I usually transfer from germ and t1 plates as soon as there’s enough growth to grab some.

After that I’ll start waiting longer to look for sectors.

I’ve always saved early plates. If something turns out really good it’s nice to be able to go back to it.

Plus like somebody said pins are good to clone.

And you can use old plates for experimental stuff like li or different subs.

That'd be really hard to do with glass petris tho. You'd spill more than a few. You can open your PC tho and they will cool much faster.

With glass petri, I use an aluminium wire cage (aluminium wire from the hardware) that comfortably holds a stack of 6. I have used both stainless containers and foil to ‘wrap’ for the PC. . .

Moving them whilst hot is difficult without making a mess, easier when poured real thin. I also find some boil regardless resulting in empty petri. . . It can be done if you are careful, and the racks help with lifting. . .

Josex suggests cooling in the PC is poor practice, which was the case until I met a flowhood, and can now get great consistent success. . . If you design your cages well, you can ensure they sit level, and set level.

I’m led to believe that if I have more water in the PC I may have less trouble with boiling agar due to slower heating/cooling. . . Will report back when done. . .

I got spills every time I tried to move glass petris lol. Anyways, opening up the PC after it drops the pressure is almost the same as putting them out. Also, all that condensation and droplets of water seem to disappear away for me in a few days. Never had any boil over either.

If you are moving them as a stack from a pivot point above the stack, there’s less chance of a spill movement. I’ll post a pic of the cages I’ve made when I get a chance.

If you’ve never had a boil over, how much water do you have in the bottom of your PC? Would far prefer to not pour. . .

I'm putting the rack on 3 espresso cups and water level is below that. I'm aiming to have them filled halfway up.

The only thing I do is reduce the power on stove first for like 10 minutes before shutting it off completely. And I added some weigh to the weight.

Funny thing happened today, my MEA came out with all the agar sunk to the bottom. I microwaved it til very hot, but not til a bare simmer like I usually do. I still poured a sleeve of plates, but they're definitely soupy. Figured I may as well try my hand at sagari. Chopped up a few nice cloned T3's and now have a dozen sagari plates incubating.

I noticed that when I tilt these plates the MEA slides around slowly. I figure if I just don't move them til they're ready to use I won't have any problems — It's basically just thick, goopy LC in a plate, right? What's the worst that could happen? :smile: ...Anybody think I've already failed?

Thanks for the info, will mess around with no pour again. . . No pour glass petri would be awesome to sort consistently.

Time will tell. . .

Consistency of soup is not a major problem, you are half way between LC and agar. . .
The problems will occur if you didn't get hot enough for sterilisation, which I assume you haven't as boiling alone suggests 100°C, not enough on an element. Not sure how you microwave owners get shit done and what the threshold is. . .

Please let us know if they contam. . .

Here are some examples of the cages I've been using for glass petri.

I found some stainless containers that are great to contain during PC, as long as you remove immediately as the pressure drops, otherwise some end up with contams.

I've also added foil covered as an example, this works well also. . .

Hold by the top any they 'swing' when you handle them minimising 'spill' movements if you are gentle.

Make a stack of pre-poured petri, distort cage bottom too fit over stack, slide bends under base of stack. . .

Cages are made from aluminum wire, inert in the PC and easy to work with. Cages made from one piece, starting and finishing as a handle. . .

I'm doing some no pours today. . . Well I'm teaching the wife to. . .

Definitely a bit more work with glass, though less criminal in my mind. . . Polystyrene is nasty! I've been through all mine at least 7 times. . . 3 more to go. . .

Only broken 2, and not by dropping, by stupidity. . .

Would really like a way of dishwashing, as this would make the process much easier. . . They get flipped in mine though and fill up with water and aren't clean. . . Any ideas?

I like those cages and ss containers and glass. I'm using 75mm pp petris. I just use my old muda bottles and load them upside down. I foil the lids to make sure the stay sterile till I pour. When it's time to pour I just pull the foil, flip the muda bottles upside down on my rack, unscrew the lid and lift the bottle up and stack them off in a corner then pour.

You can kinda see the workflow in this picture. It took maybe 45 mins including syrup to pour these 40 petris. Ang just under 600ml of agar

I also do glass petri no-pours, but wait til the PC is completely cool before opening. Some times I wait a couple days. So why are you kids pulling them out hot and risking a hot mess? Advantage unclear.

Also, regarding washing, I dump the agar with the twist of a spoon, the dish is practically clean. A quick wipe with a wet paper towel is all it gets.

Nice writeup.

I'm using glass to avoid using single-use plastic. . . Cling wrap I will be avoiding. . .

I find it takes a bit of scrubbing getting the marker labels off, and I'm a bit too lazy and leave plates dirty for too long and they get stinky and mouldy. I also use 1.5% or softer agar which is a bit messier, though not that much. . .

Hi all, I'm brand new to messing with agar. It's time for my first transfer from some clone plates, thought I'd ask if what I'm doing makes any sort of sense :smile: This thread (among many others) has been a fantastic resource- thanks for sharing the knowledge!

Pics of the plates:




Plate 1 I'm thinking the area from 10-2 looks dodgy, so I'll give that a wide berth. But will take transfers from the main colony at about 4, 6, and 8, and probably one from the satellite colony at about 3.

Plate 2 will avoid 11-1, but everything else looks ok to me, will take a few from around the periphery.

Plate 3 looks weird to me from 3-5, and from 8-9. Will prob take a few spots from 10-2, and maybe one from 6-7.

Anything look off with my plan here? Any feedback appreciated.

(And yes, I've learned my lesson about labeling in the center! That'll go closer to the edge in future.)

Submerged myc or contam? I can see little rhizos in the contaminated looking area.



Is this t2 toc showing me my wild temp swings in my cabinet during lights on or mold? I'm working on a new cabinet but it's been slow going should correct my temp issue.

First plate is bacterial AF... if you were to touch the snowflake (or around the edges), its slimy as hell... looks like thick mucus yeah? second plate MIGHT be very weak, tomentose growth, or mold... either way, it needs work before I'd run it personally
