Selective breeding for increased potency
    #29681509 -

Is there any good way of doing this as a layperson? I've heard of test kits and stuff to get an idea of psilocybin content. I'm not afraid to do a little math or journaling and documenting and reading up on Gregor Mendel's story of how he selected pea plants just right for his preferred traits.
I got my one strain of golden teacher. I want to nurture it into one that is strong and potent. A golden teacher that is surprisingly potent for a golden teacher :poison:

You need a metric fuck tonne of Q-Tests.

Do a grow. Select a bunch of random shrooms. Make a clone from each. Dry the fruits 100%, test each one. If one has higher potency than others by a significant margin, run that clone. Repeat the process again, except this time do spore prints.

Repeat all that again and again and again and again until your samples are higher than baseline without much potency difference between the samples.

Grow out their spores in 100s if not 1000s of tubs and repeat the process to see if what you're seeing is stable.

If instability shows up, go back to the last stable clone/print and start over.

Honestly, it's way too much work when you can just grow exotics instead.

Are you saying this because exotics are just naturally more potent?

Many are, yes.

For example, GTs are an average 0.5-0.9% of actives, where TTBVI is around 3.2%.

How many years and how much money on qtests/growing supplies is it worth to spend on a mediocre cube variety when some basic growing skills gets you something thats 3-6x as potent immediately.

Even an average PE variety blows GTs out of the water potency-wise

The best way to intentionally do this would be to start with wild genetics. You want maximum heterozygosity in the expression so you can at least find what you're looking for. Once you do (and it might take a while) you can look forward to the next filial generation. Do that for like 7 generations and it should be pretty stable.

Most of the older varieties were mostly stabilized for their spore production not potency  sure there are outliers but they are hard to find.

Ouch. Yea its not that I wouldn't have fun with that. But to grow that many tubs would be scary. Wouldn't be able to hide it from guests. And then the quantity there sounds like WAY above what the law would consider "just for personal use". I wanna selective breed. But I also don't need more shrooms laying around than what satisfies me.
Damn..
I need to work for a dispensary in colorado or something so I can have that kinda fun.

When I was hunting phenos while searching for RW in the second Gen I just did smaller and smaller grows. Eventually found my master line pinning in a no pour plate. I don’t recommend running a big tub of anything you hadn’t already tested.

so just make tons of small pf tek jars and examine for highest potency? Then make a tub of a potent generation?
I think for fun I wanna select for other traits too like # of flushes, sizes, deeper color. Deeper bruising. I want to make a designer golden teacher. Tame it to be what I want.
Doubt I'll get something as wild as x7x but hey. I wanna have fun with this if I can get the opportunity.

Running small substrates for pheno hunting is a great idea.

Was kinda the inspiration/drive behind my invitro straw method where you only use a little and then fruit in the container, just untwist the lids for FAE. I’m not even sure if you can buy those ziplock containers anymore. All kinds of little grows that tossed out disappointment

It was actually one of the cheapest ways to grow. Each container contained mere pennies worth of media.

Great method for sure.

I just found a 5 gallon bucket full of chopped straw I forgot about and instantly thought of the invitro straw tek.

Just in time to start a few bottles to get me some pan clones.

Agree with what Pasty mentions about heterozygosity. If you start with a stable variety like GT, you’re not going to achieve anything while spending a lot of resources. If you want to select for potency, you could also start by crossing two varieties that are known to have high potency. That would mean less heterozygosity, but a higher potency to start with.

If I wanted to start breeding for fun or learning or to release the next stellar variety into the community, I would probably start selecting for visual traits instead. Much cheaper, and more direct feedback which would make it more fun for myself. If it’s just to have potent mushrooms, grow subtrops or pans.

Since doing cake wars, I've been thinking much more about this in general.

Consider the classic recommended method of growing nowadays

1- Spores to agar
2- Transfer best first clean growth
3- Keep transferring, 2 or 3 more times most likely
4- Send to grains
5- colonized grains to coir

This will likely provide a nice stable flush with some variety, but the reality is steps 2 and 3 remove the vast majority of the gene pool available. Probably over 90% if higher? STill, for someone who wants quantity, that is the best way to start off.

This is a massive bottleneck that selects for growth on agar, which is good, but isn't necessarily the best potent fruits.

Let's look at a classic PF Tek method, which is the safest for MS syringes.

1- Inject cakes with MS syringe
2- They grow, hopefully clean
3- The entire genetic potential is within that cake, and there are thousands if not millions of spores that germinate and add themselves to the wholes.
4- Yields are not as good and standardized, but you have more variability. What fruited was the winners in a big ass competition.
5- Select for the most interesting phenos, which ones bruise the most, which ones have solid stems. Bruising doesn't guarantee potency, its actually a sign that it lost some to oxidation, but it at least means it did initially contain a lot.
6- Usings prints from step 5 for crosses, and cloning the best fruits (test them yourself before dedicating yourself to the cloning to not waste time with something that looked good but wasn't what you hoped for potency wise.

Seems better to get clone samples from PF Tek by a long shot, otherwise you are hoping that the bottleneck you made on agar happens to have the best genes for potency selection, which considering the bottleneck, isn't likely.

On my plate, KSSS, Great White Monster (AA+), Australian Murbah F2, Columbian Rust Spore, and some attempted crosses of those by mixing the spore solution. Will likely output both parent strains, but a few of them should have notable crosses.

If you want coincidental spore crossing you need them to germ at the same time. I feel the best way to cross is to use fresh spores. When I did RW I swabbed both parent fruits gills at the same time on the same swab. This ensure lots of crossing.

That, or at least swabbing both prints simultaneously? Better when fresh I suppose.
