I'll be honest, I was a bit disappointed in the book.  While there is some good info in there, there's a fair portion of woo as well.  It's not the best compilation of information, but it is a really cool book none the less.

As long as they're sterile. The problem with a lot of plastic tubes is that the tubes are autoclaveable but the lids aren't. PC one tube to test test see what happens. If they're sterile and from a reputable source, you should be good to go.

I picked up some I'd these 50mL self standing tubes from Cell Treat for the same reason. 50mL distilled water and 100mg LME.

Are you using them as your standard LC container for short term storage/usage or are you using the tubes also for long term storage of an LC? If yes - how long you are keeping the LCs and at which temps?

Be careful I got a 50 pack of those from celltreat. The lids were not autoclavable. I tried to do no pour slants.

They are sterile and can be poured.

The plan is to use the LC to inoculate larger containers of SSLC. I don't know how long it will stay good for but I imagine that it would be a very long time. A few weeks ago, I used a jar of SSLC that is almost a year old to inoculate grain. Mycelium had colonized the surface of the SSLC and gone dormant. It kicked off on grain right away like nothing had ever happened. That jar has seen temps anywhere from 50°F-80°F.

I assumed that the lids would melt because Cell Treat's website only mentions that the tubes are autoclaveable and says nothing about the lids. I sterilized 500mL of LME LC and used a pipettor to squeeze 50mL of the LC into those tubes.

For long term storage I would go with just sterile water in the tube and keep them refrigerated.

If you use a sugar broth the culture would excrete the digested sugars and toxify the liquid. (Not good living conditions):lol:

They actually sell autoclavable lids for the tubes. You just have to order them separately.

They work well and come in bags of 50

Can you clarify this statement for me please, or provide a source?

As far as I'm aware, hyhae do not produce toxic (toxic to itself) metabolic waste products other than c02, and an array of secondary metabolites that are utilized for defense and "digestion".

Consider that slants are long term storage and contain nutrients from complex carbs (sugars).

The subject was long term culture storage (up to ten years) in sterile water in tubes.

Every plain corn syrup liquid culture I’ve made has lasted at least two years in the fridge.  I think my oyster is goin on five years old now.

Yes I'm aware of long term storage methods, I was curious about your statement regarding the byproducts produced by the presence of sugar that harms the mycelium.

I can't find the particular write-up So I would say, I'm bad...

Right now I'm not finding It.. The file went south when my hard drive went south..

The write-up was on five and up to up to ten years in vacutainers using sterile water for culture long term storage.

In the write-up I remember how they would use a syringe and needle for putting a vacuum in the tube.

They definitely wrote on any kind of sugar broth would not be inducive for long term storage because of what they excrete.

Excrete: of a living organism or cell) separate and expel as waste (a substance, especially a product of metabolism).

When I think of long term storage In sterile water I think of five seven ten years.

Why Is it you never hear of lc. storage for five or more years?

I'm not a trusted cultivator or a mycologist, So I don't totally understand allot of this..
