Hey I'm just curious how did you inoculate? Just shoot in a drop, or did you use an inoculation loop?

shot in 2 drops, some cups got 1 drop

I've been told that's bad and is way too much. I have not done it yet, but I will use an inoculation loop. Hopefully that may help you too. You can make homemade ones, bod has a tek or you can buy em, they're cheap.

Definitely use a drop on a loop and streak OR put a drop to a BRF puck and transfer to agar form that.

ya I made one but forgot to grab it. Was already gloved up in front of the SAB and said beginner's luck let's go! Turns out it doesn't work that way ha

That jmf syringe was growing ick spots faster than the lc syringe grew myc so I'm not sure if it's even worth doing anything with it

ive really only seen rings during the summer. im leaning towards temp these days. when i was a noob i thought it was light because night and day were the easiest variables to observe.

i honestly dont know.

Anyone have tips for germinating old improperly stored prints?

I'd atleast like to try a hail Mary or 2 before I deem them dead

Temperature fluctuations accentuated by winter, the out ring coincides perfectly with the turning on of an oil heater.

I have these plates which all have rings forming at more or less the same time. Checked the temperature over the growth time and after one week around the 12.8 the temperature raised by about 10 degrees and stayed there. Definetely supports the theory of temperature fluctuations.
Either way im happy to hear this because i was thinking its a sign of bacterial contamination :rolleyes:

That used to be said pretty often, yet I've never seen evidence of or seen anyone show a light cycle cause rings. I've also seen cultures in no light have rings.

I like to be proved wrong, but an old post from mushboy, (that you have to filter to his posts to find) isn't gonna convince me. I'll test it when I get a couple cultures going. And try and cause some with light just to see if I can.

All i did was search rings on agar and click the small tc box pimp .
As should all your searches be before trying to correct some shit lol
But fr doe i asked this question about rings on agar a year or 2 ago .

My point is, a lot of things are taken at face value and passed on without evidence. TCS aren't infallible.

I'm sure they are better equipped to prove me wrong than I am at the moment, and I hope they do.

Made a thread but didn't get much answers so I thought I'd ask here

I put some agar plates in the fridge 15 days ago, most of it were in tupperware but this particular one was just in a plastic baggie.
As you can see its all dried out and the agar is kind of folding over itself.
Now I'm paranoid and not sure if just tupperware is enough.
My fridge is 3ยฐC and it's not a dedicated myco fridge

How do you store your agar dish for maximum shelf life ?

You can try rehydrating the print in sterile distilled water. Add a drop of surfactant before sterilization cycle, jet dry or polysorbate 20. After 24 hours, streak spore solution to agar.

I'm my experience, plates aren't really meant for long term storage. I've had success for a few months with them in the fridge but after 3-4 months they start to dry out a little. If you want long term storage try slants or for really long term storage distilled water storage. If you just want to use plates you will need to transfer to new plates a few times a year.

It doesnโ€™t really matter if they dry out though does it? Kind of like dried grains except itโ€™s with agar?

I store my plates for a year plus, then make fresh transfers. It really depends upon the quality of refrigerator, and the culture cleanliness.

Don't use beer fridges with internal freezers or you'll lose a bunch.

:whathesaid: In my experience storing in my mini fridge with a top separate feeezer the wrong spot in the fridge will still freeze plates quick. I stear clear of storing in it now.

Yup. I've revived a dried plate by chipping a piece of agar with myc on it and putting it on a new plate.

I'm using a keg fridge but no freezer. It dries my plates pretty quick. I see your point about freezing if they were in the wrong spot though.
