What do you guys think is in this plate? It's not growing on top or on bottom, it's looks to be more within the agar. I hadn't changed my process which has severed me well for years. The only thing I can think of is maybe the LME is too old? The agar recipe (LME and agar) as well as the PC process used were the same as I've always used but all of a sudden this started showing up in most the plates. Do you think it's some sort of sediment and the plate is okay to use?

Honestly. That kind of looks like the remnants of potato flake when I don’t completely dissolve it in my agar. Just watch it.

Majority I’ll say a pray and flush them, but if they are pretty or chubby, I’ll play taxidermist and keep them in cryovials for an afterlife.

That and my room is small, I had to get fancy to fit a 3x2. It gets warm. If left on it would be like a sauna up in there. Even after an hour...

As others have mentioned a print is better than a MSS. Making a MSS just dilutes the spores and creates additional unnecessary vectors.

Still learning the search function but I'm having trouble finding any write ups on Master/inoculation jars. Are there any up to date explanations on the process and follow up steps like storage/use? Wondering for educational purposes. Links would be greatly appreciated. :crazy2:

When do you know when to take a second transfer when cleaning on agar? I made my first transfer when it looked like this:


It's been about 5 days and they have definitely spread out on the dishes but the myc is so light that I couldn't even good a picture of it. It's probably spread out about an inch from the center. But it looks like it's still very delicate at this point.

Then I took another couple transfers from the original dish 2 days ago because the myc had gotten much thicker and ropier, so it looked stronger. That dish has already gotten some extra  sprouts off it and they are much thicker strands.

Besides my wonky, not thoroughly mixed plates, I don't see much in the way of contamination. Should I just let them go until they are more solid white and ropey? Or is there a tek I'm missing that shows when it's good to make the next transfer?

This is about as good as I could get. You can see I put 3 transfers on this same dish. It's spreading out but it's very light looking. Should I just leave it a while longer? Like I said, this is about 5 days of growth since that picture above.


Here is the original plate today, obviously a little mangled from transfers being taken:


And here is the transfer I took 2 days ago after it got a lot fuller. Not as much growth but it's coming in a lot thicker.

How do these plates strike you? Sendable once grown? To be honest they look similar to what I transferred from, but the growth looks uniform and quite nice to my eyes despite being very tomentose.

I'm never a fan of the halo around mycelium like that, your 3rd plate has such a large halo it would concern me.

I'd let that plate continue to grow to see if the growth becomes more organized.
