So what’s your take on the blackhead removing “inoculation loop”, Verum?

It'll work fine for some things but suck balls for others. Firstly, I like my loop to have a little give in it so that it bends a bit when streaking. This makes it possible to streak plates faster without piercing into the agar. Sencondly, the loop portion looks a little too wide to hold a drip. I may be wrong but it looks that way.

I get your point but, swabs can be sterilized and can also be used more than once if done correctly

How can one sterilize a swab? I suppose to be clear, how can one sterilize a swab that has spores on it?

You can not, as far as i know. Haven't researched the temp that kills spores so i can't be sure. Non spored swabs, as I'm guessing you know, can be wrapped in foil or thrown in a mason jar and PCed or cooked in the oven to sterilize.

Ok, that was my point. I appreciate the need for clarification. I was a bit broad when I said swabs were one use only. I meant that in this context, when using a swab with spores, there is no way to sterilize it (should it get contaminated), so best open it in the SAB and don't dip into anything aside from the agar.

And now that I think about it, if the sterile technique is good, I suppose one could put the swab back into the foil to be used later. Not entirely sure on that though. I guess I don't see why not, aside from the possibility of contamination.

I have had zero luck with swabs. Not even contams, just nothing. I would think agar residue and moisture in a swab would cause something to germinate in storage but I've never seen it with swabs I've tried.

ok guys germination time!
(edited)(plate 1) (plate 2)(plate 3, streaking)
(unedited plate 1 pic)

so if i'm not mistaking this is spore germination? to my eye it seems that the spores went brown, is it because they are bacterial? and also the further the line goes the cleaner the myc development seems, can you confirm or is that bs?

what to do now? wait? transfer? thanks for staying with me
much love  #nohomo

Strange, Ive had several mediums with this kind of growth and its been called contamination. If the streaked dots really are myc in these dishes, I cannot tell the difference from this:

Im able to make a transfer tonight, as far as I can see on my plate here, there is only one small spot of bacteria, and one small spot of trich.

Any suggestions?

It's a lot easier to start clean than it is to fight contams the whole way. I personally would start over and if you get trich transfer before the trich turns color and makes spores.

If you have some skill you can use a variation of the sandwich Tek, or hotpour, or hotpour+ peroxide. Or transfer to cardboard until you can get clean tissue. I'm sure there will be opinions on these methods bc some are old but they work well for me.

When you open that you're going to have trich spores all over. I'd focus on getting one clean plate colonized the first time instead of transferring dirty ones. There's plenty to be learned either path you choose.

Okay, so the problem with using this plate is that the trich turned green.
I must have transfered before it sporulated.

opening this in the sab today != good idea

I would just keep transferring from those plates you have that look like that.  That looks to me like fine myc.  It looks like you dropped a whole bunch of drops onto it and now they are germinating.  I could be wrong though because I've never dropped from syringe to agar, only scraped from prints.  Plus it's hard to see on mobile

Edit_i just saw the green in there.

I have been unsure whether or not that was mycelium, but now Im more sure than ever on how they will look when they germinate. This is a big improvement for me

I think we will have clean spawn in this thread soon (1w)

While waiting for new dishes, i might as well just keep trying to get clean growth on jars :sun: I just clean them and PC them, thats what they are for

It looks like lots of little colonies for sure. I used to just drop a drop in the agar and streak but lately I've been cutting a little slit in the agar where the drop sinks into. I don't like when the drop is able to flow across the agar to the edges bc I've noticed my contams usually don't start in the middle of the agar, I don't like the drop carrying things across my plate.

To me trich Mycelium has a dusty look to it before it starts to produce spores. Idk that helps that's what I look for early on. Fast and dusty.

Don't transfer that myc into grain or brf jars.  Make more agar jars if you don't have dishes to pour in.

Edit * don't transfer that *stuff* into grain or brf jars, I meant.  I'm still not entirely sure that's mush myc.  But I'd go with it of I were you.  You'll know once you transfer if it starts to behave like myc

The streak plates are def mush myc
