Birdseed and Air Exchange

Is air exchange a neccessity?

I have 4 pint jars of birdseed innoculated with agar wedges. Since I used agar wedges with new jars, I didn't bother to poke any holes in the lids. After about 4 days, I started seeing good growth on the jar sides, so I shook them up a little. Its now been another week, and I am not seeing any more signs of mycellium. I thought that maybe the fluctuation in temp during the day/night might be the problem, so jars have been sitting in an 86 F water bath for the last 48 hrs, still no signs of continued growth. Should I just let them keep going?

4 days to see growth from agar? Sounds more like a multispore innoculation. I get 100% colonization in rye pints in 12 days with 3 shakes and no air exchange. Polyfill filter floss is good -- go read the thing from anno about polyfill it can be found at:

Thanks, guess I wont worry.

I noticed the mycellium is not exactly aggressive on the agar either (versus another strand I plated earlier this week). I poked the wedges into the middle of the jar, and sort of buried it in the seed. It took 4 days for the growth to radiate out from the center to the sides. If you don't worry about the air exchange, then I am just going to let them sit. One other thing I forgot to mention in the original post, the jars don't have much in the way of headspace either (about 4/5 -100% full).

hopefully one day these things will give me something to post in the pictures forum.

I once inoculated 6 birdseed jars and had the polyfil for air exchange. 3 I left covered with tinfoil over the lid *no air exchange* and 3 I left uncovered. The three that were covered took 10 times longer, I took the foil off and in a week later they were done.
