Contaims at inoculation points

Hi I just started my first attempt. It was also the first time I used a pressure cooker and I think thats where I went wrong because my jars all had water under the tin foil and seems very wet. I used three sperate syringes with different stains. My A strain syringe started colonizing after 48 hours and within the next 12 I noticed it was getting blue spots and now It is most definitly contaimed. My question is since the contaims all started exactly at my inoculation points whould that suggest a contaimed syring. ( my other jars were innocculated with a differnt syring and have no signs of life yet)

it could be the syringe or it could be your technique. maybe you weren't as sterile as you should have been. if your jars gained water during the pc process then they will have a hard time colonizing if they do at all. this depends on how much water was soaked in by the cakes. what are your mix ratios and how much did you inject into each site? peace

contams*
And yes, it was probably the water...
Take one of the contamed ones outside and open it and see if it's really wet..
Then wash your hands and sterilize them with Rubbing Alcohol...

Where'd you get the syringes from?
Use:
1/4 cup BRF 1/2 cup Horticulure Vermiculite 50cc's water
The PF tek says use 60ccs but after i made my first jar with that it was soaking wet and pooling at the bottom, so I dumped it and redid it with 50cc's.
I did ALL my jars with 50ccs, and there is still water in there, but none pooling on the bottom like with 60ccs.
This was 10 days ago, and 5 of my jars are 60-80% colozined, and 3 of them are 20-45% colonized.
When you PC use 1/2 - 1 inch up on your glass jars.
I used 1/2 inch...
And double foiled the tops, and I made sure that the foil on the tops didn't touch the PC water.
Hope this helps and Good Luck...

Well I varied my mix ratios. But the jars that were contaminated were 45cc, 50cc, and 55cc they were quick to start to colonize but so were the contams. I didnt flame my needle because it was fresh out of the package that read steril on it. I used just a little over 1cc per jar. The syringe I got came from the FSRC. I think were my problem is that never using a PC before I just turned it up on high and left it to start to shoot steam out when I heard it I came back in to find water bubbles coming out the pressure relife valve. I think tommarow I will try again just keeping it on low. Original you said not to let the tin foil touch the water..I'm pretty sure that it was because I basically tryed wraping it around the whole jar. Trying to keep the water OUT. Thanks for the advice I gotta work on my techniqe I think.. I just didnt know weather or not to bother trying to use the rest of that syringe seeing as the rest of my jars are so far so good I checked em again not to long ago and there is some nice healthy whites poping up on the glass

Hi Musher, you shouldn't have to flame your needle right out of the pakage, but I would reccomend wiping with an alcohol wipe between each jar. By all means try the rest of the syringe, but in the meantime, PM me your addy and I'll send you out another. Just in case it was the syringe
