Substrate for an InVitro grow?
    #3900982 - 03/11/05 12:55 AM (19 years, 1 month ago)

I'm wanting to do an invitro project, as i don't quite have the space
for a complex (read: obsessively sterile) growing apparatus, but I'm
feeling a bit adventurous as far as substrates are concerned. Would
rye or wild bird seed be appropriate for fruiting invitro? Is there
another substrate that I'm overlooking? or should i just stick with BRF?

Whole grains require a PC for sterilization and are not really suited for invitro cultivation. The PF tek works great for invitro.

Cool. Hopefully this will turn out better than my first attempt.

Anyways, off to buy jars, fish heater, and cooler from Wal-Mart at 2 AM.
NOBODY WILL SUSPECT A THING HAHA!

I'll try this with pint jars and liquid culture to satisfy my sense of adventure...

Al right. I've got a styrofoam cooler, a heat-bomb style heater, and 24 1/2 pint wide-mouth jars.

I think i scared the check-out lady at Wal-Mart though.

In AFOAF's experience, PF tek cakes shrink slightly when colonized, making in vitro an option. Grains dont usually shrink mach at all, until after the first flush.

Yeah but your supposed to leave room. I've never had any problem with grain invitro.

You'd be amazed at how well straight rye can perform invitro. Just hydrate the grains with coffee and expose to light from the day you inoculate. This is all straight rye, no vermiculite or compost, hydrated with 50% of drinking strength coffee.

Even with contams, youd be surprised.

Hmmmmm.... Would coffee be an option with the PF tek? Is there a specific way to use the coffee?

Do I even need the coffee if i'm going to be innoculating with liquid culture?

Those jars are beautiful. I'm going to build smaller and slightly
less-tasty versions of them.

Off to the hippie-mart to get some rye grain!

Whole rye grain works great for invitro. I use 1/2 gallon jars with 2-3 inches of cooked(and pc'd) grain on the bottom (only additive to grain is a pinch of gypsum). After full colonization I case it in the jar with 2-3 inches of a coco coir based 50/50+ mix. This picture is of the latest 1/2 gallon jar. well over 50 strong looking pins.
