If that’s two weeks old definitely don’t goto grain. Looks iffy to me, Mold perhaps.

Well fuck.  What a waste of time.  I took some better pics.  Ive been fighting a constant nagging mold issue with my monos.  For like months.  So I started from the beginning.  Guess It looks like my source is the problem.

nah mane dont go to grain with a plate that has obvious contam on it. keep trying to clean it up with more transfers. though it doesnt look so good to begin with , so also start fresh just in case

For what it's worth, the agar I'm using may not be the best.  I bought some agar from seaweed laboratories and I found a single thread on this forum that said it was food grade agar and more geared for bacterial growth.  The poster suggested adding potato flakes, which I did, and from then on was able to successfully grow cultures on the agar plates.

So, maybe that is why it takes so long to grow, but this is 5 transfers from what I was suspecting a contaminated LC. I thought I could clean it up, but I guess I don't yet have the eye for it. I guess maybe I'll just scrap it and start from MS  that I have laying around.

They have more than one type of agar. I’ve used their MEA and have had success

Confirmed.  Since it's looking bad I pulled the lids off of two of them.  They smell like mold. Not myc.    Back to the drawing board!

are you using their premade MEA mix? im pretty sure that is way higher nutes than people usually use here, like WAY higher, since they use 56g per liter. adding additional nutrients (potato flakes) definitely isnt gonna help. id recommend making your own, its easy af. check my sig. most people here use 15g LME, 20g agar, 1L h2o around here. alternatively, you could probably just use half the amount suggested by seaweed solutions (add more agar agar powder if needed)

proper sterilization will kill any bacteria present, so so the issue is almost certainly your clean technique (based on the satellite colonies of bacteria), and/or dirty spores. are you starting from syringes or prints? SAB or FH? doing any control plates?

Ahhh I was hypnotized by those plates immediately, I haven’t been using food coloring but this inspired me to maybe make some up! I think I saw that brand of food coloring at Vons?

20 Days after my very first agar plates, having no clue what I’m looking at, too developing these beauties! These are third transfers! The Fatass I know is a huge piece transferred to the plate but I’m a noob and am trying different things, also the plate it was transferred from was being tossed out anyways:smirk: Thanks for all the useful information from the community! My question is when to transfer these to jars now? And how many one quart jars can a plate be split into?

Yeah... the first stack gets poured around 120*F... by the time I'm finishing the third bottle its juuuusssssstttttt starting to set... and I've also noticed that if I just let them sit for a good day or two (which almost never happens...) then they seem to even out completely.

B+ ms syringe from a reputable source.  It's a year or two old but it's been stored in the zip lock and box it came in, in a drawer in my refrigerator that has nothing else in it.

Guys! noob alert

So I stand before for 3rd transfer from the MS I started with and I would really love some professionel advise now, from what to transfer and what not to and more specifically from where int he given plates.

I only have 20 new petri dishes left and I'm having a hard time getting more now because of all the corona stuff going on, so have should I use what I have left?

So here are all 20 plates I have... I spot a couple of plates with some yellow spots (contam I guesss?!?)

What’s your plan here?

Honestly none of them look ready for grain yet. You might get away with 7, 17 or 19 but it’s hard to tell from the pics. Do more transfers but don’t use up all your dishes if you can’t get more. Take about 3 transfers to 3 plates from strong clean growth to try and clear them up and work on your sterile tek. Any more would be a waste imo

My understanding is that there is very little point to isolating MS because you've got no idea if the genetics you're isolating are any good.

Put some of those plates to grain, fruit, look for a strong cluster, take a tissue sample *then* start playing with agar.

The only point of agar with MS is to cut out contams which you've certainly done at this point.

Take a tissue sample like this...RR picks a clone  be sure to label the shroom and plate...eat the shroom!

I used to be like this and have loads of plates.  No I work like this.  For every culture I have I am looking to do 2 things.  I am trying to  A- get a clean culture to use for grains, LC's LI's ect and B - 1 to store.

With this in mind from every clone plate(I could take 3 clones at 1 time), spore plates (usually 3)  I take 2 transfers per culture.  Once I get a clean culture on 1 or both new plate, 1 goes to grains or LC and 1 goes in storage.  With spore plates im trying to grow so I can get clones. With clone plates If that culture is really good I G2G as long as I can till finally it gets dirty or i get bored and then go back to the storage plate and make new plates- One goes to grains/LC and the other storage, repeat.  If only one is clean I keep transferring from the non 100% plate till I am happy with it, then store.

In the beginning, like you,  I used every vessel in my house to grow cultures and it got overwhelming.  I using maybe 30 plates at the mo but have 12-15 cultures im experimenting with.
