@Huskies thank you! And I agree, calling them by the locality they were found in makes the most sense and it gives a certain respect and kinda an ode to the area they came from so I most definitely agree on keeping it local. County names for them is also a great idea. I'm down for that

The experience on these was actually quite nice. Smooth for the most part with what felt like a little drunk buzz affect in the beginning. Everyone I gave these out to liked them :] one friend however did get a little bit of an upset stomach but that was about it. And Cubes tend to do that sometimes but overall nice.

And I personally have not done a cross with a wild and commercial strain but I'd love to

I agree with what @cjbinks said. Sending out some tissue would be the best bet. A buddy of mine does analytical work in Oregon and so sending stuff to him would be a good idea with sequencing. He has a site called https://www.tryplabs.com <:D

He's a wonderful person and a badass analytical chemist and mycologist

I would like to get mine sequenced, but have no idea where to even start with that. Can I just mail some spores somewhere or does it need to be fresh tissue? Ive done some reading on it and I'm still lost.

The following partial quote from a fascinating thread on a wild find by 3.A.M makes me think tissue. I am not certain, though.

Alan Rockefeller said:
Definitely save the collection, well dried at low temperature and labeled with the iNaturalist observation number.  If you want to send me a little piece of a cap I can have it DNA barcoded.  You should carefully save the rest of the collection in case it turns out to be a new species.

Not sure if the same invitation/openness would apply to a wild cube, but it could be worthwhile to ask him! He recently shared the article from this thread's OP here, which makes me think he may find both Tico and this thread interesting :takingnotes:

Btw, for the study

I understand that wild genes might have a bunch of variables in potency/yield that make them discouraging compared to a commercial one, but I'm very interested in seeing what happens when that gene pool gets revitalized. Maybe its best of both worlds?

I feel like the goal of genetic variety and cultivation just not might go hand in hand. Even if one were to find something interesting in a much wider gene pool, for cultivation purposes it would make sense to then still select towards it.

One thing that I immediately thought were wild apple trees. Large majority of the time they are significantly worse than tasty and large commercial cultivars and I can't imagine lots of effort going towards finding something novel from them. Though I admit, it's apples and oranges, considering the length of time apples are domesticated vs mushrooms.

That is very valid, and might very well be the case. Still, I'm sure at least crossbreeding commercial with wild would be beneficial and easy to find good phenos.

@cosmic: glad to hear! not anything different than usual though, right?

Btw, this is how the author did his samples for sequencing (not that I can fully understand it hehe)

Quote:
Best way to get haploid cultures is to do a 16-streak of basidiospores to separate them. You can pick individual germinating basidiospores with ease. A haploid genome is easiest to work with because there is no heterozygosity to consider in the assembly. I sequenced siblings from the same parent, and most likely captured the genetic diversity that was present in the dikaryotic parent. It is not the standard, different scientists will have different approaches. I used haploids for the sequencing, and then to test their mating and formation of clamp connections. Ultimately, I have a library of haploids that I can cross to make new genotypes heterozygous at the psilocybin locus.

I’ve sent out samples to Alan and also to Caine Barlow here in Australia, both just asked for pieces of cap and stipe dried at a low temperature (I went for 60c) which is what I dry my pans at, also have other friends involved in sequencing and they’ve just requested plates, another guy here in Aus is researching genetic links between cubes and I’ve sent him agar wedges in ziplocks, as long as there’s enough to work with and no cell damage you’ll be fine, not sure if freeze drying would be a good idea though.

As far as crossing I’ve kinda come to the conclusion you’d need to stabilise a little too, simply because most wild clones survived in the wild and managed to fruit in those conditions, give them a high nutrient food source and perfect conditions and they don’t seem to know what to do with it, same with F1’s, majority of those grows are actually pretty shitty, there’s always exceptions though, I’m sure if you crossed F1’s you’d get some very cool mutations happy to grow in tubs. But you’d also get a lot of trash.
My first and only cross ever to fail was a wild F1 x APE, obviously 1 attempt is nothing to go by and I’ll try again and see what happens but it took the wind out of me for the time being.
This is a standard clone grow

Not the most satisfying.
Got one pinning pretty nicely at the moment from this guy

Very curious as to whether it’ll repeat those ochra like stipes.

Yea definitely nothing too unusual than other Cube strain experiences but this one did last a bit longer than a GT dose. The effects lasted longer on both a low and intermediate dose

Yea there's definitely a few ways to send in mushroom matter of sorts to get some proper sequencing done. I asked my friend what a good way to go about it n he said some similar to Alan's info. A piece of dried cap and or stipe would suffice

They haven't really flushed for me, I'm assuming its from the multispore, or that none of the spawn was 100% clean, but they're not all maturing at the same time. I'm looking forward to see the F-2 grows.

Nice! Axman, thats a nice cluster you got.

3AM: I'm guessing the clones are always providing a better flush compared to non-cloned grows?

Has anyone found unique phenotypes so far? Axman, I saw the gnarled fruit with wavy cap in the marketplace and it was very pretty. Did you clone that one by any chance?

I have a neighboring pasture that produces abundant wild GT’s….can confirm they are significantly stronger than any vendor bought cubes, I’ve grown. Happy to share spores or flesh to help out.

Nice flush, 3.A.M! (and I like the picture angles. They are soooo satisfying )

And cool on the giveaway Ax! Not quite of "age" to check it out just yet, but I might look back in ~2 weeks. Definitely interested  And echoing Huskies, real neat cluster there!

@bhg, do you have some pics/have you worked with them indoors? No pressure ofc. I just find this thread catching on absolutely thrilling!

I did not clone that one, currently the only F-1 clones I'm working with are that cluster, and a couple of the ones that were the largest and fastest growing.

My plan is to keep going down the filial line, selecting the first grown/healthiest from my tubs; but also try to go for some solid canopies by cloning and recloning clusters.

The giveaway spore donor was the largest fastest from its own tub.

I fully expect the clone tubs to outperform F-1. I'm hoping the F-2 does as well.

Since you're in LAGM I'm willing to send you one of the prints (not the giveaway fruit but some from a first flush F-1 tub) if you'd like. Just PM me a privnote with your info.

In my experience both clones and F1’s provide pretty average flushes, the one above is an outlier, as far as unique phenos, I wouldn’t say it’s unique necessarily but

Definitely hefty
