hello, question about grafting seedlings,

i grafted a trichocereus seedling,
and a lophophora seedling,

I used the saran wrap  but i didn't have a clothes pin so i used those little green twisty things from bread bags,


i then put them in my humidity chamber,
aka
a plastic rubermaid tub,


they are 2 days in.


and the loph on pereskiopsis has developed a sort of rust around the edge,

any idea what this is,

and how long i have to wait to see if they take?

The orange is not a good sign, there's a pretty high likelihood that one won't survive. Increasing air circulation might help its chances somewhat.

Next time make sure your hands, knife etc are as clean as possible. I wash my hands with soap followed by an alcohol rinse when starting and repeat the alcohol rinse every so often, and the blades get a 99% isopropyl alcohol dip followed by a flame (then wait for it to cool obviously) between each graft. The seedlings I also give a peroxide soak beforehand, followed by letting them dry off completely. Do not let alcohol or peroxide contact cut plant surfaces.

The saran wrap is another possible contamination vector, and IMO it's not necessary for seedling grafts. May as well give each piece a dip in something too if you do use it. You shouldn't need the humidity chamber if it's covered in saran wrap.

To check if the other one's on there good, wait a week or two and you can sort of lightly poke it to see if it's secure. A failed graft may be noticeable sooner than that if there's a visible gap between the two surfaces. You can regraft it if it didn't stick.

Regarding watering, personally I like to have the seedlings well watered and sappy and the stock fairly dry. I picture moisture getting sucked out of the seedling by the thirsty stock plant stock soon afterwads, with that initial flowthrough causing the union to seal. Watering too soone before or after grafting can make the scion slide off or be pushed upwards by gel-like sap.

how big is too big of a seedling to graft?

I've heard that mixes with some water work better than 99% too, but on the blade I'm using it more as fuel for flame sterilization. For my hands I use 70%.

Cacti of all sizes can be grafted, but generally for flat grafts to pereskiopsis it's best to match diameters pretty closely. For a scion wider than the pereskiopsis you have, you can use a wedge or impale style graft on pereskiopsis, or do a flat graft to a wider stock like trichocereus.
