I agree with you. I guess we will see. I hope they're both good so I don't have to retest, not knowing which is which. I know the one looks far more suspect at this time.

I don't think your stick-on SFD is going to heal the way silicone or rubber does when you stick a needle through it.

You are trying to make it too complicated. Just open the lid for a moment to draw water in. Using shallow jars (e.g. 4 oz or maybe wide mouth half-pints) makes it easier.

There's an explanation a few posts back on what I'm trying to do and why. I don't need anything with the ability to self heal, I just need an entry point that will maintain the jars sterility until I use it.

You’re kinda overcomplicating getting water in a syringe ain’t ya? PC your liquid, then open it and draw fluid into a sterile syringe. The filter is a whole extra step that’s just going to introduce contam, no?

Like the side of the filter you’re penetrating isn’t sterile at all

Yeah the GE filter is unnecessary and possible fail point.

As stated, sterilize water, open the vessel and fill the syringe.

No, Im going to put LC in a syringe, do a Josex Poke, and instead of expelling the myc, I'm going to suck up a bit of sterile water and if all goes as planned, I will have a syringe of LC. I dont trust myself to maintain sterile procedure while drawing from an open jar with a syringe, so I want to solve the potential for user error with a port that I can punch through, for the sake of ensuring that if this fails, it's bc my idea sucks and not bc I sucked at executing it. I was under the impression that a virgin inoculation port would be safe if I kept it covered with foil until I was in the SAB, thereby making it safe for one time use. All input is welcome!!

Ill explain what led me to the idea in the first place, so you have an idea of the issues I'm trying to work around.

1. Im clumsy with small tools and fine movements and still very unsure of myself during SAB work.

2. I dont feel confident in my ability to completely open a jar and draw from it with a syringe. I dont think I could extract the plunger on a syringe without both hands, and there's no way for me to keep from hovering over the open jar with my hands, in that scenario.

3. I want to use a syringe for innoculating grain instead of pouring it right off the LC jar/bottle bc I have zero faith in my ability to keep the pour to anywhere in the ballpark of 1cc per jar. I will turn my spawn jars into soup.

4. The single use nature of LC is kind of a bummer. If I could create the LC inside the syringe, I could do small batches of grain and still have the leftovers for the next batch. I'm not at a point yet where I'm innoculating a ton of grain all at once, so if I could make one or two syringes in one shot and have the ability to get many uses out of it,  I would really only need to make LC once in a blue moon.

So yes, the way I want to do it myself might differ from what someone with a steadier hand and more experience might do, but right now, I just want to find out if it will work out well, as a concept. I'll gladly do extra work to ensure the results are accurate and not due to my rookie ass botching the whole thing. I just want to find out if it will work as intended. Id love to be able to contribute something useful to the community instead of feeling like I'm just riding coattails and asking to have my questions answered, ya know? I'd love to figure out it works and then have someone more skillful fine tune the whole thing and do a write up on it, but I'm not that guy...yet.

I intend to get a nice freeze dryer at some point but for now I don’t see an issue just throwing it into the dehydrator for about 12 hours.

Mine doesn’t go up that high, but everything comes out cracker dry.

Hells yeah I'll get a normal food dehydrator then and I can make my apple chips ^^ it does say you can set it to 95-104 degrees.

I dry at 160 Fahrenheit for 24 hours minimum. Give this a read. Notice the responses from the TC’s Cracker dry comparison

Edit: fixed a word

Both very good information to have I'm going to really have to study. Thanks

open air non sterile grain inoculation/colonization for outdoor?

I've seen contam spawn and jars grow outside with no worry about contam, wondering if i can take a bucket of grain, soak it outside , introduce some spores, myc or LC/LI and cover it with a lid or towel and let it colonize skipping all pc or pasteurizing etc?

What you are planning is way more complicated and likely to fail. The list of vectors in that process you are describing would make this not a "quick answer".

When planning anything if you find yourself saying:

The idea has plot holes and needs work before launch.

I understand you have concocted a method from pieces of other methods and you want it to work, and you want to feel validated about your idea, but the fact of the matter is, there are people with tons of experience who are fully confident in skills like sucking water into a syringe, who are advising you to KISS. I love you and I hope you are successful. Anyone here is just trying to help you get there faster and with less headache.

If you "don't trust yourself" to do clean work, your complicated method is doomed, or at least in the hands of mere luck. It seems like you should just brush up on simple practices as a goal and build that confidence in doing clean work. Complications and ad-on's will not compensate for a lack of skill.

Man LC is actually pretty simple. You just add a clean sample to a vessel of broth. If you can PC a jar of nutritive water and then place a sample in that jar, you’re good. You don’t really NEED a syringe at all. Syringes are an additional vector that needs to be controlled for.

Open a jar/bottle, take a sample, put the sample into the jar/bottle. Then, if the inoculation was successful, you open the jar and pour it into your grains.

Here’s how this hobby works: Learn 100 things. Then over the course of a year stop doing 95 of them. It’s really so much easier than your mind is leading you to believe, and any extra steps and tools involved creates additional vectors for contamination.

is a low g2g ratio slow, i did a g2g with a ratio like 1:30 or 1:40 and it has been around 7 days and the jars are around 5-10% colonized. i also believe now that shaking after g2g is bad because one jar i didnt shake at all after g2g and its the fastest

I agree with Nichrome. As an avid Josex poke to LC fan I like your idea of trying to do it all in one syringe. But if you have to poke and then draw water up, it doesn't seem any less complicated to me as opposed to poking to LC, letting the LC grow out and then drawing it into a sterile syringe if you absolutely need to have it in a syringe. If you're gonna give it a try I'd at least ditch the injection port idea.

I always poured LC, however since getting a FFU I've been more adventurous. Here's how I've gotten drawing a syringe one-handed to work.

Start by holding the plunger with your middle finger and thumb against the little finger tabs on the syringe:

Draw the plunger back using your index and ring fingers as far as you can while pushing against with your thumb and middle fingers:

Eventually transition to gripping the plunger with the palm of your hand to pull the rest of the way:

Everybody's hands are different but hope this helps

So anyway, uh...

LC in a jar is extremely easy. You can still use a syringe to noc with too, just suck up some lc into a syringe. That simple.

The LC in the jar is literally 2 ingredients PC'd for 15 at 15 post vent. Wedge went in the jar and swirled some times over some days. Used a 60 ml syringe with a 4 inch stainless 14 ga luer lock dispenser to suck it out of there.

Very interested in this. Im following along. You should start a separate post for it.

I'm curious what the LC does in the syringe in the PC. If it will stay in or boil out. Create a vacuum and pull in air while cooling?

I'm wondering if you should just fill it 3/4 full or fill it 3/4 then pull the plunger all the way back to fill the void with air.

I'm gonna try myself when I get back home. Hoping to see your results here before that.

Great idea.

I appreciate all the constructive criticism, and I will say that this was 100% a "hold my beer" idea that I came up with yesterday while sitting at work. I would absolutely agree on the point that it would make more sense to develop my skills to the point of being able to do it the way that has already been established. But the problem with that is that I was never great with fine, precise movements to begin with, and then I severed two tendons going to my thumb in my dominant hand, which has had some lasting effects, though not as bad as it could have been. Im not worried about transferring a wedge to the broth - not a problem. Im worried about trying to pour an amount to my grains that isn't going to result in too much water in the grain jars. You could offer me a date with Jennifer Aniston if I pull it off, and I'm still sure I'd screw the pooch on it. I'm sure that I will get better with things as I get more practice, but I think I will always have an ceiling lower than most. Im going to practice drawing liquid with a syringe using only one hand - I think if I could figure that out, it would take a significant bite out of what's holding me back. I appreciate you guys providing feedback!

Idk if air would be enough to draw the cells into the syringe, and it feels sketchy to draw air into a syringe and hope I didnt bring contamination with it. If I had an ffu, maybe...idk. idk wtf I'm doing, but it's gonna cost me maybe $1 and some of my time to try it, so cowabunga it is!

if fruiting a clone, how different are spore prints from one fruit to the next in terms of stabilization?

if you fruit a clone and take a tissue sample from any of those fruits it is all the same as any other fruit from that spawn? and/or the same as a transfer.

selecting one fruit to clone from each ms grow, then taking prints from every fruit of the clone grow, mixing them in one solution, growing it out and selecting a clone...

Thanks, I took your advice and this is what I ended up doing. put around 2.5 or 3 cups of water around the sides waited 24 hours and removed about .25 cups of water that hadn't absorbed into the sub. It's looking a lot better now. There seems to be more healthier growth coming in now. Still not that dense but at least it growing a lot better.

Hoping it will pick up more after a full harvest and probably a dunk. Thank you for the advice.
