Whenever I’m told to just throw away something that’s not working, I try to save it, with minimal effort, while advancing other projects.

That’s why I can’t bring myself to throw away these disgusting, but densely colonized bags.  Keep thinking about casing them, or soaking them and covering with verm and putting them in a water chamber, or burying them in a flower pot, or even spawning to bulk.

At a minimum, I’ll put a sample of each to agar, and eventually I’ll chuck em if they don’t fruit.

Meanwhile…

Agar pieces stuck all over the jars in inconvenient spots, no matter how much I whack em.  The avery albino agar mostly landed in the grain, but the albino mvp just wanted to be high up out of reach of the grain.  According to a post I found somewhere from a TC, it’s fine, the myc will crawl down to the grain, but laying it on it’s side is also acceptable, so we’ll see how it goes.

Attempting Pasty’s unmodified lid tek.  Once they’re colonized a bit I’ll give em a good shake, stand upright and loosen the lid for fae.

I have 4 more grain jars to inoculate.  The FP+ plates are just about ready, as are a couple of other plates, but I want to transfer a little wedge to a fresh plate to keep the culture going.  I used my last two blank plates for the two aggressive albinos, but made four new jars of brf agar last night, as well as a jar of water agar.  Haven’t poured anything yet, but I have such plans…. More pics and rambling to follow.

Second flush of Vogon B+ seems to be on its last gasp.  Watching for now.
Why is this APE trich bin still sitting in my grow area?  Who is responsible for this?!

Auto-Summary
The post discusses various experiments and observations in growing mycological strains. Key points include:

1. Bacterial recovery challenge: The author pours water agar plates and cuts out bacterial blobs to potentially transfer mycelium.
2. Four empty grain jars: Plans for inoculating jars with B+ PFPuck, Tampanensis, Avery's Albino plates, and Golden Teacher strains.
3. Avery's Albino Plates: Three transfers from 11 days ago are ready for grain, while the original and three-day-old plates show mixed results.
4. Albino MVP Plates: The plate from 18 days ago shows potential pinning, while the three-day-old and 11-day-old plates are ready for fruiting.
5. Golden Teacher Rescue Plates: Several plates show signs of potential fruiting, with plans to decontaminate and transfer to BRF agar.
6. FP+ The Mail Order Agar: The author plans to fruit the awarded strain on a 66qt BEEFYTUB, possibly using Pastywhyte's EZ Dial Monotubs.
7. "Azzie" Psilocybe Azurescens: Plans to transfer the strain to a new plate or use LI for fruiting.
8. Vogon B+: A

Bacterial Recovery Challenge

As part of my ongoing education in cleaning up agar, I have this plate of mostly bacteria, but also some spores.

I theeeeenk there's some mycelium buried in here in a couple of places.

I'll be pouring some water agar plates, and I'll cut out these cursed blobs and see what they do.

Might make some pfpucks to transfer them onto if there's any myc recovery

Their looks are laughable, unphotographable, but here's the best I could get.

Just take my word that in-person, these blobs have some shimmery white filaments underneath.  I think some of the yellow is the fluid mycelium makes to kill bacteria.  Weeee shaaaalll seeee...

Four Empty Grain Jars

I've got four rye jars ready to inoculate.  Candidates:

B+ PFPuck
This B+ PFPuck is pinning.  After transferring some pins to BRF Agar, and maybe doing a Josex Poke, I could Tiger Drop 3.0 this puck into a jar, which could become a master for G2G.

Tampanensis
I absolutely want some long term rye jars of Tampanensis to harvest for truffles in a few months, and the sooner I start, the better.  I have three decent candidates for inoculation source, buuuuut I might be better served using these first jars spawn masters or to be spawned to bulk, so maybe I'll just do one jar to serve as a master for three or four jars from the next PC round.

I'll put some of the pins to agar, transfer myc from the edge of the invisible colony to another BRF agar plate, then use the rest of the entire plate to inoculate one grain jar since this seems to have pretty slow growth, and leave the PFPuck alone to see if that sclerotum actually sclerocifies into a scrote.

Avery's Albino Plates

The original plate was inoculated with purchased LC 18 days ago, and it's curling into a shroom rollup.  I'm hoping it will pin, but probably not going to use it to inoculate grain.  Three transfers from 11 days ago are ready to go to grain.  The plate from three days ago, surprise surprise, is not.  Strong candidates for grain, and a good reason to prep more grain jars to accomodate.  Looking back over the forums, people have used plates to colonize up to 6 jars, though BeefSupremeJr does 1/2 plate per jar, which seems about right in terms of inoculation points.

Albino MVP Plates

The first plate, inoculated 18 days ago, is not curling up, and does look like it's getting ready to pin, so I'll be watching it and then putting pins on agar or PFPucks.  The plate that was transferred three days ago is moving along fine.  The three plates transferred 11 days ago look excellent, and are really hungry and ready.  I should use at least one of these soon.

Golden Teacher Rescue Plates

This is going exceptionally well.  The original plate from 18 days ago of what I thought was badly bacterial infected Golden Teacher myc, the last of my GT genetics, looks like it might be getting ready to pin.  The plate from 14 days ago looks like it wouldn't mind being on grain, and the three-sample plate from 11 days ago looks a little weird but not explicitly contaminated.  I think it would be wise to put this through a couple other decontamination steps, probably water agar to pfpuck then josex poke to BRF.  If I get any pins I'll just put them straight to BRF agar.

FP+ The Mail Order Agar

BeefSupremeJr awarded me this extraordinary strain by sending a thin wedge of agar through the mail, a process which, by every scientific measure imaginable, should not be possible, according to the experts on this ridiculous thread.

Yea verily, witnesseth a witchcrafte most foule, for doth the fungus yet prospere!

I could just toss these into grain and see how it goes, would probably be fine, but I really really want to fruit this strain abundantly.  I've been planning to shoebox my next grows, probably 2:2 quarts of grainspawn to coir, since I still have a pretty terrible ratio of success:contam.  I'd like to fruit this strain on a proper 66qt BEEFYTUB, which is 2:8, and perhaps (?) more prone to contamination by my shaky noob hands.  Probably transfer these out to some full BRF agar plates and let them get nice and strong before sending to grain.  Hell, maybe I'll make a half dozen Pastywhyte's EZ Dial Monotubs and fruit up a veritable forest of this glorious Shroomery original strain from the illiminicent Fahtster.

"Azzie" Psilocybe Azurescens

According to everyone, this strain won't fruit without either being planted outside in the spring under a woodchip bed after winter sets in, OR being fruited in a humidified monotub contraption inside a well-lit refrigerator, so even though the myc looks really happy on its transfer plate and the original is blueing up nicely, I'll probably either transfer this to a new plate, make LI, or do some other third thing.  PFPucks perhaps?

Vogon B+
One of my UB "TEK" bins that triched out donated a chunk of itself to this blobby fucked up agar plate, and is absolutely shredding it up.  Some of the myc reached that plateau and is literally reaching out centimeters into thin air looking for something to destroy.  It might also be running away from hidden contams, so I think this will get the water agar pfpuck josex poke to BRF agar treatment before I let it near grains.

Josex Poked Puck Plate

Funny enough, this plate - produced with a method touted for being an excellent way to weave out contams, appears to be contam'd.  Unless the two oddly circular fuzzy spots are just the B+ myc expressing its individuality, I'm assuming it's some competing myc.  I also know that my Josex poke technique was deeply flawed.  I stabbed a dry needle in without attaching it to a syringe of sterile water, and smeared it onto the agar.  I did this with three separate plates and only this one took off.  I think this guy is destined for water agar, it needs to take a bath.

Risen from the Dead!

Saving the best for last, all 7 of the B+ PFCakes have returned from beyond the grave!
Now I just have to figure out what it looks like when it's consolidated, and then set up a water tub to fruit em.  Huzzah for resilient mycelium!

Tonight's Activities
I've just finished briefly dialing up the water agar and a bottle of brf agar to 15 atmospheres for a few minutes to melt it, I have the sous vide going at 190.  I'll take them both out, pop em in the sous vide and bring the temp down to 140.

While the temps are coming down, I'll sacrifice one of the Albino MVP plates and one of the Avery's Albino Plates into two grain jars each.  That will give me four jars of each Albino cooking, so I can afford to lose some to contam but still have spawnable grain to send to bulk or use as master jars.

Once the temp is down to 140, I'll pour water agar and brf agar plates.

If I'm feeling spunky, I'll boil some more rye grain, following Hamloaf's Easy AF Rye Prep and then scoop it onto my herb drying rack, following the last part of Beef's Wheat TEK.

Thank you to all the TC's, Sophomores, Oldheads, Noobs, and weirdo Dickwads who frequent this forum.  You've given me encouragement, good advice, bad advice, outright trolling, and reliable entertainment for the past year six months three months not quite two months I've been here, and I couldn't be happier.

Don't touch my turds!

Just read this entire thread.

What a ride, where do you get the energy? I feel tired just reading about your journey

You are going to have shrooms coming out of your ears at this rate, you will be bartering in shrooms, making lotions and postions, rubbing them into your armpits as deodorant and popping them under wobbly table legs as a stabiliser.

Love how you put tek's in coloured text with links to them.

As a noob, I definitely like shoeboxes, 1 quart jar of spawn per shoebox to 4 parts coir, which is half a 650g brick with enough coir left over for a pseudo casing, and I cram 4 without lids into a 90L tub with the Pastywhyte’s Ez dialed monotub hole configuration, set and forget so far, never had to mist yet.

I did lose 2 shoeboxes to trich but oh well at least I had that degree of separation and it wasn't a whole big tub, and then I just mixed up some more shoeboxes to replace the ones I lost.

I've got 10 different varieties on swabs here and I've been only growing nothing but Nats for months, I have put one other variety to agar about 4 days ago, but after seeing your journey I'm starting to think I should of put all of them to agar.

A big old smorgasbord of shrooms :awesomenod:

If for no reason other than it’s fun, agar is great.  It’s also an incredibly versatile tool to move myc safely from one form to another.  Feels like alchemy.  Go for it!  Hitchhiker guide in my sig links multiple reliable teks for getting started.

I haven’t read up on nats, what do they need to be happy?

Loads of activity since the last post, tons of transfers to water agar, brf agar, grain inoculated, will post another summary soon maybe.

Wanted to update though because I decided to birth the three most promising appearing B+ pfcakes.  The B+ PF Puck had pinned, so after transferring the pins to brf agar, and seeing that the sides were covered in knots, I figured - what the hell, put it in a water chamber and see what happens.

The water chamber is also where the pfcakes are soaking, weighed down by 4oz jars, with the pfpuck on top.  After 24 hrs I’ll case them with verm and rearrange them in the chamber with the puck.

One of the next projects will be a Bakedbeings Bottle Tek grow.  I pasteurized a brick of coir with rye grain boil water.  Following Yoshitrainer’s low grain prep with 4oz jars, and then add the grain coir, PC, add agar wedges, hopefully have a bunch of mini bottle grows to fruit.

Nats seem super easy, the mycelium eats trich (Too an extent of course, I did lose 2 shoeboxes to trich,  but below plate pin was from a culture that ate trich) and handles bacteria infection well, below also is a picture of 2nd flush of a ziploc that was spawned with bacteria contaminated popcorn.

Same things as a cubes, very closely related to cubes, they enjoy a little more FAE.

I'd offer to send you spores and wedges, but I'm from New Zealand and I bet a kind member from America could supply you with better Nats than myself, faster and cheaper :awesomenod:

I'm really impressed with how many tek's you're going through, you obviously love experimenting.

I find something that works and never change.

:rofl2:

For example, I found the 2% Agar and 2% LME recipe for agar 2 years ago, I throw the ingredients into a media bottle, add warm water, shake shit out of it, put bottle into hot water bath on a trivet on the stovetop until melted, shake the shit out of it and put into 10 min vented instant pot on trivet with a loose fitting cap for a 50 min pressure cooking cycle, let it chill for an hour in instant pot once finished to let temp come down, tighten cap, shake shit out of it, let bubbles settle out of agar and then pour, works great and comes out clear :awesomenod:

Edit: If you want to try Nats, but can't find anyone closer to send you something, I'll absolutely hook you up brother! Free of course, but you might be waiting 4 weeks for a letter

That is fascinating!  The great Alan Rockefeller recently posted microscopy video of a  Ps. Natalensis x Ps. Cubensis hybrid (external link). Makes me wonder if the trich resistance of Ps. Nat could be bred into Ps. Cub.  Might make all those doomed UBTeks viable .

That's my problem, haven't found anything that works!  Right now I'm trying to do everything, get a feel for which workflow I wouldn't mind repeating or scaling up, and also building a repertoire of techniques in case I come across a strain that doesn't like whatever main TEK I end up settling on.

Welp, now I've got another TEK to try.  Thanks!

I was going to try some low tech Nat x Cube Crosses myself, I'll probably try mixing Nat and whatever Cube I fancy spores together on an agar plate, and mixed into sterile water which I'll shot into PF tek cakes, and finally with something like Fahster's method outlined here - https://www.shroomery.org/forums/showflat.php/Number/28117746 :awesomenod:

You said no rush, but I was sending mail out to a man in need in NZ anyway. When you've got the SAB setup anyway, you should do everything all at once right?

Edit: Forgot to mention something, you probably alright know this, but I forgot just now and possibly fucked up my agar, if you leave your media bottle lid on too tight while the agar cools before pouring it will create a vacuum like canning jars, note to self don't do that... I only left my lid on a few turns and it did it.

Auto-Summary
The post discusses various techniques and experiments in mushroom cultivation, including mixing Nat and Cube spores on agar, PF Tek cakes, and Fahster's method. The author shares their experiences with different techniques, such as leaving agar lids on too tight and boiling over, and cleaning up their lab area. They also mention PC'ing rye jars with boozy coir, making viscous LC, and preparing grains for cubes. The author plans to use agar as a tool for cleaning up samples and has high hopes for future experiments with various strains.

aw man, another TEK to add to my growing list

You said no rush, but I was sending mail out to a man in need in NZ anyway. When you've got the SAB setup anyway, you should do everything all at once right?

I always get like this, end up spending hours with the fan off and window closed breathing through a mask and sweating heavily bc I’m also running the PC at the same time and the room turns into a sauna while I’m doing dozens of transfers and any other sterile work I can think of

I’ve definitely done the opposite and had them boil over into the pc water.  It’s not an entirely unpleasant smell, but it sure is a pain to clean up…everything’s all sticky

Got a lot done today.  A friend gets great results hydrating coir with grain boil water.  He immediately steams it for 16 hours while I…left it in the bucket for a couple of days.

It had a distinct sour alcohol smell, but nothing ventured nothing gained…right?  I sacrificed a quart grain jars and mixed the prepped grain 1:1 with boozy coir and made two quart jars of the stuff.  PC’d it and will noc it up at next opportunity in hopes of a bottle tek a la BakedBeings.  We’ll see if cubes like rye liquor.

Also PC’d two rye jars, which will be home to some tampanensis agar.

Since I was making a PC run and didn’t want to waste it, I made a small pile of 4oz pftek jars and a couple pfpucks.  There was still room in the pc, so I mixed up a flask of brf agar and put a splash in a bunch more 4oz jars.  Want to try something I saw on here somewhere- after the agar is colonized in the jar, top with a sprinkle of pc’d grains, anx when they’re colonized add coir to topfruit.

Also removed three pfcakes from a 24 hour soak, covered in verm and set in a water chamber shoebox, along with the knotting pfpuck.

I had some agar left over in the flask, what I’m thinking of doing is just adding water and pc’ing it to be used as viscous LC.

ArkaNaut said:
Semi-Solid Liquid Culture

Small amount of added agar thickens and allows better clinging when shaking incoculated grain spawn

My lab area is an absolute wreck.  And yet my next steps aren’t cleaning up.  Instead, the plan is to scoop all the rye liquor coir into myc bags which have just arrived so I can oc it and stop the fermentation.  I suspect there’ll be some pH problems, but I don’t have litmus paper or a ph meter, and I’ve already gone way over budget for the moment so I’ll just let it ride.

Got more quart jars as well, so I’ll need to prep some more rye to PC alongside the boozycoir bags, if there’s room.

Gave my four noc’d jars from 6/17 a good shake since they were at about 30%.  I suspect my grain’s a bit on the dry side causing slower colonization, but that’s probably better than being too wet from all I’ve read.

Two days ago I transferred samples onto a couple dozen water agar and brf agar plates.  Continuing to clean up my rescued golden teacher mycelium, one plate looks ready to use.

I bought some travel size bottles of distilled water, thinking I might drop a slice of colonized agar into each under the SAB to make LI for long term storage and grains.

If I do make that thickened LC, and it’s probably enough agar and brf left over to make two 500ml media bottles for the pc.

I received apple wood smoker pellets, which I’ll be soaking and mixing with wheat bran, wood chips (a mix of smoker chips) to use for growing edibles.  I have LC coming for lions mane, blue oyster, king oyster.  Might be able to run the azure strain on that as well.

Also expecting lc of mazatapec and hillbilly, so I’m gonna need some grains prepped.  It’s a reliable LC source.  Everything I’ve put to agar has been clean, though I’ll probably start the new cubes on pfcakes as well as grain.

I have high temp silicone and those air filter discs.  Though I’ve been using pastywhite’s unmodified plastic lid tek, but I’ve found that the lids warp in the PC and I don’t know that they’re fitting loosely enough for good fae without popping off.  So I’ll be modifying some lids with injection Ports and fae ports.

My overall plan is to have pfcakes fruiting while getting uo to speed with grains and spawning to bulk/bottletek, eventually switching to grain bags and LI/LC.

Agar will remain as a tool for cleaning up samples.  I just put some b+ spore prints streaked to agar for instance.

Mkay, that’s enough.  Onward and upward!

Only took a few pictures

Birthed, soaked, vermed B+ PFCakes

Grains Shaken

Vogon B+ was giving some sad pins, I had harvested then and dried them since they weren’t maturing, just turning dark and wilting.  0.4g, took them to see if pins are unexpectedly strong - not really, barely level 1.  Took 2.6g remaining first crop stressed vogon B+, barely hit level 2.  They had been dried in a fancy toaster oven with a dehydrator setting at 150F x 20 hrs, but definitely lost a lot of potency compared to fresh.  They looked normal, but results don’t lie.  Since then I’ve purchased a cosori dehydrator, which does a better job, naturally.

Today, if there’s time, I’ll unload the PC.  I need grains, might give this a try from 99.99, though he uses wheat.

99.99 wheat prep without soak and boil

I have red wheat, but I’ve heard that’s similar to rye, so maybe I’ll just use rye.

Yoshi Trainer low preps his rye with almost exactly the same ratio as wheat:

Sterile Work
  • Inoculate Tamp plate into 2 rye jars
  • Inoculate Dirty Teacher onto PFCakes and PFPucks
  • Inoculate Distilled water bottles with all available genetics for LI
  • Inoculate agar jars with clean agar myc
  • Post images of unusually fast growing myc - is it trich or legit? - if legit plate it on brf agar

Documentation
  • Write out inventory of all plates, jars, their sources and derivatives

I just recieved a pack of LC from my fav vendor.  Mazatapec and Hillbilly, and a free bonus LC of Leucistic Blue Magnolia!
I'll be branching into edibles, with blue oyster, king oyster, and lion's mane.  I have some supplies for that, but I think I can't reasonably tackle any substantially new TEKs.  Managing the growing Agar library, deciding what transfers to make, managing the PFCakes, Pucks, and the grain jars, is enough.

My true next step is clearing up my grow area.  Have to move some furniture, set up shelves, tables, decide on storage area, some sort of organizational system, think about investing in a flow hood...

Pictures or it didn't happen...

The second flush off of my only surviving Vogon grow was pitiful, but the bottom had a lot of pins, so I messily flipped the fragile thing over.  Only doing grows with high substrate to spawn ratios from now on, nice thick beefy cakes

It was sopping wet, too, so I left the lid off until it dried up a bit, and I'm surprised to see some acceptable fruits peeking out the sides.

My first proper sanctioned Hitchhiker grow, these back from the dead B+ PFCakes are already colonizing the verm nicely, and the bare assed puck is pinning again, just 24 hours after birth.

That puck donated some pins to agar, and 24 hours later they're exploding into mycelium:

My pinning tampanensis agar plate was crispy dry, and the pins were stringy as hell, I was worried it wouldn't take, but we're looking good:

The badly contaminated Golden Teacher ("Dirty Teacher" DT) donated a few dried out crispy cubes to water agar for further cleaning, not looking too aggressive but it's only 24 hours.

Avery's Albino on the other hand is racing across the agar, visible even under condensation:

Of interest, I've got a big supply of 2" Medipore, which I thought was micropore, and I've been using it to wrap plates.  Holds them together, keeps out the largest dust particles, but does nothing to preserve moisture.  Several of my plates have dried to cracker crisp, but I have read that this is actually a viable storage method, and the myc will wake up.  Gonna try it out with this piece of Albino MVP that didn't make it into the grain.

I made some dirty spore plates off of my first B+ grow.  Used a Bakedbeings technique to drip them onto agar and streak them.  So far just growing bacteria, but it's only 24 hours, and they're nicely separated colonies.  Hopefully some of the spores will appear soon.  You can see the streaking guide I made for myself under the plate

This plate from a Vogon APE grow looks pretty strange.  Maybe trichoderma in between the colonies?  I sampled from areas 1, 2, and 3, and plated to water agar.  Backlit, there is translucent mycelium flying away from the wedges.  I'm worried it might be trichoderma myc, but 48 hours after plating it's probably time to sample the edge and put it to BRF and see what grows.

For comparison, this is trich, though I think this may be on clear nutrient agar.  See this awesome thread by Hindsight for more:  Experiments with Trich

Running To-Do List

Clean and Organize Lab Area
  • Build grow tent
  • Gather loose tools and small objects together
  • Gather and take out trash - Done
  • Rearrange equipment for better walkthrough flow
  • Bring lights up and arrange on grow containers

Cooking/Pressure Cooking
  • Grain + Coir Bottle Teks - Done
  • PFCakes and PFPucks - Done
  • Grain Jars - Done
  • Thicc brf LC

Container Prep
  • Modify lids with silicon and micropore
  • Clean used PFJars
  • Make PF Substrate and Fill Jars - Done
  • Make various bags of raw rye, water, verm, boozecoir
          * 1c rye, 1c water, 1c verm x 2
          * 1c rye, 1c water, 1c boozecoir x 2
  • Make bags of just boozecoir to stop fermentation in PC

Spawning
  • Set up larger water chamber for hydrated PFCakes
  • Place fully colonized PFCakes into water soak
  • Shake 6/17/24 jars - they are approximately 30% colonized - Done

Fun thread to follow!

I don't see any reason to have a grow tent unless you are growing a plant. I also don't see a reason for a water tub if you can just shred them to normal tote.

Just a bird's eye 2 cents

Appreciate the feedback!

The growtent is mostly just for the shelves right now.  Someday I might turn it into a martha just 'cuz.  Now it's just nice not squeezing into my closet to see my progress.

As far as the "water tub" it's just a shoebox with water in it.  what do you mean by "shred them to a normal tote?"  I'm all about simplicity, believe it or not

You can shred a cake and treat it like straight grain by spawning into a tote of the corresponding size.

If your plans are turning the tent to a martha it is worth keeping set up if you have the space
