I only own one scalpel. I see no need for more than one with replaceable blades. An inoculation loop can be made very easily by twisting thin wire. Practice using the search function a little. You'd be amazed at how many people have asked the same questions over and over again. It's all there to be had.

My handle isn't nice. All that matter is that it's flame sterilize-able and long enough to keep your dirty hands away form being over the dish.

I get cases of 250 for like 30$



The benefit to these long inoculation loops is that they are long enough to reach into an lc without having to flame sterilize a long piece of wire.

By all means, pour more agar. But don't toss those unless they contaminate. 2 days is hardly enough for the freshest of spores to germinate.

Update

Agars

Agars look bacterial, many of them. Some yellow satelites.
A few dishes have spots that look they may be satelite colonies of mycelium.
This means either complete redo or a lot of transfers. Its only been 3 days so I dont see much and im entirely speculating based on what I see.

More loving teaching from this hobby if I have to redo.
And I have already ruined like 10+ dishes. Only got 8 left.
Got to order better petri dishes.

General Jar production

Made 3 jars of WBS with dry verm lyers that were rinsed 10 times, soaked for at least 24 hours, boiled for 20 minutes, then rinsed again, and PC cooked for 120 minutes. The foil is left on and the jars stay inside the PC cooker for now.

These are jars that I plan to use the agar cultures on later.

I use an exacto blade for my scalpel. I have a 50 pack of blades. I’ve used maybe 3 blades in 10 years. Works fantastic!

If you're inoculating with agar you don't use a dry verm layer. That's for syringes and pf tek.

Some of the dishes had too little agar in them which may explain the «seas» and «continents» on the dishes. One dish for instance, has a part that is liquid.
But only that dish.

Let me guess, even if you have mycelium on your dish, if it has bacteria or something else, you cant use it? Even for transfer?

Im not gonna throw away the agars just yet, but please start to kill my hopes in the harshest way possible.

Q: Under what circumstances does agar change color other than organisms?

Gonna inject the remaining agar of the agar jar with 1 ml of spores. If it works I will have a huge petri dish

Had major problems in the SAB today. Spilled agar everywhere, the fucking plastic dishes are cheap and pissing me off. Thanks for the great non-pp5 petri dishes, mother.

My sab doesnt have a flat floor, this was what was causing problems today.
Net result was only 2 agar dishes made, now cooling in the refrigerator.


Gonna try again to make more agar tomorrow, with spirulina. This time I will inject a jar with it, after reading this^

Huh, I have an X-acto, but never even thought to use it. I just bought a scalpel instead. I'll give it a try.

Anything sharp that can reach incandescence can be used as a scalpel. They just have certain ergonomic qualities that are preferable to biological applications. Surgical scalpels are usually flat. I, personally, like the cylindrical design of a hobby knife because I've had a lot of time handling them in delicate situations. Flat ones feel clunky to me.

because of huge problems in the SAB today with spills and generally being incompetent the jar idea is postponed until tomorrow, and i will downsize the spore use to 0,2ml, :crazy2:

I ended up with only making two dishes which are cooling now, both with condensation, but with pristinely clean top parts. (no spill)

Avoiding condensation

Q: Why dont people just let the dishes cool with the tops off in the SAB, leaving the room for a while, and then wipe down the jellified agar with isapropanol after it has cooled for sanitization? Will contaminations on the top surface of the agar actually survive being wiped with isopropanol? And since it evaporates fastly, it will probably not cause lasting condensation.

the small enclosure could create enough air isopropanol inside the air space of the dish too probably sanitize the inside of the dish too

is it worth attempting?
