do have a thin layer of coir for casing. The casing is uncolonized in one tub and about 50% in the other.
I'll get some pics up later but there isn't much to see really.

You got some really nice rhizo growth in those red plates...try to transfer a small slice of one of those strands. :thumbup:
