I think bags are generally for fairly small amounts. Pasteurizing large amounts of substrate (usually straw, for oysters) tends to be done in big mesh baskets in drums of water heated over a propane burner; the basket is then just left to drain while it cools. Compost or manure based substrate could probably be pasteurized the same way, though I'd guess with a finer mesh (brew bag, paint strainer bag, or pillowcase).

Scale is a funny thing - a completely overwhelming amount of substrate for me would be pretty small for a commercial farmer.

Edit to add that I found sous vide pasteurization of casing to be a bit tricky since the sealed bags still had a lot of air in them. You have to weigh them down. (Though I used a fairly weak vacuum sealer for those and cheap poly tubing as bags. My chamber sealer would probably have done a much better job.)

That's super helpful now that I think about it pasteurizing in brew bags or something makes a lot more sense to me.  I guess you can always strain to desired capacity.

Antifungal will stop spores from germinating but not cultures from spreading

As Nichrome already stated If you can't get good jars all you will do moving to bags is ruin more grain.

Learn proper grain prep and good aseptic technique for your agar till you get it down.

I feel like I've experimented so much and realistically eliminated so many variables at this point that I wonder if it actually could be dry grain. I have generally been doing no-soak in all but one attempt, which involves boiling until a few burst grains and opacity all through the thickest grains. Then I spread out to steam off, load and PC.

My impression was this should be adequate hydration. Just about willing to follow any lead though at this point. People will point to culture and sterile technique, and I get it, but I would happily bet my house at this point it's the grains. I've just tried too many different cultures and emphasized sterility in the SAB too much to have the same exact problem every time in every jar.

Hi!

I poured a few plates yesterday and left them un-innoced for a few days to make sure they're clean. But today I noticed that several of them have these small brown irregular specks on the surface.

This picture's taken about 15 hours after pouring. Can you tell if I'm looking at some sort of contam or is that just burnt agar that got scraped off the bottom of the pot (I'm continuously stirring the agar while boiling).

maybe not so quick question please fellas

When pasteurizing, are people ramping up slowly and ramping down slowly to/from temperature? What is the best practice?

I have been wetting the sub in the vaccuum bags with 80Β°C water to get close to 69Β°C from the start, then sous vide for 2 hours once bath is at temperature plus an extra ten or fifteen minutes to allow for temp equalisation of my bags, which are made very thin, so that they come to temperature with the water very rapidly as it is climbing to temp with the sous vide

When I finish I just remove them and leave them on a shelf till cooled, then cut a slit to allow some air back in so everything is not anaerobic in there while they wait a day usually to get spawned

does this all sound alright and like best possible practice?

Or should I be rapidly cooling bags after pasteurization in a cool water bath and/or allowing them to rise to temp over an hour or more as the sous vide warms the bath?

I also assume I should be allowing air back in also and not leaving them sealed and vaccuumed there until use?

My process works fine it seems but I wonder what the gold standard is?

Thanks, good times

I use warm water as well to get the substrate closer to temp faster. Don't go by the water temperature when starting your timer, you have to go by the temperature of the center of your substrate. Mine usually takes about 45 minutes to an hour after the water hits the target temp to get the center up to temp. I use a cooler and a sous vide, and when I pack my vacuum bags I flatten them out to be about 2 inches thick. I pack them close together in the bath but not touching and I make sure the water circulates around them. I also wait for the water to get to temp and then add the bags. I have a sacrificial bag that I leave a temperature probe in with the substage and partially seal it so the temperature probe wire can hang out of the bag. When this gets to my target temp I start my 2 hour timer. When that goes off I pull all the bags and let them cool on a wire rack before using them. I reuse the sacrificial bag of substrate for the next batch once it's around the same temp as the new bags that need to be pasteurized. I leave them sealed for up to 3 days. Don't open them and let them sit or they might get all the bad things you just killed back into them.

It’s probably just a clump of lme or something. You don’t need to make agar on the stove. If you don’t have a stick blender, get one. But what I do is dump my agar and lme into a quart jar, measure my 500mL of boiling water, dump that into the jar and hit it with the stick blender. Then if you’re sterlizing it, you can put your lid on and into the pc.

I do pasty plates still, so I pour those and then pc em.

Now that I have a Excalibur dehydrator, I notice that stuff gets cracker dry WAY quicker than 24 hours. Is that your experience?

Also, I left my desiccant packs open accidentally. Should I just plop all of them in the dehydrator to reset them?

Also, I didn't prep my buckets last night, kinda feeling like trying out warm water for my coir instead. Is it truly fine?

I know of other methods.

I use pasty plates because I have around 100 of them already, they've survived hundreds of PC cycles and I'm not wasting any plastic.

Jesus christ I'm so pissed, second time in a row the media bottle boils over in the PC, spouting half the agar out. I shouldn't even bother pouring what's left of this into plates, right? I'm trying to make some PDA, give LMEA a break, but this potato starch I bought seems to just make the agar explode when in the pressure cooker. I didn't have this problem with potato flakes. Anyone been in a similar sitch? Why's it keep doing that? I noticed when I'm boiling it in the microwave there has been, both times, solid chunks that refuse to liquify. I'm going back to the malt for now but fuck me, I got plates that need to be transferred and I just delayed myself another two days.

Not really a "quick answer" anymore since it's been three days but you could just boil a few potato slices instead of using potato starch or potato flakes. It's how I make my PDA and I've never had any issues with the bottle boiling over.

Quote from another thread where Bod quoted the following:

Quote:
Potato infusion can be made by boiling 200 grams (7.1 oz) of sliced (washed but unpeeled) potatoes in ~ 1 litre (0.22 imp gal; 0.26 US gal) distilled water for 30 minutes and then decanting or straining the broth through cheesecloth. Distilled water is added such that the total volume of the suspension is 1 litre (0.22 imp gal; 0.26 US gal). 20 grams (0.71 oz) dextrose and 20 grams (0.71 oz) agar powder is then added and the medium is sterilized by autoclaving at 15 pounds per square inch (100 kPa) for 15 minutes.

My 1650w home made dehydrator will knock out 10lbs of cracker dry fruit in 12 hours, but the 24 hour thing is a general guideline used since there is alot of variability in fruit sizes, heating power and airflow amongst commercially available dehydrators that should get everyone to the goal of completely dry fruits.

Yeah, just toss the dessicants in the dehydrator to reset them.

How long should agar be out of the frige before using it to inoculate grains?

And after inoculation do you guys seal the rim of your jar with cling wrap or parafilm?

ru colonizing ur agar in the fridge? or ru storing uncolonized agar in the fridge?

i wrap cling wrap around the edges of agar dishes. if u mean jars then no u don't have to wrap anything around the rim

I got dishes pretty well colonozed then have been storing in the fridge so they stop growing abd I can take transfers to jars/ new plates. I just had to catch up abd make the jars

I do cling wrap plates, but could swear Ive read a few threads where people will seal up the area where the lid meets the jar on their grain jars and liquid culture jars as well. I was wondering how prevalent that practice is

And i need to make plates too. If anyone has a link to good pp5 deli sauce  containers I wanna try those

I think you could put the agar straight from the fridge to the jar if not then just let it warm to room temp, i'm not sure tho. an i haven't read about anyone wrapping their grain jars, if it's PC'd and sealed idk why you would need to. only thing i can think of is putting foil on it for PC'ing
