Tried to germinate APE spore swabs on Agar without much success.
Out of 5 plates this is the only one showing some activity. Not sure if I am seeing mycelium or a contamination though. The little bright white spot seems to grow like a little mountain instead of spreading out for now.

I have had 3 varieties pin (B+, GT, Tamp), so I have little doubt about what I'm growing there, but all of my AA+ plates look questionable:

What do you guys think? Any of them look worth continuing to transfer?

I should note that other than a few plates where myc outran initial contam, none of my plates have rhizo growth, so the lack here doesn't mean much. I reduced the nutes in my last batch of agar, which will hopefully help with that going forward.

For germinating stubborn APE spores from swabs I like to wet the swab in sterile water and then bury small pieces of it in PF-tek substrate, it works really well. The last link in my sig has the details, scroll down till the end.

My first plate that kind of looks like it's supposed to. Pic isn't the best, but could that go to grain or should I do another transfer? I think that is transfer 3.

If it were me, I would xfer the tip of each spoke to a small jar of grain. Then fruit each one in the simplest way possible, in vitro even. If you have more Petris I would clone each one until you find out which fruits were most potent and ascetically pleasing, pinset, size etc. then make a master jar with that once it grows out. Really so many options depending on what you are shooting for

I would chuck that to seed and look for something to clone.Take a transfer first though just in case.

For example rusty white always produces leucistic fruits from ms. How did it get to that point without ms stabilization and strain isolation and in what order

Some invitro pins back on agar.

I only had one blank plate when I found these just barely pushing on the lid of the plate. I am going to transfer them to their own plates as I take some transfers.

side lighting is upping the macro game considerably.

What are the reasons for cloning invitro agar pins? Is it just because of the clean sample? Or does agar pinning have other implications? Like good genetics?

Well there are only two reasons I take pins off my plates.
-Narrowed genetics
-Because it pinned on the agar dish it could possibly be a better pinner off substrate
However ime doing so I've run into some agar clones that are just absolutely worthless so its a gamble tbh.
