inoculating pucks with a print and an old syringe * 9
    #28243180 -

anybody who knows me knows i pucking love BRF fucks, but starting them from prints can be annoying. loops get messy, dropping a swab in there is weird, and making a spore syringe is extra work

the solution?

fill an old syringe with water and sterilize it, scrape spores into the tip of the needle, and flush them out. repeat for as many inoculation points as you want

for anyone unfamiliar with pucks:

1 part brown rice flour mixed with 2 parts vermiculite and then  1 part water, loaded into small jars (in the shape of a hockey puck) and PCed for 45 minutes or steamed for 2 hours. this mixture is magical for reviving stubborn spores and outrunning bacteria. cultures can be grown out and fruited on pucks, or tissue samples can be passed to agar for further expansion

The josex poke for prints, I love it!

I've started using BRF pucks because of how much you tout them, I've had a lot of success. Thanks for spreading the knowledge.

I'm going to attempt making my own syringe this weekend... what do you find is "extra work" compared to what you did? I may attempt this way...

if you have reason to make a syringe, by all means make one. i have nothing against them. but for inoculating pucks this way is just faster, and allows you to work multiple prints with one syringe

It seems that there's no holes for filters or anything. Is that correct?

Also, would it be ok to use those 4oz containers typically used for no pour Agar?

filtered and unmodified lids are both okay, and pretty much any container is fine

I've also come to love pucks after Josex' Poke helped me get rid of some annoying, pretty persistent bacterial issues. I very much fucks wit pucks, too.

That being said, I wasn't planning on using them for prints yet but if I ever decide to start with pucks, this will come in handy, cheers! Bookmarked. :super:

The puck turned out great in this sauce container!

The myc of those Atlantis sclerotia grows very fast. Let's see if it can outpace bacteria this time. Third time's the charm they say.
