Funnily enough that post by Fahtster was my jumping off point. I was thinking about DMing Fahtster, but I was kind of assuming I was just searching wrong.

I've made few transfers to the water agar 3 days ago for the first time. After the fact, I've read somewhere that you should use larger wedges than usual, mine were like rice grain size. So far no visible growth at all.
Is this a sign it's dead? Can I just place larger wedges on the same plates?

I like to use a syringe needle and take a little needle nose pliers and slightly bend the tip so that it's Barbed .. this becomes a perfect scrape and grab tool for Ariel mycelium.

I want to try some cakes of just rye flour. I figure that should definitely work, just like fruiting off of grains.

But I'm wondering how much I should hydrate it. PF Tek suggests 2 parts Verm 1 Part BRF 1 Part Water, but that's specific to the characteristics of verm. I'm not sure if 1 part water 3 part rye flour would work (equivalent amount of dry matter).

I don't have any vermicultie, but I do have some hay, although that wouldn't have the same characteristics as straw used in invitro straw teks.

This is a very helpful write up thank you!

From the totality I think that I need to add food coloring to my agar for sure. I had been skeptical because of my early attempts with vendor produced petris that were blue and seeing the plain MEA petris of the same cultures outperforming the color cultures.

I have tried dental picks, and still regularly use them for other tasks, but have had difficulty with avoiding touching something else when trying to pull the hyphae.

I have had success using a syringe to extract a samples and then depressing the plunger to transfer. The issue with this method is about 50% of the time sending the sample across the workspace and no where near let alone in the petri.

Were the plain MEAs also from the same vendor and at the same nutrient level? Either could acct for the difference.

Some food coloring (at least one of the reds IIRC) can be vaguely fungicidal and I've had very pigmented plates underperform, but dv8 is def the big dawg on this front.

One aerial cleanup technique I didn't see in there that I'll sometimes use. IDK where I learned this, in my mind it's called "the tickle" and I swear I got it from somebody on here but IDK:

Sometimes you want an aerial transfer from something so dirty that you don't want any ground/plate surface to come with you. Rather than trying to cut /twist the myc, get a thin surface from the recipient plate stuck to the side of the scalpel, lightly brush/dab it over the aerial mic on the dirty plate, and then drop it back to the new plate. Like the wedge equivalent of a spore swab.

Alternatively (this is harder in a SAB), but if you can get both hands free and the aerial myc is long, it can be easier to grab the tip with tweezers and then scalpel across it lower down to take some without bringing the base up. Fresh blade pretty critical here otherwise it just pushes it around.

Just flour is likely not going to enough, it needs something to serve as a reservoir for water, plus it will probably contaminate easily due to being a humid paste of nutrients.

You can totally use coir if you have some instead of verm btw. I never buy vermiculite anymore.

Hay, I am not sure. I'd try an adapted Muda Recipe for Straw from the Updated Bottle Tek thread. More chance of success than just rye flour.

Yes the petris were from the same vendor and the colored ones were sold as "rapid rhizo". The vendor claims to pour on the same day the order is shipped and both types of plates were in the same order. It very well could have been mistakes on my part, I'm learning how little I know every day, and loving it!

I have been thinking of using a small pair of precision shears to cut the base off when it sticks, but a scalpel is probably the right call instead of adding another tool to my workspace.

I did recently move to an FFU, so along with reworking all of my workflows I also get to learn about vibration isolation!

Is this ok?

Flipped lids with foil
PC 15psi for 90min
Left for 8hrs to cool overnight with PC sealed.
I opened the vessel in the morning to see if the jars were cool enough to tighten the flipped lids.. but..

They were still warm so I put the pressure cooker lid back on and went to work.

I'm worried about dirty air getting into the jars as they cool.

The risk should be negligible right? With flipped lids, foil on top of jars tight, lid rings loose.. even tho a small amount of dirty air was let into the vessel when I checked the jar temps, hopefully I didn't do any damage.

When I get home after work if should all be cool and I can tighten my lids and inoculate tomorrow night.

Should be totally cool. But if you're worried about it and want to be super careful give it five days or so before inoculating to see if it decides to turn on ya.

Opinion on fruiting AIO jars: top-fruit in the jar or โ€˜birthโ€™ it? If I birth it do I soak it before casing?

What do you mean by "AIO jars"? I'm confused because AIO jars usually are not soaked, but BRF cakes are. And I only read the term to "birth" used to refer to BRF cakes. If it's a BRF cake you're talking about, then yes, birth it and soak it, and proceed with the standard PF TEK protocol.

If you're confident enough and want to experiment a little with dialing in surface conditions on mini monos or other "bulk" approaches, you could break your cake and mix it with hydrated coir. But in the link above you can find complete informations on that (section 7.2: Top Fruiting & Bulk in a Mono).
If that's actually an AIO jar, and it's fully colonized (be sure that's the case, because uncolonized parts can kickstart unwanted contaminating organisms), then I would prefer to break it and put it in a container with a shape that allowed for more surface, mantaining around 4-5 cm of depth. That's because in my experience it's much more efficent. If it's an AIO, it should be already well hydrated, but if it needs more, just mist it after spawning.

I wouldn't reuse the plates, but if you are low on them, I feel you and understand why you're thinking about it.
In case it's a "desperate" situation, reuse them, but transfer as soon as possible and keep everything as far away as possible from the other transfers.

Regarding the size of transfer, it can work out even with super tiny pieces of tissues, that's why things like Josex's poke work. But in my experience bigger wedge transfers catch up better, and when doing super tiny aerial myc transfers, sometimes there are no signs of lifeat all.
Do you have pictures of the plate you transfered from?

Someone in another post wrote about the pasteur's swan neck experiment which shows lab glasses in a shape that probably creates a condition analog to the jar rim covered by the lid (+ the foil in your case) which should create the conditions for it to be totally safe!

Working in front of hood.

6 jars. Same culture.  Lc.

Jars lined up next to each.

Nocced up one after the other.

5 jars fully colonize. Clean.

Last jar.

Black pin mold.

What happened. Had to be me right?

I wonder where I went wrong?
