Hello,
Trying to do spore print transfer here. can't seem anything to grow but this.  Does this look like mold vs myc?  If I keep transferring this sample, will it eventually show more myc that this fluff??

Hello and a happy new year!!

New year & new grows… had consecutive contam problems with my substrates by the end of last year and set everything aside until now.

I still consider myself a noob when it comes to agar and I’d really appreciate comments on these two for my learning curve!



These are from my second transfer, donor plates of first transfer were cold stored and a few months old.

1. Some of my (poor man’s) agar plates look healthy and strong, some look disorganised and weak. What is the likely cause for this?

2. I would personally now transfer all of the plates to grain as long as they look clean, would you? Or would you do more transfers until they were all thick and uniform?

3. Is there a difference in transferring before 100% plate colonization versus pre full colonization?

Admittedly I’m clueless when conducting a transfer on which part of the mycelium growth to carve a slab out of. Been doing this on pure luck. I’m 99% confident that this has not been the cause of my previous contam problems as they have arisen toward the very end of the entire process, during pinning. Going to adjust my tubs for a more sterile setup first hand. See where that gets me.

The second plate looks clean to my eyes. The first...maybe, if you remember the wedge rolling around near 11 o'clock.

Assuming identical media and external conditions (temperature, light, gas exchange), weaker growth can be either genetic or due to varying levels of (likely bacterial) contamination.

Transferring before the plate is filled gives you an opportunity to grab growth from a leading edge, which is harder for contamination to hide in.

Spawning to bulk is not sterile. Increasing "sterility" at the fruiting stage is not going to help avoid contam at pinning. Clean and fresh spawn and being careful not to have free nutrition (uncolonized grains, starch from porous utensils or buckets previously used for grain or food and not sufficiently washed) added to the bulk substrate will.

Hey yall. I did some pre-pour agar this weekend. I used dollar tree green lid sure fresh containers. They kinda compressed. I can upload a pic soon . I used to use these 6years ago for NO POURS so idk if that's where I went wrong? or do we think the material used just isn't pp5 anymore? Also...how do I avoid condensation? for PRE pours this is my first time . Tanks in advance

Made some water agar and LME. Gel food coloring wasn’t coming out very easy and accidentally squirted way more than intended, also ran out of agar before filling all the plates. :epicfacepalm:

I have made over 100 plates using ketchup cups.  I will continue to use them, they work really well.

Something that really helped me is a $30 Amazon Sous vide to keep you agar temps during pouring

I've been lazy with my agar work, I have a few bisporus washington plates that are fully grown, but then they bruise brown a bit. Is this normal for their mycelium?

These are left overs. I did a small run of prepour and decided it was more hassle than it’s worth. Bought a case of 500 celltreat grip ring plates. First time I touched one of those plates there was no going back. :lol:

I have these there are 50x, they're PP5 and 65mm (measuring the agar from the bottom). They've worked well for me thus far, not as clear obviously since they're PP5 but I like that they lock so I don't need parafilm, clingwrap, or grafting tape. If you need something that has been reviewed there are these which are basically identical but more expensive.

Note to anyone doing this don't put the bottle into the water bath right after taking it out of the pressure cooker it will crack the glass I found this out the hard way :frown: and I used to work in a lab lol but never worked with things coming our of a pressure cooker hot.. :/
