Re: Top Tea Prep method test per potency testing of dregs, teas, & extracts [Re: ihansterx4i]
    #29606215 -

I went down the rabbit hole of reading almost all of this entire thread (skipped some of the sidetracking on the last couple pages), but found most of my answer on page 17ish.

I really liked the idea of storing dehydrated tea for various reasons, mostly for saving space but also for ease of use. Yes it's not too hard to roughly grind for a tea, but the procrastinator in my wants to knock out the hard work one time and then cruise on easy mode and take up less storage since this hobby seems to keep wanting more and more.

Excited to see the day 300 update, and as a really out there elevated shower thought as someone who doesn't care about getting the highest possible yield on a personal level, I'm putting this out there for fun:

Has anyone tried using a series of gravity flow columns packed with mushroom material assembled in a small rack so they drip into a shared bowl or something? You can continue to apply water to the top so it flows through the mushroom matrix and extracts goodies. Maybe something like a burette where you can adjust the flow would be preferably in case nothing moves through or if water moves too fast...

That pump along with some good lab glass would be great for vacuum extraction. I am liking my half gallon jars with standard wide mouth top and an electric mason jar vacuum sealer for now. But that might be the way to scale up.

So i made some active tea out of 200g fresh. I kinda screwed up the drying process because i put them on some agar plates. It ended up being impossible to scrape off so i rehydrated with some hot water and then transferred it to a silicone mold. I was able to break that up but it was still a little sticky so I dried it again and that second dry got it fully dried. I then pulsed it in a coffee grinder to get it powdered. Anyways, I ended up with 5.7g of the final product. Hopefully all the drying and rehydrating didn't kill the potency but should I expect 4x strength? Was able to get about .8g of the dried active tea in each capsule. That one capsule would be like a standard dose or am i doing the math wrong? if so , that's amazing. One capsule for one dose would be really cool.

I had a thought the other day when reading the label of baking powder; it has corn starch in it, which i assume helps the texture, or granular nature, of the powder.

It makes me wonder if there's an additive that could help the consistency of dehydrated tea. Of course, it could backfire and lead to inconsistent dosing from not mixing thoroughly/adequately across the batch.

Just a thought :sunny:

Test the potency of dregs of teas prepared in various ways to determine if one method of preparation is more effective at extracting the actives from the mushroom materials into the tea.

Thanks for your thoughtful experiments! Super interesting. Friendly suggestion to update the original post with either the results or a link to the results. I can't find them in the pages of replies.

I cut one of the trays to fit larger caps (~2in clearence). Mold is 1.96in and Fits perfect, holds about 1L tea. Super easy to scrape!

Something that occurred to me a while back, just wanted to chime in...

In laboratory conditions extractions are done with agitation. Putting organic matter into aqueous solutions and bringing it to the boil results in numerous microlayers, physical, chemical and thermal that strongly influence heat transfer, phase separation and the movement of solutes. This is why vortex and ultrasonic agitation is used for these types of extractions in professional environments.

It appears this is why we saw unexpectedly low results for simmered extractions in the initial data sets, and if we applied agitation to simmering it may present different results.

I actually tried one of my active tea capsules (.8g) today and it was interesting. First 2 hours or so I felt the sort of anxious come up but nothing happened. And then after 30 minutes of playing basketball with my son it started slowly kicking in. I  don't know if the physical activity making my blood flow move was jump starting it or not but if definitely was noticeable. I don't think It was a 4x potency like I originally thought but of course every grow is different and this was not a grow I tested in other ways before doing the active tea method. All in all if I took 2 capsules and not 1, i'm almost positive I would've blasted off into space which is cool because if its was just dried ground shrooms into capsules i would have to take like 10-15 capsules. Definitely more work doing the active tea but if you have the time why not.

I came across an extraction research article from January 2026, they claim to have a better yield than other methods.
Figured I link it in this thread in case it's useful:

Interesting because ethyl acetate is used to extract pure mescaline from cactus. Kinda wondering now if it can do the same with mushrooms…

very strong claim. why do you think this study has poor methodology? can you explain?

reverend myc. the comment i'm quoting is like 1 year ago lol, but i'm very interested in his reasoning. i've seem some other mods here in the forum saying this study has flaws in the methodology, but never entering in details on that.

There are a couple of weird red flags in the paper.

First is the following figure. This table is pure extrapolation and they didn't even control for the Y axis going to 110% ethanol, why wouldn't you catch this and fix it? It's a model based on extrapolation that appears to show all of these different concentrations in different conditions - but what was ACTUALLY tested are those 5 little blue circles.

Which begs the question, why didn't they actually test something that resembles a range of pH, temperature, and ethanol percentage so we can actually get a good idea of the relationship and build a genuine model? I'm not even saying they need to test every possible combination, but at least test pH of 2, 6, and 10 under all of the same conditions. Regardless, it's no where near enough to genuinely convince anyone that the figure is meaningful at any point apart from the 5 circles.

The biggest weird flag to me is the e-grams of their chromatography software. I'm a biologist so I don't use this stuff extensively, but I dabble in Chromeleon/Empower. Why on earth are the traces so poorly lined up with the label (basic crap you should fix), why can't you see ANY peak in 3 of the traces that still had 5-6 mg of psilocybin. Did they just not autoscale or zoom, is the baseline incredibly high, is there no normalization or correction being done to account for baseline?

Why is there a raising baseline in ALL their samples versus the control compounds. Did they really not put them in the same solution to give us a good idea of what the controls should look like under the same conditions? :crazy2:

I can't say for certain, but to me it looks like covering for bad data by zooming out and only seeing the peaks some-what recognizably in the highest concentration samples.

The one that rubbed me strangely initially, is randomly claiming they identified Tetronomycin for the first time in fungi. They characterized an antibiotic produced from bacteria within cubes. How many people have fully sequenced cubes at this point but haven't seen this produced once, and what would the most OBVIOUS explanation for this be? There's bacteria in their sample, or in the soil they didn't rub off from the sample.

Instead of thinking this through at all, checking BLAST or any other protein/DNA database to even explore this possibility they just claim they identified it in cubes. Show us a single orthogonal method that confirms this please...

Finally, I will say this may be a misunderstanding from me. I'm still struggling to understand this as well:
They started with 200mg of liquid nitrogen frozen and homogenized mushrooms. This 200 mg had 7ml of solvent run through it 6 times, and that solvent was dried in this 35C oven for 12 hours.

How on earth did that single sample from 200mg of mushroom generate 50mg of psilocybin in the form of those 6 separate extracts? Did this number come from some weird formula I missed or some other deceptive model they decided to use?

It would be nice if they included the actual mass of psilocybin recovered from that 200 mg starting sample so we can actually see the raw yield instead of the number they gave us.

i'm not talking about that paper. i'm referring to this one:

"Stability of psilocybin and its four analogs in the biomass of the psychotropic mushroom Psilocybe cubensis"

reverendmyc and a few other people have argued that this stability paper has poor methodology. personally, though, i find it to be a well-designed and highly relevant study that provides a lot of useful data.

As I have previously requested, I would prefer to keep this thread on the topic of testing extraction methods rather than discissions of the quality of other peoples work. I do not wish to muddy this thread up.

If you would like to discuss the merits of studies that you did not participate in, please feel free to create a thread of your own. It could be very enlightening to have a good faith argument there. However, there is a distinct pattern of long circuitous discussions around these topics that do not get resolved and only serve to derail threads.

This thread is about testing methods of extraction at home and the merits of those methods. Discussing the methodology of another study does not further the objectives of this thread so I will not participate in that discussion here. Please do not derail this thread.

If you do create a thread for that discussion, feel free to link it here. I look forward to following the discussion there.

I agree, that paper has some serious issues in it although I doubt most bona fide mycologists would be able to pick up on them. Peer review can be easily manipulated and manufactured, Paul Stamets made millions on his ability to manipulate peer review to sell his shitty rice products. Any paper on potency that doesn’t begin by outlining how they obtained true isolates used to obtain the fruits is already sus IMO. The number of these I see where they use ms and kits is laughable. IDGAF if it’s Penn state doing the work, the actual results need more than an appeal to authority to carry weight in this community.

Let’s not debate this paper in this thread please, if some people feel it’s an authority or good science make a new thread somewhere else, don’t hijack this one.

Questions:

1- Is there a reason why alcohol extractions weren't very much explored? Or did I miss it in this thread?

2- Let's say I wanted to transform some unnapetizing looking grows (blobs and carpets) into gummies: the optimal method would be brewing a tea, 1g to 20ml water, make a batch of tea, and then reduce it into instant tea, which can de dissolved into gummy recipes?

Sorry if covered, it's a big thread and the numbers make my head hurt
