Guess what Q tips suck ass for tall jars.

Comments:

- Shaky, fiddly in the beginning.
- A few times I hold my hands over shit, getting more conscious of that.
- Obviously the short length of the swabs betrayed me this round, but hey at least Im not using them for anything else than cleaning up the spore print.
- Torched my finger
- Its not a good idea to cool this inoculation loop in agar, because it will
actually bring agar back with it to the spore print
- having to clean up agar spill on your spore print obviously isnt a great idea
- double dipping the last jar

You are improving and it sounds like you are re-watching your videos for where you messed up :thumbup:  Second video - don't put your bare wrist over the spore print and scrap the whole Q-tip thing completely.  In my opinion they don't have any place with what you are trying to do. 

It doesn't take very many spores to inoculate a plate.  If you are concerned about contaminating your print with agar from your loop, one option is to tear the foil and the print into smaller sections (while inside the SAB).  This way you can store away the larger print and just work off of the smaller piece you tore off.

P.s. Sorry for calling you a troll, but when I watched the video with the Q-tips and the jar of water I genuinely thought you were screwing with us.  Still looking forward to seeing you succeed.

Thank you a lot, that means something to me. Made some transfers today, having a ball with buying these glass jars with metal lids at the local convenience store, tomato paste 0.70$, taco sauce even cheaper 0.55.

There is now a mason jar without any visible contams, and with seemingly strong mycelium.

If you got white growth, and it bruises blue, you can be sure right?

The reason I am asking this, is because when I worked with Agar
transfer today in the SAB, I noticed the agar plate mycelium
that I was messing with, started to bruise several places.

It depends on the stage of growth. If the mycelium is past its vegetative/colonization stage, it can bruise blue, inevitable of what it has grown on. RR was speaking specifically of colonizing.

Yes, myc can bruise blue when transferring.  I often swipe the whole surface of a plate just to watch it turn after my transfers are finished.  It can take anywhere from a few seconds to 15 minutes, in my experience

Smoked 1/4 of a joint
Made 8 transfers using a steel wire 
Since the pore popper is missin. Made sure to give
it a good and thorough burning with my bic lighter.
Wearing face mask and gloves, but no ISO sanitizing,
this is because the burning of all the hair on my forearms
earlier today.

Im getting a camera for better pictures so we can have
more informed discussions on the growth I'm seeing in
my agar jars - ETA on this is next sunday.

Im experimenting with being generous with the amount of
colonized agar I transfer to new agar jars or to PF tek
jars. Everything I have read makes me believe that its
advantageous to the impatient and to the beginner
to use as much spawn as possible. The reasons
for this is contaminations, which the mycelium
receives a really big head start on.

Im also feeling more comfortable with my 6 L PC
because Im seeing that control jars or PCed content
stays not visibly contaminated as long as I dont
open the lids on any of my stuff.

Many agar jars now exhibit what I believe to be clean, white, puffy and uncontested mycelium. There is even a PF tek jar style jar, that is about 10% into colonization. Things are looking up and I'm feeling better about the project. 

Today's task is to PC 3 jars of coir only, which I plan to make agar piece transfers into when new agar jars are colonized.

I left the coir in a bucket with water for 1H, and now Im PCing it for 60 minutes. The lids will be tightened when the PC is done, and the jars moved out of the PC to cool, so I can make some more agar jars.

DO NOT PUT AGAR INTO JARS OF ONLY COIR


EDIT -You must transfer agar into a nutritious media, which coir is not.  Grains are my choice, however you can do a multitude of things, but not coir.  Wild bird seed is cheapest for me and you can find many teks on many different things to use as a nutritious media with which to transfer your agar to.

After these have colonized, you can transfer these to coir.

No.  It might branch out looking for nutrients, but there is nothing more for it to eat.  On agar, it eats whatever nutrient you have in the agar.  for me its BRF.  There is no nutrients in coir (aside from perhaps microscopic levels that are negligible and not enough for myc to live on)
