Just a Question ? Has anyone tried Pectin as an agar substitute ? I know that when you cut a large grape stem a large amount of pectin comes out which is very gelatious at room and higher tempatures . Just a thought and if it worked a whole lot easier to find . I know that grape jelly does not liquidify during open air at room temps so perhaps store bought jellies would even make good growth mediums ? Just a thought guys , give me some feed back .

Usually stuff like that denatres and becomes clumpy when you heat it enough to sterilize it.

there's a dissenting opinion floating around that petri culturing is antiquated and obtuse. there have been some arguably more refined methods discussed.

One of the key aspects of a petri dish is unhindered visibility. It is rather simple to establish a secure seal on petri dishes. Petri dishes are stackable and durable and autoclavable. Manipulating agar inside petris with a scalpel and spear is a simple task for anyone capable of writing with a pencil and possessing some common sense concerning airflow and/or gravity.

i find it worth noting that pectin is very present in apple skins.

hold on.... Pectin is used to make jelly and I have pCed jelly to seal it. There was no breakdown or clumping..

I have wondered about pectin my self.

Has anyone tried it?

Pectin is sugar. I would think culturing in pure sugar would have detrimental effects.

That is odd everything I have says it is a colloidal carbohydrate found in fruits and vegetables.

"a white, amorphous, colloidal carbohydrate of high molecular weight occurring in ripe fruits, esp. in apples, currants, etc., and used in fruit jellies, pharmaceuticals, and cosmetics for its thickening and emulsifying properties and its ability to solidify to a gel."


agar-agar is a colloidal carbohydrate found in sea weed.

"The water soluble colloidal carbohydrate of the red seaweed, Gelidium; forms gels with as little as one part to 500 of water."


I am still wondering is anyone has tried it?

I suggest that you try it and let us all know what happens. Be the pioneer! I have read about using malt extract from home brew stores.

Something like 1:25 malt extract to distilled water.
boil,
put in mason jar with a hole in the lid,
autoclave or pressure cook (or maybe even microwave),
innoculate, and then watch the white fuzz grow.

Does this work?

yea but it defets the perpose of using agar. agar is solid so you can isolate things you want from things you dont. with malt(liquid) this is impossible. or nearly immposible.

No.

sugar=carbohydrate , yes, but not the other way round.

The reason why agar is used is that it is not easy for microorganisms to metabolize thus is stays firm. Gelatin for instance liquefies once colonized by mycelium.

Try what happens with pectin and report.

i didnt say carb=>sugar. just the equal sign. so how can that make a difference?? ina equation you can flip things around if you want. glucose , frustose is a carbohydrate.

please explain im confused.

carbohydrate == An organic compound consisting of a chain or ring of carbon (C) atoms to which hydrogen (H) and oxygen(O) atoms are attached in a ratio of approximately 2:1; a compound of C, H, and O having the generalized formula (CH2O)n; carbohydrates include sugars, starch, glycogen, and cellulose. Carbohydrates are excellent energy FEEDSTOCK.

So sugar is a carbohydrate (Sugar = Carbohydrate) and Starch is a carbohydrate (Starch = Carbohydrate) as well as others. But the implication was that a Carbohydrate = Sugar which is no always true.

heh, i typed out a rather lengthy retort to Anno concerning his use of the equals sign this morning but figured i was being a picky bastard. i even wrote out the correct mathematical expression for what he was trying to convey. i thought better of it as surely the intelligence in the crowd was capable of seeing what he was getting at................................hmmm
