PURE WHITE LIQUID CULTURE PANCAKES

Have many bulk liquid cultures going.

I forgot to stir a few for awhile (week - 10 days?) :confused: on the stir plate.

Ones I didn't stir, grew about an 1/8 or 3/16ths thick pure white myc pancake floating on the top.

Have on stir plate now, trying to break it up & get everything back into fluid suspension.

LOL...... myc pancake is pretty tough - to get apart :eek:.

Stir bar goes in the jar - pre/PC.

if you get one, PM me & I will show you how to build stir bars, for almost nothing. Store bought, they run about $9 + each. Which can get spendy, running a dozen or 2 culture jars - at once.

Go to auto parts store. Buy several feet rubber wind sheild wiper type vaccum hose. Also buy RTV high temp gasket sealer.

Find steel nail, bolts or threaded rod(magnetic) that will fit tight in rubber hose (Ace or True Value). Cut hose into into short lengths, cut of heads of bolts/nails - or cut rod to size, insert in hose - seal both ends with RTV sealant. Allow to cure overnight.

Good idea on the stir bar.

I had a myc pancake form in a culture I think. I didn't stir for 3-4 days though. I didn't think the myc would form a solid that fast so I tossed it thinking it may be a contam. It was pure white and looked just like your pics but only took up 25% of the surface.

Surface myc starts small & grows. If it was pure white. Sounds like you tossed a good jar.

question on those stir bars, can that windshield tubing withstand autoclave temperatures. Say for making those nifty lids you have?

agar- do you believe harvesting the mycelium is worthwhile? I have been toying with the idea of a 50l container. I already have it all built for another purpose and it could be easily modified for a bulk LC.
An idea that I tried before was readding sugars to a spent LC, theory being that after it uses up the 4% solution it is thick with mycelium, you bring it to 4% again and it will multiply again very quickly ending up in a densely populated mycelium LC. It seemed to work for a while, I never drank it though. After a while its metabolites should become toxic to itself.

Most plastic is fine in a a/c. You could always just cover the nail completely with silicon

Years ago, I did a 5 gallon batch that was stirred & provided with filtered air exchange. The results were not worth the time, effort or trouble. Moreover, LC's are very prone to contamination. In the sense that a liquid nutriant broth is an ideal medium for them.

The big LC culture was incubated in darkness. I have always wondered - if once the culture was fairly thick - if it was exposed to steady light & incubated/stirred for more time (a week)- if that might increase potency?

All in all.... I believe fruiting out an optimal substrate under ideal conditions will get you about the best potency - possible.

Speakin of potency increases in LC's. I've been fiddling around with vanilla in LC's. Vanillas loaded with amino acids and anti's. I used some extract in one to see if it would be worth while. Just to get a colonization and MAYBE a potency increase when knockin up grains. You know all that jazz about Tryptophan increasing potency? And to my surprise the damn LC is taking off like a rocket. Haven't put it to use yet though.
