I'm growing on agar from MS syringe just to make sure there's no contams before transferring to 4 quart bulk jars, would it be better to transfer 4 small wedges from the same petri that is growing the fastest/cleanest or mix it up with multiple dishes or does it not matter?

If the dishes are all from the same syringe it doesnβt matter I donβt think, but I donβt like the idea of spawning germs plates.
Aside from not being able to properly spot contams on a germ plate, thereβs also always weak myc that I donβt want anything to do with.
Uniformity. Consistency.

I took a transfer from a clone plate of a plate pin. I dont care for my "pastyplates " and I'm gonna order a case of 500 petris in the next week. I'm gonna get another transfer deep then start expanding with multiple transfers till I can tic tac toe a few dozen pints.

I was having condensation issues too and it definitely was temperature swings for me. I have wood heat stove in the house so temperature fluctuation is bad. I cranked up an electric heater in the room so it is more consistent. Hoping that cures it, helping so far.

Yeah I should have taken a clone of a good fruit from my last grow but only just getting into agar so I'll do that next time. I figure it's got to be at least a little safer for contams than shooting the solution into the jars.

Gonna play a little; going to attempt to transfer these 2 entire RW cultures away from the bacterial colonies near the edges... they're clean otherwise
And taking a transfer from this PE6 clone slanted in July....

thinkin about trying grain soak agar for the first time. I'm not seeing any specifics out there, so I'm guessing nutritional potency of the water doesn't really matter? Just boil some grain in water and put agar in it, no fuss?

I've seen grain soak diluted up to 1:4... I'd start with 50/50 and see what type of growth you get
I'd take 3 small transfers from 4-7... a good bit ahead of the rest of the culture, and looks like it's going to go rhizomorphic... nice pick, and nice pic!

Yeah I always make my GW from at least 6-8 quarts of grain, no straining or anything.
I freeze what I donβt use (mix first to homogenize sediment) in ~300mL aliquots.
I then mix 200-250 mL of the grain water with my agar and fill to 500 mL for GWA.
I donβt like it full strength, but that may be due to how concentrated mine is to begin with

I am going to try it when I actually have a clone I like. What would you use for malt extract? Iβm thinking about 15g per 1000ml.

I use 6.5g LME (light malt extract) to 500ml of water... so 13g per liter... but as much as 20g is standard MEA... just play around with different nute levels with different cultures: see what gives you the type of growth you like

That's how I do it. I capture the grain water when I strain grains and then add agar powder to the grain water (10g agar/500ml grain water) per how much agar I plan to make. I also use the grain water to make LC as well. Rye seems to work best IMO.

Still experiementing with this myself. I'm thinking Rye would get better results than oats because of the hull. Here's from my most recent pour:

You can see the grainy bits floating around. My first plates were crystal clear... got down a little further in the jar and here it is.
Also, I'm starting to think that 10g agar is too much for low temp pours. I noticed that I was pouring solid bubbles trapped in the agar (you can see in these pics too) and I poured at 120f. I'm going to drop my agar down to 7g or less.
I bought some LME and I'm going to make that next time I pour agar. Just to see what I like. Here's my next recipe for 500g I'm going to work with:
Sound like a good test to folks?
