Can't figure out where I'm going wrong with rye

I recently did a couple rye grain spawn jars for ATL #7 (just a little side project since it's set it and forget it if you aren't going to bulk). I thought I had hydrated it perfectly. I did an 8-12 hour soak, then I simmered it, stirring constantly for about 25 minutes. I kept checking the grains throughout the simmer, and pulled it off when the majority of the rye berries had no white, starchy center anymore and become fully translucent. I could see the split down the center but not sure if they were truly "butterflied." I then dumped them on a towel and let some of the excess water steam off before jarring it. I thought my hydration level was going to be PERFECT, but neither jar I inoculated started at all. It has been 3 days and I see no growth whatsoever. Is ATL #7 really slow to start, or did I somehow not hydrate the grains well enough? I'm completely confused, I really thought I had nailed these jars. There was no external moisture but they were well hydrated internally. Starting to think maybe it was contam, because I had used large tub filter discs on the jars for more air exchange that I had seen another very successful truffle grower do, but because they took up the entire lid, I couldn't install injection ports and had to inoculate by cracking the lid in a SAB. Would really appreciate anyone telling me which rye tek's they've had success with.

I hope you don't mind filling in some blanks.

You covered soaking and simmering.  What happened after that?  Pressure cook or steam pasteurize?  And for how long?

How did you inoculate?  Agar, LC, or MSS?

Sorry, idk why I just blew those off like you guys knew what I was doing. I've been a bit frazzled the last couple days. Anyway, after the soak and simmer I PC'd for 90 minutes, let go back to room temp and inoculated with 4-5cc of ATL #7 LC on each jar. Like I said, I did not have an injection port on these jars because I used a large (about 2.5inch) filter disc that took up almost the entire lid so I had to crack the lid and inoculate in an SAB.

I had a tough time with rye once as well.  I can't definitively say this is what you have happening.  Maybe it is, maybe not?  I'm just sharing my notes. 

Getting better with grains:
I went back to ye olde 20-24 hour grain soaks.  People are going to tell me I don't need to go that long and to focus on what the grains look like after the simmer.  Things fail on me when I do shortcuts and work when I go old school. :confused:  I gotta stick with what works for me. 

It also came to my attention that I was contributing to drying my grains out by letting my steam regulator purge every 20-30 seconds (as instructed by the pressure canner manufacturer).  But doing that is continuously releasing necessary hydration from the grains.  It makes sense NOW, duh.  I keep my stove temp a little lower all while maintaining the appropriate PSI, but don't run it hot enough to be blowing steam quite so often.

Not accepting LC's as is:
I've had too many failed grows by blindly trusting LC - even from known, vetted, experienced sources.  Contams get in those syringes either in the source or from the (mis)handling that happens in transit or storage. I still buy LC's, but I insist on growing on agar to inoculate the grains.  This allows me to reduce the contams and ensure the myc is healthy to do its thing.    I can't say your LC's are not viable, but it is a common issue.

I don't know enough about tampanensis to help with expected timeline or growth characteristics.  Maybe your culture is just a little shy?  Someone else might know something and shed light.

This is how to prep rye, don't soak it for any length of time prior to cooking it.



Sterilize for 2 hours after at least a 10 minute purge. Do everything I say in this video and your grains will come out perfectly. If you're using LC before you're capable of running properly sterilized grains you need to switch back to a proven clean culture. That's where I'd start

I use rye all the time. Your grain prep technique is lacking. Watch that video Stipe just posted, or read the hitchikers guide to the shroomery.

I'm still not exactly sure how it was lacking. I did the exact same process in terms of rinsing and checking the grains while hydrating to make sure the interior was translucent and there was no white starchy core left on the majority. This actually was one of the videos I watched before I tried the first time. I think it just came down to still not hydrating them quite enough. Mine definitely didn't get that puffy.

I use rye or wheat, but dont soak, I simmer in pot on the stove for about an hour until they are chewy. Then let them air dry for at least a few hours.
  After it goes into jars I pressure cook for 2 1/2 hours at 15 psi. I think thats your issue, not pressure cooking long enough. I haven't had any issues with the last 40 jars this way.
    The only 2 jars I had trich were from injecting straight from a syringe into jars...

Soaking isn't beneficial, it's an unnecessary step which may actually cause problems, theoretically.
