What is the Psilocybin/Psilocin content of Subaeruginosa (per gram)

Yeah so I'm planning my next trip and I want to use the calculator on this site to figure what level I'll be aiming for but I need to know the Psilocybin/Psilocin content per gram of dried Sub. any kind of support will be much appreciated.

I wouldn't mind knowing the answer to this as well since dosing pure psilocybin is something I'd like to do...

Psilocybin has been isolated from this species in 0.45% yield.[8].

0.45% psilocybin... So, 10g of fresh subs (1g dry equivalent) would contain... 0.045 grams of pure psilocybin, right?

That 0.45% yield is for dry weight Painkiller, so 10g of fresh subs will give around 0.0045g psilocybin.

No worries buddy, I had to read the referenced journal article to find out how it was done - twas an interesting read. I wonder if the scientists who do these sorts of experiments are psychonauts themselves :bigblunt:

The "milligrams per dried gram" is not necessarily accurate, since alkaloid content varies from
mushroom to mushroom. As such, the dosage calculator is basically rough guess work - the best
way to gauge, in my opinion, is to try a small or moderate dose to see how far it takes you.

Even then one might be in for a surprise, i.e. if 1 dried gram only provides a "level 1" or "level 2"
experience, adding half a gram might just propel you intro a full "level 4". The higher the dose, the
less predictability, is my experience. That's mainly with liberty caps though, so I'll have to count in
the presence of Baeocystin too, which is not included in the calculator.

So true Anglerfish. There's also tolerance level to take into account too.

I had not dosed for a long time, not in the last 9 months or so, and had 2 medium-sized fresh caps yesterday (subs) which almost blew me away. Was starting to see faint trails in my vision and the body load was quite uncomfortable... all on 2 measly caps which I cooked and made into an omelette with 2 eggs and some leafy greens from the veggie patch. What a trip it was, 3 or 4 caps would have put me into a delirium!

Beware that 0,45% psilocybin is most likely not correct. The study (Picker, J.; Rickards, R.W. (1970). "Occurrence of psychomimetic agent psilocybin in an Australian agaric, Psilocybe subaeruginosa") was made in 1969 and could have weaknesses, I haven't checked thoroughly.

I have made a note several years ago about another study where Psilocybe subaeruginosa was found to contain 1.41% psilocybin and traces (0.03%) of psilocin. The problem is that I can't find the exact reference when I look for it now due to a broken link. I referenced Anastos to this and found a study, but I can't find the number in it: Anastos, N., Lewis, S. W., Barnett, N. W., & Sims, D. N. (2006). The Determination of Psilocin and Psilocybin in Hallucinogenic Mushrooms by HPLC Utilizing a Dual Reagent Acidic Potassium Permanganate and Tris(2,2’-bipyridyl)ruthenium(II) Chemiluminescence Detection System. Journal of Forensic Sciences, 51(1), 45–51. but it might be in another of his articles or maybe someone asked him and wrote it on a forum or something... Anyway, this made me investigate this a bit deeper.

In 2015 Wikipedia had another note about the potency which was later removed:

Now, I looked through some tables in
Occurrence and use of hallucinogenic mushrooms containing psilocybin alkaloids which could have uncertain information, mistyped numbers etc. It's not a very good scientific report... Anyway, on page 50 we find the different references and numbers for :

The numbers are mg/kg dry weight which translates to:
0,45% - (Picker and Rickards, same as the first reference in the top of this post)
0,2% - (Perkal, M., Blackman, G.L., Ottrey, A.L. and Turner, L.K. (1980) Determination of hallucinogenic components of Psilocybe mushrooms using high-performance liquid chromatography. J. Chromatogr., 196:180–184.) Observe that this is not the same article as the removed wikipedia reference)
0,11% - (Anastos, same as I referenced above)

Now, there is a methodology problem when reading and comparing different studies of quantitative extractions. Perkal only extracted during 2 minutes which could explain the result above.

And we also know that psilocin in reports often are lost due to oxidation. We can for sure know that psilocin is in the mushrooms since the blue color is a breakdown product of psilocin.

So I definitely think it's much stronger. Just have a look at the Shoomery species page for Psilocybe subaeruginosa:

There´s no way a 0.45% mushroom give level 5 experiences at 3.6g. But with >1% it makes more sense. :wink:

So.. what do you say? Is there any more reliable sources?

And I guess you experienced hunters from down under can testify that subs are more potent than Psilocybe cubensis for example? How is it compared with Panauolus cyanescens?

i find potency to be highly variable to the extent i would likely advise away from the dosage calc. for example the species stuntzii... i have heard too many contradictions on this species. i have heard from many that they are pretty potent but have also heard from many more that they are super super weak maybe the weakest of all.

the dosage recommendations for stuntzii are numbers so high i wouldnt even attempt them, i usually hover around a level 2 on my doses, maybe approaching a level 3 but i would NEVER want to go past that.

stuntzii is listed at 4.6g for a level 2 and 8.1 grams for a level 3 lol, those are scary numbers of grams to me. the only species i have experience with are cubes and ps cyanscens, and ive had a batch of cubes that were on the same level if not more potent then the cyanescens. granted the cubes were cultivated but it was a MS inoc, wasnt a cloned strain. however i have also had cubes that were not potent at all and all the cyanescens are pretty potent, although some more potent then others.

i have attempted micro dosing with horrendous results lol. needless to say im not doing that anymore lol.

so i guess, i would hypothesize that subs are generally more potent then cubes. but there seems to be exceptions from batch to batch and patch to patch, could even be shroom to shroom form the same mycelium network although i wouldnt be sure about that.

i find this topic incredibly interesting as it doesnt appear to be settle science just yet. seems there are always discoveries to be made.

another factor that may be contributing is mistaken ID's
for example lets look at panaeolus cinctulus.

this is another species which ive heard different accounts on potency. and have also read that cinctulus doesnt occur in lawns. which is where lots of people 'find them' including myself. i believe alan has said the 'cincts' in lawns are not panaeolus cinctulus but some other panaeolus. (correct me if im wrong)
so this leads me to believe that many of the reported cinctulus are not cinctulus and thus 'dosage charts' will not be accurate i they are averaging active compounds present over many collections. or where those collection were from.

also if i remember correctly, beta knight back in the 15-16 pnw season, found a patch of subs here. i could be wrong as i dont know if any follow up was made but i believe he got them sequenced for a proper ID. lots of the wood lovers are similar to the point they may get mixed up and inaccurate data gets unknowingly recorded.

i mean if you look at the psilocybe alenii page on this site, it states somewhere that paul stamets says its microscopicly similar to cyanescens and thinks they are just cyanescens. granted that could be an older paper from him or something along those lines, or perhaps i misunderstand the phrase microscopically similar as microscopically identical.

No one can give you accurate information on this,  due to this being listed as an illegal drug, with no approved medical use under the United Nations charter, no one has been able to do studies that will answer your question. However, the FDA in the U.S. has given Psilocybin breakthrough status, so, this may change, but it may not, especially since they can synthesize the compounds now.

Keep in mind if they extracted a certain amount that doesn’t mean there is that amount in the mushroom.  No extraction has a 100% efficient yield.  So you’re likely getting more psilo by eating the mushroom than what these figures may suggues

It's not that it's complicated, it's the legality of doing such testing. There are field reagent tests that check for the presence of psilocin and Psilocybin, it's part of the NARK II test kit, just to name one.
