Okay I may do that, now I see some small pins forming, any way too much moisture on surface may delay em or the opposite, I know those areas grew a strandy candy cane white color(like the white looked spiraled),just try to find out all easy fixes the next go around. Thank for advice, am I correct in that you take the inside flesh of one the large fruits and extract a sample and go to agar to isolate and reproduce or is it isolated already so just grow more cultur res from the culture?

Taking the inside flesh of your best fruits is good.

You can put it on agar, in liquid culture, in a grain jar..

I would take a spore print, and work from spores. It will act as a genetic "reset".

The 2nd flush might come in better too.

You can also access the Marketplace and pick up some prints or swabs.

That's one way to do it. Advantage being that you see the fruit in the tub, maybe even eat (or analyze) some of it to test potency, and therefore have a solid idea of what you'll get from that culture. On the other hand, if your spawn is iffy- quite likely if you started with spores to grain- then you might end up with some bacteria in the mix that are hard to shake. Also, you have to be careful and/or a little lucky to ensure that no contaminants from the surface of the fruit make their way onto your sample (one of the reasons people often recommend pulling the fruit apart rather than cutting it in half).

But even just germinating spores on agar and then doing sequential transfers will narrow down your genetics. And there are tricks to make that more effective, like catching the growth while it's still expanding across the plate and pulling from the sectors that look most promising. All of this is typically done to capture clean growth and get away from any contaminants in the spores, but it also helps get the genetics into a tighter range. Especially if you're focused on that as a goal.

I personally have just settled into cloning plate pins: once I'm reasonably sure I've got clean growth in a plate, I'll just let it fully colonize, and then sit, in the hopes that it'll kick out a few pins. Less worry about avoiding or escaping contaminants, as the fruits formed in a sterile environment as opposed to open air. The drawback is that I'll still have to grow it out to see how it actually performs in tubs, but most cultures have worked out just fine.

Is there any drawbacks to not letting the plate fully colonize? I guess I've just been waiting bc I figured more mycelium, but I suppose it wouldn't really matter if its more time on agar or more time on grain. I guess it never occurred to me to send a plate that's half colonized, but fugg it - I've got a PE plate Im anxious to get moving.

My first attempt at:
Holy Grails
Using a leather punch for transfers
and
Cloning a fruit to agar using the Josex poke incorrectly :foreheadslap:

"White" Golden Teachers - store bought LC syringe -> LC broth -> agar (pastyplates) -> agar (holy grails)




Plate #1 (top right) - the dark stuff on top is a result of not cleaning the new leather punch to remove any oily residues, as many steel tools have... When I put it to the torch, I didn't see the gooey, burnt, black stuff that had formed inside the tube. So that one effectively has a little grease on it (2 tiny dark spots on either side). But the myc seem to be moving onto the agar.

Plates #2 and #3 look pretty good so far. #3 got a double up because part of the transfer bit got stuck on the punch, so I dropped it in there also.

Clone plate - who knows? I used a sterile needle to poke through the stem of the biggest outdoor fruit I harvested the other day. I forgot about the sterile water part, but I had some nice, clean air in there to help spoot it out

I will watch it with radical optimism, but tempered expectations. Can't win 'em all, but I can learn from my losses.

It may take a bit longer if it’s not fully colonized but most people only add a wedge or two in a quart.  You could take a plate that’s only 25% colonized and chop it into 2-4 pieces and throw it in a quart and that’s basically the same thing.  I’m sure there’s something to be said about it growing out more so you can ensure  any sneaky contams don’t show up but that’s more personal preference.  I think the majority of the time I send plates that look similar to this one
